Description
【発明の詳細な説明】
リポタンパク質及びアポリポタンパク質の測定方法本発明は、リポタンパク質又
は特にアポリポタンパク質の定量的及び/又は定性的な免疫化学的測定法用試料
の新規な処理方法に関するものである。本発明の免疫化学的方法は、それらの対
応抗原と反応する抗アポリポタンパク質抗体又は抗すポタンパク質抗体を必要と
するものである。
リポタンパク質は、トリグリセリド、リン脂質、コレステロール及びそのエステ
ル、Tim脂肪酸及びタンパク質が互いに非共有的に結合した集塊(aggre
gate>から成るものである。リポタンパク質中に存在するタンパク質はしば
しば゛アポリポタンパク質”と呼ばれ、それらの個々の名前は当該タンパク質の
タイプ名の前に接頭辞“アポ”をつけたものである。従って、例えばアポA1.
アポAI[、アポB、アポC1,アポC■、アポC■、アポD、アポ(a)、及
びアポE等がある。これらのアポリポタンパク質のうちの幾つかは異型(iso
form)を有している。
また、いわゆる冬型性も生起し得る。
集塊全体の形態で存在しているリポタンパク質はよく大きさと密度に従って分類
されることがある。即ち、キロミクロン。
超低密度リポタンパク質(VLDL) 、低密度リポタンパク質(LDL)、リ
ポタンパク質(a) (L p(a)) 、及び高密度リポタンパク質(HDL
)がある。これらは更に様々なサブグループに分類され得る。これまでに、アポ
リポタンパク質の主なものは一つ以上の大きさの分画中に存在していることが示
されてきた。
唯一の例外はアポ(a)であり、アポ(a)はこれまでにアポ8と共にLp(a
l中でのみ検出されて来た。
アポリポタンパク質の多くがレセプター結合及び/又は酵素制御機能を有してい
ることが明らかにされてきた。アポリポタンパク質は脂質代謝に於いて極めて重
要な役割を果しているものと考えられている。
血清及び血漿中に於けるアポリポタンパク質及びリポタンパク質の様々なレベル
は先天性及び後天性疾患と関連のあることが判っている。更に、該レベルをある
秒の収縮疾患の危険可能性と関連づけることができる。特に、高レベルのアポ(
a)及びしp (a)は心臓血管に関する危険性を示すものと考えられる。
血液の様々な両分から、特に超遠心、クロマトグラフィ、ポリアニオンによる沈
澱、及び電気泳動という手段によってリポタンパク質を互いに分離することがな
されて来た。こうして分離されたりボタンバク質は定量され、特にその中に存在
するアポリポタンパク質によって特徴付けられてきた。この特徴付けには様々な
免疫化学的方法及びその他の方法が援用されている。
アポリポタンパク質の検出測定方法としては、放射免疫拡散法;電気免疫、散開
免疫及び酵素免疫法:等電点電気泳動、比濁計(nephelometry、
turbdimetry)、ポリアクリルアミド電気泳動及びデンジ1−メータ
ー等を挙げることができる。例えば、蛍光及び化学ルミネセンスのような他の検
出方法も同様に使用することができる。
リポタンパク質及びアポリポタンパク質、それらの構造及び定量、並びにそれら
の血清及び血漿中のレベルの臨床学的意義については多数の参考書及び文献中で

リボ及びアポリポタンパク質の免疫化学的測定を標準化することは困難であるこ
とが判明した。試験の場所(研究所)及び時が異なれば得られる結果も互いに違
ったものになるという事態がしばしば発生した。試験試料の処理が重要な因子で
あることが知見された。異なった結果は、試験試料の保存に起因して生起する傾
向のある該試料の非再現的変化という現象に特に由来していることが判明した。
この問題を避ける為に、脱脂することによってリポタンパク質中に存在する抗原
決定基が更に露出されるであろうという考えに基づいて、脂質溶解溶媒、様々な
型の洗浄剤及び酵素による加水分解(例えば、リパーゼ)により試料の脱脂を行
なうとする目的の各種の実験がなされて来た。しかしながら、それら脱脂方法の
うちのどれ一つとして実際の成功を収めたものはなかった。
優先期間中、スウェーデン特許庁は国際調査報告に於いて以上が本発明の先行技
術の正確な記載であることを強調している。
その中では以上の文献が引用されている。

処理方法が開示されている。試料をイムノアッセイプロトロール(例えば、比濁
if )に援用する為゛に、ある種の脂質分解(加水分解)酵素及び/又はテン
サイド(tenSideS)を該予処理ステップで用いた。
2 、 Albers J、J 他:関節硬化症に関する第5回国際シンポジウ
ム(ベルリン) 、 1979年、 p8N−15(発行1980年)に於いて
、適当な試料の加水分解処理によって、アポリポタンパク質(特にアポB)の抗
原決定基を露呈させることの重要性が確認された。該発行物によるとこの露呈に
関する問題は未だに回避されていない。

ることなくアポリポタンパク質の特徴付(キャラクターライゼーション)につい
て記載している。
本発明の第一の目的はアポリポタンパク質の免疫化学的測定に用いる試料の安定
化方法を提供することである。第二に、タンパク質測定の再現性を改善させるこ
とである。第三に、ある種のアポリボ及びリポタンパク質の異常レベルに関係の
ある疾病の診断の信頼性を改良することである。第四に、タンパク質、特にアポ
(a)の抗原決定基を露出させる方法を提供することである。
本発明は前記タンパク質の異型(isoforms)及び冬型性の免痩化学的測
定及び研究の為の改良さ、れた方法を提供するものである。
上記目的は、所望の抗アポリポタンパク質抗体又は抗すポタンパク質抗体と試料
を反応させる前に、該試料のpHを予処理ステップ中で約pH9,0より高く又
は約3.0より低い非変性値、好ましくはpl(3,Q−9,0の範囲外に維持
する本発明方法によって達成され得る。試料はこのI)Hで、免疫反応に対して
最適なようにリポタンパク質の抗原決定基を霧出させるに充分な温度・時間で維
持される。本発明の予処理の最終ステップとして、自体公知の方法で免疫化学的
測定が実施され得るように該免疫反応に必要とされる値にpHを調節する。“非
変性値”とは、試験に際して決定的に重要であるアポリボ又はりボタンバク質の
抗原決定基が例えば加水分解又は不可逆的変化によって破壊されないようなpH
値を意味する。
試料としては、全面又はそれの各種分画、例えば、血漿又1ユ血清分画を用い得
る。あるいは、血液中で自然に生起するりボタンバク質分画の一つ(例えば、キ
ロミクロン、VLDL、LDL、Lp(a)又はHDL)を自体公知の方法で分
離して試料として用い得ることができる。
予処理ステップの間、pHを好ましくは0〜3の範囲、例えば0〜2.5、又は
90〜14.0の範囲に調節する。接客の方がより好ましく、その中でも更に約
10以上、例えば約11より大きく13より小さい範囲が好適である。pHの調
節は非緩衝酸、例えば塩酸、又は非緩衝アルカリ溶液例えば水酸化ナトリウムに
よって実施される。勿論、希釈溶液を用いて該調節を行なえば適切な添加コント
ロールが容易になされ得好ましいことである。上記範囲内で強力な緩衝能を有す
る緩衝系の助けをかりてI)Hを調節することもまた容易である。適当な緩衝系
としては、前記範囲に−pKaを有する酸−塩基対0例えばHPO4/H2PO
4−及びCl13COOH/C13COO−(pHO〜3用)、又はHPO/P
O(pH9−14用) カ9ケラレル。9.0ヨリi!いpH値の場合、水酸イ
オン濃度及び/又は添加緩衝成分は10−5Mより大、例えば10−3M、適当
には2Mより小、例えば0.5Mより小である。pHが3より低い場合、H30
+濃度及び/又は添加緩衝成分は101Aより大、例え・ば10−2“5Mより
大で適当には2Mより小である。
必要であれば、様々な脂質分解酵素(前記予処理ステップを実施するDIに於い
て活性のあるもの)を加えることもできる。
予処理ステップに於ける緩衝能は次の免疫化学測定に必要とされるpH値への調
節を容易にし得るものにすべきである。予処理中に、酸性物質が放出される。こ
のことは試料が部分的加水分解を受けていることを示している。M’ffl系は
この放出物質を中和することができる程高い能力を有すべきである。
該予処理ステップの効率を次の免疫化学的測定との関係で研究することができる
。即ち、例えば最適温度、時間及びpH条件は、これらの各々の変数が実際の試
験操作にどのような影響を及ぼすかを別個の実験で決めることによって、夫々求
めることができる。実施例に於いては、いわゆるサンドウィッチ操作によってこ
れを行なった。温度及び濃度が余りに高いと、又は予処理時間が余りに長いとア
ポリポタンパク質を変性させ得る可能性があることが判明した。このことはサン
ドウィッチ法に於ける取り込み(uptake) 量の減少を意味する。
本発明の予処理は好ましくは0−50℃の範囲で実施され、1゜−40℃が最適
である。該予処理はステップは幾つかの一連のステップのうちの一つであっても
良い。
酸及び塩基の温度及び濃度は前記選択pHに於いて変性を生起させないような値
であるべきである。
予処理時間は実際的見地から決められる。温度及びpHは予処理ステップが5分
〜20時間、好ましくは5分〜5時間以内に実施されるように選択される。
自体公知の方法で免疫化学的測定を実施するとは、十分な感度及び特異性を有す
る免疫化学的方法であれば所望の如何なるものも使用することができるというこ
とを意味する。リポタンパク質及びアポリポタンパク質に対する数多くのこのよ
うな方法が知られている。例えば、前述した引用文献を参照されたし。
本発明の予処理はこれらの全ての方法において有用であり得る。
特に、分析検出可能基、例えば、醇累学的に活性なもの(酵素、補酵素、補因子
、基質等)、放射活性なもの、化学発光性のもの、蛍光性のもの、粒子(ウィル
ス、ラテックス、金)を有する免疫化学的反応体の少なくとも一つを使用する方
法に於いて有用である。
これらの測定に於いては、抗原(ハブテン)及びそれの対応抗体間のアフィニテ
ィ以外でそれと等価の生物特異的アフィニティを利用することが可能な場合があ
る。このようなアフィティを有する物質の例として、プロティンA−ICIG:
炭水化物−レクチン:C1q−イムノコンプレックス:RFレセプターイムノコ
ンプレックス:ビオチン−アビジン等を挙げることができる。
現在得られている結果によると、本発明はりボタンバク質(a)又はアポ(a)
に対する免疫化学的測定に有利に適用することができる。
特に、標識免疫化学反応体を、例えば血清、血漿及び全血中で定量化するように
、特異的アポリポタンパク質をこれらのタンパク質同志を予め分離することなく
定量化することに利用する免疫化学的方法に於いて1要な利点を得ることができ
る。
以下、本発明を数多くの非限定的実施例で詳説する。そして、リポタンパク質及
びアポリポタンパク質の免疫化学的測定に用いる試料の脱脂に関する以前の方法
に較べて、本発明のそれが優れていることが明確に示されるであろう。
牛血清1%、 EDT八0へ 01M及び静菌剤0.15%含有)抗アポリポタ
ンパク質抗血清
ヤギ抗アポ(a)、羊抗アポ(a)、ラビット抗アポAl及びラビット抗アポB
抗血清はしp(a)、アポAl及びL[’)Lを用いて公知方法により免疫感作
してl[L、た。
アポ(a)及びアポBに対するモノクローナル抗体夫々精製Lp(a)及びLD
Lで免疫したマウスのN1ji!細胞とマウスミエローマ細胞をハイブリッドさ
せてマウス抗アポ(a)及びマウス抗アポBモノクローナル抗体を調製した。ハ
イブリダイゼーション及びに続くl\イブリドーマの培養及びクローニングは以
下の文献に従って実施した。免疫学に於ける研究論文(Rese、arch H

mical Press、New Yorkマウス抗アポ(a)モノクローナル
抗体を1251でクロラミンT法に従ってラベル化し ■−標識モツクローナル
抗アポ(a)抗体を得た(Hunter & Greenwood、Natur
e 194/1962/p、495参照)。
アポATに対するマウスモノクローナル抗体はメディカル・ダイアグノーシス・
インコ(CaIIlvridge、 UK)から購入した。それらをクロラミン
T法で標識した。
デカンティング・サスペンション(deCantir+g suspensio
n)(Phramacia AB、 l1ppsala、 Sweden )は
アガロース粒子に共有結合した馬抗羊ICIG又は羊抗うビット■gG抗体を含
有する。
馬抗羊[G抗体がヤギIaGと強く交差反応する為に抗羊抗体試薬を使用するこ
とができる。
5O8=ドデシル硫酸ナトリウム
Renex (レネックス)R30−アトラス・ケミカル・インダストリー(英
国)製の洗浄剤(登録商標)TritonRX−100= トライトンX −1
00<登録商標、メルク・インデックス第10版、1983年、971頁、He
rck & Co、 Rahway、US、A、参照)リパーゼはシグマ・ケミ
カル・カンパニー、U、S、A、から購入した。
実験1〜4に於いて、アポ(a)は対照血清と比較して111当りのユニット(
U)で測定した。これは該結果から得られる結論とは関連のないことである。
1.1予処理
(a) アルカリ性pH0血消及びそれと等量の0.138 N a OHを混
合し、室温で2時間インキュベートし、その後、リン酸緩衝液Iで試料を18倍
に希・釈した。最初の測定′pHは12.0であり、これがインキュベーション
の間に約11.7まで減少した。
(b) 酸性pH0血清及びそれと等量の0.2HHC1を混合し、v潟で20
rt間インキュベートし、その後、リン酸緩衝液■で試料を18倍に希釈した。
最初のpHは約1でありインキュベーション中に僅かに上昇した。
(C) アルカリ性緩衡液系。血清及びそれと等量の0,2Mリン酸三ナトリウ
ムを混合し、室温で1時間インキュベートし、その後、リン酸バッファIで試料
を18倍に希釈した。
インキュベーションの間中ずつとpHは約11.7であった。
1.2 予処理ステップの抗原決定基利用性に与える影響の測定試験法
ステップ1の反応混合物50成を18倍、54倍、162倍、489倍、145
8倍、4374倍に夫々希釈した。各希釈物を50鱈のマウス抗アポ(a) 1
25T (標識抗体)と混合し、室温で1時間インキュベートし、その後50成
のヤギ抗アポ(a)を添加した。反応混合物を室温で0.511i)間装置した
。次に固相結合形態体(2dのデカンティング・サスペンション)と0.5時空
温でインキュベートし、得られた複合体を沈澱させた。懸濁液を遠心分離し、そ
の上清をデカンテーションによって除去し捨てた。残存する液体の放射活性を測
定した。得られた結果、を第1表に示す。
別の実験に於いて、ヤギ抗アポ(a)の代りに羊抗アポ(a)及びモノクローナ
ル抗アポ(a)を用いた。これらの抗体は溶液状態及び固相結合形態に於いて実
験に使用した。固相結合抗アポ(a)を使用した際にはデカンティング・サスペ
ンションを除外した。得られた結果は第1表に示されたものと一致していた。
結果は読み取ったUアポ(a)#)mKとして表示した。第1表から、本発明の
予処理が免疫反応を最適に進行させる為に決定的に重要であることが明確かつ疑
う余地なく知見される。
同様に同表から、NaOHは塩酸に較べてはるかに効果的であることが判る。試
験血清は表中に示されるように希釈された。
これらの種々の希釈物を1.1の記載に従って処理し、アポ(a)を1.2の記
載に従って定量測定した。
0.13HN a OH処理時間を増加することによって、所与の希釈物に関し
て取り込み量が減少した。これは抗原決定基が不可逆的に破壊された為と考える
ことができよう。
実施例 2
先行技術及び本発明の予処理の比較
2.1予処理
血清及びこれと同伊の予処理溶液を混合し、2〜16時間インキュベートした。
その後該試料をリン酸緩衝液■で20倍に希釈した。1.2に記載した方法に従
って反応を実施した。処理溶液、処理時間及び処理温度は第2表に示す。結果は
第3表に示す。

実施例 3
本発明の予処理を施した血清試料とアフィニティ精製したアポ(a)含有対照試
料の比較実験
3.1 1.1(a)に従って処理した血清からのアポ(a)のアフィニテイ精
製
製造者の指示する方法に従って(jl S −A −3,645,852も参照
のこと) 、 CNBr活性化5epharose 4B (Pharmaci
a AB、 Sweden)にヤギ抗アポ(a)抗血清を共有結合させた。得ら
れた20dの免疫吸着懸濁液を1.1(a)に従ってNaOH処理した試料5d
と混合し、リン酸緩衝液(1))I 7.4.0.05M)で希釈し全容量を6
5!dとした。1qられた懸濁液をカラムに詰め第4表に記載の条件で溶出させ
た。

*グリシン緩衝液の溶出液はリン酸緩衝液で中和されている。
*傘グアニジンーHCj画分は、溶出後直ちに、SephadexRG−25(
Pharmacia AB、 Sweden)に詰めたPD−10カラムで脱塩
した。
3.2 1.1(a)に従って処理したアポfalを含む血清の連続希釈物及び
既知量のアフイニテイ精製アポ(a)の連続希釈物を1.2の方法に従って測定
した。アポ(a) 濃度は006〜20U/jの範囲であった。得られた値を各
濃度に対してプロットし血清試料及びアフイニテイ精製アポ(a)について夫々
一つずつの深度依存曲線を確定した。この二つの曲線は完全に対応しており、本
発明の予処理ステップは免疫学的見地からは完全に満足すべきものであることが
示された。
実施例 4
1.2に従って分析した予処理試料及びいわゆる“ロケット”法にヱつで分析し
た未!!!lI押試斜の間の相“ロケット”分析は電気免疫測定法(Laure
ll B、 Annal、 Biochem、 Is/1966/p、45−)
を意味する。この方法はアポリポタンパク質分析に於いて最も普通に用いられる
方法の一つである。
血清試料15gを(i) 1.1(a)による予処理及び1,2による測定、及
び(ii) Laurell Bによる電気免疫測定法にかけた。この結果を第
5表に示す。

傘 “ロケット”法の低感度故にアポ(a)は測定不可。
上記結果からこれらの二つの方法によって相互に関連する値を得ることができる
ことが明らかとなった。
実施例 5
アポA■及びアポBを含む試料の本発明による予処理三種類の異なる血清試料に
ステップ11(c)の予処理(但し、18倍希釈ではなく20倍希釈)を施し、
その後適当な抗体試料を用いてステップ1.2の試験操作を行った。抗体試薬と
しては、前記“J”の項を参照のこと。予処理にかけずにリン酸緩衝液で20倍
に希釈した同じ試料も同様に1.2に従って分析した。
本発明の予処理の結果は、免疫測定によって対象アポタンパク質を測定するのに
15?すべきものであった。アポAlについては、第1図に結果を示しであるが
、図中、■及び■と記されたコラムは夫々予処理及び未予処理に対応する。縦軸
は添加量に対する抗アポAI Iの結合パーセントを示していいる。
本発明は本明IlBgの一部を構成する添付の請求の節回に於いて更に定義され
る。

Claims (4)
- 1.リボタンパク質及び/又はアポリボタンパク質の免疫化学的測定法であって 、試料と適当な抗(アポリボタンパク質)抗体又は抗(リボタンパク質)抗体と の反応に先立って、予処理ステップに於いて該試料のPHを約pH9.0より高 いか又は約PH3.0より低い非変性値に維持し、その後、該PHを免疫反応P H値に調節することで自体公知の方法での免疫化学的測定の実施を可能にするこ とを特徴とする前記測定法。
- 2.前記予処理ステップのpHがO〜3又は9.O〜14.Oの範囲内で選択さ れた値に羽節されることを特徴とする特許請求の範囲第1項に記載の測定法。
- 3.前記予処理ステップのPHが10.0より高い値に調節されることを特徴と する特許請求の範囲第1項又は第2項に記載の測定法。
- 4.前記予処理ステップのPHが9〜14の範囲内で選択された値に調節される ことを特徴とする特許請求の範囲第1項又は第2項に記載の測定法。
