Claims (31)
- 제1 혈청형의 아데노바이러스의 구조적 및 비-구조적 요소들을 포함하는 재조합 아데노바이러스 벡터로, 상기 벡터는 기능적 E4-orf6 단백질, 그의 기능적 일부, 유도체 및/또는 동족체를 암호화하는 서열을 추가로 포함하고, 여기서 상기 서열은1. 상기 제1 혈청형과 다른 제2 혈청형의 아데노바이러스로부터 유도된 E4-orf6 암호화 서열;2. 하나 이상의 코돈에서 결실, 돌연변이, 첨가 및/또는 치환을 포함하는 상기 제1 혈청형의 아데노바이러스로부터 유도된 E4-orf6 암호화 서열; 및3. 상기 제1 혈청형과 다른 제2 혈청형으로부터 유도된 E4-orf6 암호화 서열의 일부와, 제3 혈청형으로부터 유도된 E4-orf6 암호화 서열의 일부 사이의 융합물을 포함하는 (여기서 상기 제3 혈청형은 상기 제1 혈청형과 동일하거나 또는 다를 수 있다) E4-orf6 암호화 서열로 이루어진 군으로부터 선택되는 것인 재조합 아데노바이러스 벡터.

- 제 1항에 있어서, 상기 제1 혈청형은 서브그룹 B로부터 유래되고, 상기 제2 혈청형은 서브그룹 C로부터 유래된 재조합 아데노바이러스 벡터.
- 제 1항에 있어서, 상기 E4-orf6 암호화 서열은 아데노바이러스 혈청형 5로부터 유도된 재조합 아데노바이러스 벡터.
- 제 1항 내지 제 5항 중 어느 하나의 항에 있어서, 상기 제1 혈청형이 아데노바이러스 혈청형 11, 14, 16, 21, 34, 35 및 50으로 이루어진 군으로부터 선택되는 것인 재조합 아데노바이러스 벡터.
- 제 1항 내지 제 6항 중 어느 하나의 항에 있어서, 비-아데노바이러스 단백질, 폴리펩티드 또는 펩티드를 암호화하는 서열을 추가로 포함하는 재조합 아데노바이러스 벡터.
- 제 7항에 있어서, 상기 비-아데노바이러스 단백질, 폴리펩티드 또는 펩티드는; 세포-치사 유도 폴리펩티드, 병원성 유기체의 항원성 결정기, 종양 특이 항원, 바이러스성 단백질, 호르몬 및 시토킨으로 이루어진 군으로부터 선택되는 재조합아데노바이러스 벡터.
- 제1 혈청형의 아데노바이러스의 구조적 및 비-구조적 요소들을 포함하는 재조합 아데노바이러스 벡터의 생산방법으로, 상기 방법은a. 발현 형태로 제2 혈청형의 아데노바이러스로부터 유도된, E1B-55K 암호화 서열, 그들의 기능적 일부, 유도체 및/또는 동족체를 정착시키고 있는 보충 세포에, 상기 보충 세포에 의한 상기 재조합 아데노바이러스 벡터의 조립을 가능하게 하는 아데노바이러스의 필수 요소들을 제공하는 단계, 여기서 상기 요소들은 상기 제2 혈청형과 다른 상기 제1 혈청형의 아데노바이러스로부터의 몇몇 이상의 구조적 및 비-구조적 요소들 및 (상기 보충 세포의 상기 발현가능한 E1B-55K 단백질과 양립할 수 있는) 기능적 E4-orf6 단백질의 암호화 서열, 또는 그들의 기능적 일부, 유도체 및/또는 동족체를 포함하고;b. 상기 보충 세포를 아데노바이러스 벡터의 생산 및 조립이 일어나는 것을 가능하게 하는 조건하의 배지에서 배양시키는 단계; 및c. 그렇게 생산된 재조합 아데노바이러스를 배지 및/또는 보충 세포로부터 수확하는 단계로 이루어지며,여기서 상기 양립가능한 E4-orf6 단백질을 암호화하는 서열은 그렇게 생산된 재조합 아데노바이러스 벡터에 존재하는 방법.
