Claims (22)
- 25G 또는 그 보다 가는 바늘(needle)을 통해 주사가능한, 무균(steril) 상태의, 약제학적, 물리적으로 안정성이고 주사가능한 현탁액으로서, 하기 성분:
- 무균 상태의, 결정성의, 미분화되지 않은(non-micronized) 3-베타-히드록시-5-알파-프레그난-20-온 입자;
- 아실글리세롤의 혼합물; 및
- 콜레스테롤을 포함하며,
상기 아실글리세롤의 혼합물은 중쇄 아실글리세롤이고, 상기 중쇄 아실글리세롤은 중쇄 트리글리세리드(MCT)이며,
콜레스테롤의 농도는 0.1 내지 2.5% (w/w)이고,
상기 현탁액은 24시간 이하 내에 무균 조건하에 상기 성분을 혼합함으로써 제조되는 현탁액. - 제1항에 있어서, 상기 바늘은 27G, 또는 그 보다 가는 바늘인 현탁액.
- 제1항에 있어서, 상기 3-베타-히드록시-5-알파-프레그난-20-온의 농도는 10 mg/mL 이상인 현탁액.
- 제1항에 있어서, 현탁액을 재현탁시키지 않고 용량의 완전 회수로 적어도 12개월 보관 후에 주사가능하게 남아 있는 현탁액.

- 제1항 내지 제4항 중 어느 한 항에 따른 현탁액을 포함하는, 27G 또는 그 보다 가는 바늘이 구비된 사전-충전된 주사기(pre-filled syringe).
- 결정성의, 미분화되지 않은 3-베타-히드록시-5-알파-프레그난-20-온 입자의 제조 방법으로서,
(i) 적어도 45℃의 온도에서, 적어도 0.1% (v/v)의 수분 함량을 가진 테트라히드로푸란 용액에 3-베타-히드록시-5-알파-프레그난-20-온을 용해시키는 단계;
(ii) 단계 (i)의 용액을 10℃ 이하의 온도를 유지하는 액체 헵탄에 첨가하고, 상기 용액 혼합물을 교반하여 결정성 3-베타-히드록시-5-알파-프레그난-20-온의 침전을 수득하는 단계; 이어서;
(iii) 단계 (ii)에서 수득된 침전된 결정성 3-베타-히드록시-5-알파-프레그난-20-온을 여과하고 건조시키는 단계를 포함하며; 여기서 상기 결정성 3-베타-히드록시-5-알파-프레그난-20-온 입자의 임의의 미분화 단계가 생략되는 제조 방법. - 삭제
- 제13항에 있어서, 단계 (iii)에서 수득된 결정성의, 미분화되지 않은 3-베타-히드록시-5-알파-프레그난-20-온 입자를 그레이팅(grating)하고 체질(sieving)하는 추가 단계를 포함하는 제조 방법.
- 제13항에 있어서, 상기 테트라히드로푸란 용액은 0.5% 내지 1.5% (v/v)의 수분 함량을 갖는 제조 방법.
- 제16항에 있어서, 수분 함량을 가진 상기 테트라히드로푸란 용액은 50℃의 온도를 유지하는 제조 방법.
- 제13항 및 제15항 내지 제17항 중 어느 한 항에 있어서, 상기 액체 헵탄은 -10℃ 내지 -20℃의 온도를 유지하는 제조 방법.

- 콜레스테롤을 포함하며;
상기 현탁액은 24시간 이하 내에 무균 조건하에 상기 성분을 혼합함으로써 제조되며;
상기 결정성의, 미분화되지 않은 3-베타-히드록시-5-알파-프레그난-20-온 입자는 제13항 및 제15항 내지 제17항 중 어느 한 항에 따라 제조되는 현탁액. - 결정성의, 미분화되지 않은 3-베타-히드록시-5-알파-프레그난-20-온 입자; MCT; 및 0.1 내지 2.5% (w/w) 농도의 콜레스테롤을 포함하는, 25G 또는 그 보다 가는 바늘을 통해 주사가능한, 무균(steril) 현탁액의 제조 방법으로서,
- 결정성의, 미분화되지 않은 3-베타-히드록시-5-알파-프레그난-20-온 입자를 감마 조사(gamma irradiation)를 통해 멸균(sterilization)시키며;
- MCT를 콜레스테롤과 혼합하고 콜레스테롤이 MCT에 완전히 용해될 때까지 교반하며;
- MCT와 콜레스테롤의 혼합물을 여과하고 무균 용기에 무균적으로 넣고 상기 멸균된 3-베타-히드록시-5-알파-프레그난-20-온 입자를 첨가하고 24시간 이하 동안 혼합하고;
- 수득된 현탁액을 무균 주사기 배럴(sterile syringe barrel)에 무균적으로 충전하는 제조 방법. - 제21항에 있어서, 상기 결정성의, 미분화되지 않은 3-베타-히드록시-5-알파-프레그난-20-온 입자는 제13항 및 제15항 내지 제17항 중 어느 한 항에 따라 제조되는 제조 방법.
