Claims (8)
- ushA 유전자 및 aphA 유전자가 정상적으로 기능하지 않는, 뉴클레오사이드 5'-포스페이트 에스테르를 생산하는 능력을 지닌 에스케리키아 속에 속하는 세균을 배지중에서 배양하여 뉴클레오사이드 5'-포스페이트 에스테르를 배지중에 생산하고 축적하는 단계; 및배지로부터 뉴클레오사이드 5'-포스페이트 에스테르를 수집하는 단계를 포함하는, 뉴클레오사이드 5'-포스페이트 에스테르의 제조방법.
- 제1항에 있어서, 돌연변이가 ushA 유전자 및 aphA 유전자내로 도입되거나, 이들 유전자가 파괴되어 정상적으로 기능하지 못하는 방법.
- 제1항 또는 제2항에 있어서, 뉴클레오사이드 5'-포스페이트 에스테르가 5'-이노신산 또는 5'-구아닐산으로 이루어진 그룹으로부터 선택되는 방법.
- ushA 유전자 및 aphA 유전자가 파괴된, 뉴클레오사이드 5'-포스페이트 에스테르를 생산하는 능력을 지닌 에스케리키아 속에 속하는 세균.
- 제4항에 있어서, 뉴클레오사이드 5'-포스페이트 에스테르가 5'-이노신산 또는 5'-구아닐산으로 이루어진 그룹으로부터 선택되는 세균.
- 미생물의 모 균주, 또는 공지된 5'-뉴클레오티다제가 결실된 이의 유도체 균주를, 단일 탄소원으로서 제1 뉴클레오사이드 5'-포스페이트 에스테르를 함유하는 최소 배지 및 단일 탄소원으로서 제2 뉴클레오사이드 5'-포스페이트 에스테르를 함유하는 최소 배지중에서 배양하여 모 균주 및 유도체 균주내 유전자의 발현 프로파일을 조사하는 단계;탄소원으로서 제1 뉴클레오사이드 5'-포스페이트 에스테르를 함유하는 배지중에서 모 균주 및 유도체 균주를 배양하는 경우 이들 각각의 유전자의 발현량의 비율과, 탄소원으로서 제2 뉴클레오사이드 5'-포스페이트 에스테르를 함유하는 배지중에서 모 균주 및 유도체 균주를 배양하는 경우 이들 각각의 유전자의 발현량의 비율의 곱을 계산하는 단계; 및곱의 수치가 보다 높은 유전자를 하나 이상 선택하는 단계를 포함하여, 뉴클레오사이드 5'-포스페이트 에스테르의 축적에 영향을 미치는 5'-뉴클레오티다제 유전자를 탐색하는 방법.
- 제6항에 있어서, 제1 및 제2 뉴클레오사이드 5'-포스페이트 에스테르가 5'-이노신산 및 5'-구아닐산인 방법.
- 제6항 또는 제7항에 있어서, 단백질을 선택된 유전자로부터 주변세포질로 이동시키는데 필요한 시그널 시열을 암호화할 수 있는 유전자를 선택하는 단계를 추가로 포함하는 방법.
