Abstract
PURPOSE:To produce a pullulanase having stable activity under the action of amylase, by culturing a pullulanase-producing strain belonging to genus Baccilus stearothermophilus and collecting pullulanase from the cultured product. CONSTITUTION:A bacterial strain belonging to Baccilus stearothermophilus is selected from a bacterial group existing in a specimen such as compost, growing at about 60 deg.C and having pullulan-decomposition activity and is cultured in a medium containing soluble starch, etc. The objective pullulanase is separated from the medium. Said stearothermophilus strain is especially preferably Bacillus stearothermophilus TRS128 (FERM P-9610) having high pullulanase- productivity. The pullulanase produced by this process has high heat-resistance and is usable in combination with glucoamylase, etc., to be used at a high temperature.
