Claims (16)
- 하기 단계를 포함하는 샘플 내 항-약물 항체(Anti-Drug Antibody: ADA)의 면역-검출 방법으로서,
상기 약물은 표적 항원에 대항하는 치료적 항체인 방법:
a) 샘플 내 상기 항원의 존재를 입증하는 단계;
b) 샘플 내 상기 항원이 존재하는 경우, 항원을 중화시키는 단계; 및
c) 상기 샘플 내 항-약물 항체의 존재 또는 양을 면역-검출하는 단계. - 치료적 항체 치료에 대한 환자의 반응성의 검출 방법으로서,
환자로부터의 샘플에서 하기의 존재 또는 양을 측정하여 환자 프로파일을 획득하는 단계를 포함하고, 상기 치료적 항체는 표적 항원에 대항하는 것이고, 상기 환자 프로파일은 상기 치료에 대한 반응성을 나타내는 것인 방법:
- 치료적 항체,
- 항원, 및
- 상기 치료적 항체에 대항하는 항-약물 항체. - 제2항에 있어서,
상기 측정은 면역검정, 바람직하게는 ELISA 또는 면역-포획에 의해 수행되는 것을 특징으로 하는 방법. - 제1항 내지 제3항 중 어느 한 항에 있어서,
상기 항원은 TNFα인 것을 특징으로 하는 방법. - 제4항에 있어서,
상기 약물은 인플릭시마브, 에타네르셉트, 아달리무마브, 골리무마브 및 세르톨리주마브 페골로부터 선택된 항-TNFα 항체인 것을 특징으로 하는 방법. - 제2항 내지 제5항 중 어느 한 항에 있어서,
상기 환자는 자가면역 또는 염증성 질환을 갖는 것을 특징으로 하는 방법. - 제2항 내지 제6항 중 어느 한 항에 있어서,
상기 환자로부터의 샘플에서 C-반응성 단백질(C-reactive protein: CRP) 또는 적혈구 침강 속도(Erythrocyte sedimentation rate: ESR)의 존재 또는 양을 측정하는 단계를 추가로 포함하는 것을 특징으로 하는 방법. - 제2항 내지 제7항 중 어느 한 항에 있어서,
상기 환자의 치료를 모니터링 또는 조절하기 위한 방법. - 제1항 내지 제8항 중 어느 한 항에 있어서,
상기 샘플은 체액 샘플, 바람직하게는 혈액 혈청 또는 혈장 샘플인 것을 특징으로 하는 방법. - 제8항에 있어서,
환자로부터의 샘플에서 하기의 존재 또는 양을 측정하여 환자 프로파일을 획득하는 단계를 포함하고, 상기 환자 프로파일은 환자가 상기 치료에 반응하는지 여부를 나타내어, 치료의 조정을 가능하게 하는, 항-TNA알파 치료를 받는 환자를 모니터링하기 위한 방법:
- 치료적 항체,
- 항원, 및
- 상기 치료적 항체에 대항하는 항-약물 항체. - 제10항에 있어서,
상기 TNF알파 수준이 10pg/ml 미만이고, 상기 치료적 항체 수준이 1μg/ml 초과이고, 상기 항-약물 항체 수준이 50 ng/ml 미만인 경우, 상기 환자는 반응성이고 상기 치료는 지속되거나 또는 감소되어야 하는 것을 특징으로 하는 방법. - 제1항 내지 제11항 중 어느 한 항에 있어서,
비활성 TNF알파의 수준이 검출되는 것을 특징으로 하는 방법. - 제12항에 있어서,
상기 비활성 TNF알파의 수준이 10pg/ml 초과인 경우, 상기 환자는 상기 치료에 대해 비-반응성인 것을 특징으로 하는 방법. - 치료적 항체 치료에 대한 환자의 반응성의 검출 방법으로서,
환자로부터의 샘플에서 비활성 TNF알파의 존재 또는 양을 측정하는 단계를 포함하고, 상기 치료적 항체는 TNF알파에 대항하는 것이고, 상기 존재 또는 양은 상기 치료에 대한 환자의 낮은 반응성 또는 감소하는 반응성의 지표인 방법. - 항-TNF알파 치료를 받는 환자의 모니터링 방법으로서,
환자로부터의 샘플에서 비활성 TNF알파의 존재 또는 양을 측정하는 단계를 포함하고, 상기 존재 또는 양은 상기 치료에 대한 환자의 반응성의 지표인 방법. - 샘플에서 하기의 존재 또는 양을 검출하기 위한 시약을 포함하는 키트:
- 표적 항원에 대항하는 치료적 항체,
- 항원, 및
- 상기 치료적 항체에 대항하는 항-약물 항체.
