Abstract
Description
도 2는 분리된 본 발명의 융합 단백질(oFN) 및 피브로넥틴 단백질 제9 및 10 타입 Ⅲ 도메인(iFN)을 쿠마시 블루 염색(A) 및 웨스턴 블랏(B)을 수행하여 확인한 결과를 나타낸 도이다.
도 3은 본 발명의 융합 단백질(oFN), 피브로넥틴 단백질 제9 및 10 타입 Ⅲ 도메인(iFN) 및 골형성 단백질-2(BMP-2)을 농도별로 조골세포에 처리한 후 세포 부착 활성을 확인한 결과를 나타낸 도이다.
도 4는 본 발명의 융합 단백질(oFN) 및 피브로넥틴 단백질 제9 및 10 타입 Ⅲ 도메인(iFN)을 조골세포에 처리하고 3 또는 10일 후 ALP 활성을 확인한 결과를 나타낸 도이다.
도 5는 무처리군, oFN 융합 단백질이 봉입된 콜라겐 매트릭스에서 자란 MC3T3-E1 세포 또는 iFN 단백질이 봉입된 콜라겐 매트릭스에서 자란 MC3T3-E1 세포의 ALP 활성을 확인한 결과를 나타낸 도이다.
도 6은 무처리군, oFN 융합 단백질이 봉입된 콜라겐 매트릭스에서 자란 MC3T3-E1 세포 또는 iFN 단백질이 봉입된 콜라겐 매트릭스에서 자란 MC3T3-E1 세포에서 골 형성 마커인 OPN, RUNX2 및 Col I의 mRNA 발현 수준을 나타낸 도이다.

- 피브로넥틴(Fibronectin, FN) 단백질 및 골 형성 단백질-2(Bone morphogenetic protein-2, BMP-2)가 결합된 융합(fusion) 단백질.
- 제1항에 있어서,
상기 피브로넥틴 단백질은 제9타입 Ⅲ 도메인(FNⅢ9) 및 제10타입 Ⅲ 도메인(FNⅢ10)인 것을 특징으로 하는 융합 단백질. - 제2항에 있어서,
상기 피브로넥틴 단백질의 제9타입 Ⅲ 도메인(FNⅢ9)은 서열번호 1로 표시되는 아미노산으로 이루어진 것을 특징으로 하는 융합 단백질. - 제2항에 있어서,
상기 피브로넥틴 단백질의 제10타입 Ⅲ 도메인(FNⅢ10)은 서열번호 2로 표시되는 아미노산으로 이루어진 것을 특징으로 하는 융합 단백질. - 제1항에 있어서,
상기 골 형성 단백질-2은 서열번호 3으로 표시되는 아미노산으로 이루어진 것을 특징으로 하는 융합 단백질. - 제1항에 있어서,
상기 융합 단백질은 서열번호 4로 표시되는 아미노산으로 이루어진 것을 특징으로 하는 융합 단백질. - 제1항의 융합 단백질을 코딩하는 폴리뉴클레오티드.
- 제7항에 있어서,
상기 폴리뉴클레오티드는 서열번호 5로 표시되는 염기 서열로 이루어진 것을 특징으로 하는 폴리뉴클레오티드. - 제7항의 폴리뉴클레오티드를 포함하는 재조합 벡터.
- 제9항에 있어서,
상기 재조합 벡터는 도 1의 개열 지도를 갖는 것을 특징으로 하는 재조합 벡터. - 제1항 내지 6항 중 어느 한 항에 따른 융합 단백질을 유효성분으로 포함하는 골 재생용 조성물.
- 제1항 내지 6항 중 어느 한 항에 따른 융합 단백질을 유효성분으로 포함하는 골 재생용 스캐폴드.
