Description
도 2는 간접 저해 ELISA법에 의한 반응계의 모식도이다.
도 3은 간접 저해 ELISA법에 의한 클론 TAT-5의 각 항원으로의 결합성을 평가한 결과를 나타내는 도면이다.
도 4는 각 항체를 결합시킨 라텍스 시약에 의한 TAT로의 반응성의 평가 결과를 나타내는 도면이다.
도 5는 각 항체를 결합시킨 라텍스 시약에 의한 유리 안티트롬빈으로의 교차 반응성 평가 결과를 나타내는 도면이다.
도 6은 pH를 6.0~7.2 사이에서 변화시킨 경우의 베이스 흡광도의 변화를 나타내는 도면이다.
도 7은 pH를 6.0~7.2 사이에서 변화시킨 경우의 혈장 베이스를 차감한 TAT 반응성에 대한 영향을 나타내는 도면이다.

Claims (8)
- 트롬빈 안티트롬빈 복합체(TAT) 측정 시약으로서,
라텍스 입자에 결합한, 안티트롬빈 측에 결합하여 TAT를 인식하는 항체, 및
라텍스 입자에 결합한, 트롬빈 측에 결합하여 TAT를 인식하는 항체를 포함하고,
상기 안티트롬빈 측에 결합하여 TAT를 인식하는 항체는, TAT에 대한 반응성이 유리 안티트롬빈에 대한 반응성의 100배 이상인 것을 특징으로 하는, TAT 측정 시약.
- 제1항에 있어서,
측정시의 pH가 5.8~6.6이 되도록 구성되어 있는 것을 특징으로 하는, TAT 측정 시약.
- 제1항 또는 제2항에 있어서,
상기 안티트롬빈 측에 결합하여 TAT를 인식하는 항체가 결합한 라텍스 입자 및 상기 트롬빈 측에 결합하여 TAT를 인식하는 항체가 결합한 라텍스 입자를 포함하는 제2 시약, 및
pH가 5.8~6.6인 완충액을 포함하는 제1 시약을 포함하는, TAT 측정 시약.
- 생체로부터 분리된 시료 중에 존재하는 TAT를 측정하는 방법으로써, 제1항 내지 제3항 중 어느 한 항에 기재된 TAT 측정 시약을 이용하여 라텍스 응집법을 실시함으로써 TAT를 측정하는 것을 특징으로 하는 방법.
- 라텍스 응집법에 따라서, 생체로부터 분리된 시료중에 존재하는 TAT를 측정할 때에 사용되는 항체를 스크리닝 하는 방법으로써, 항체의 후보 항체를 준비하는 공정, 후보 항체를 일정량의 유리 안티트롬빈과 반응시키는 공정, 상기 공정 후에 얻어진 반응액을 TAT가 고정화된 기재를 이용한 효소 면역 측정법에 제공함으로써 항체의 유리 안티트롬빈에 대한 반응성을 측정하는 공정, 상기 유리 안티트롬빈에 대한 반응성을 항체의 TAT에 대한 반응성과 비교하는 공정, 및 TAT에 대한 반응성이 유리 안티트롬빈에 대한 반응성의 100배 이상인 항체를 선택하는 공정을 포함하는, 항체의 스크리닝 방법.
- 트롬빈 안티트롬빈 복합체(TAT) 측정 시약으로서,
라텍스 입자에 결합한, 안티트롬빈 측에 결합하여 TAT를 인식하는 제1 항체, 및
라텍스 입자에 결합한, 트롬빈 측에 결합하여 TAT를 인식하는 제2 항체를 포함하고,
측정 시의 pH가 5.8~6.6이 되도록 구성되어 있는 것을 특징으로 하는, TAT 측정 시약.
- 제6항에 있어서,
상기 제1 항체가 결합한 라텍스 입자 및 상기 제2 항체가 결합한 라텍스 입자를 포함하는 제2 시약, 및
pH가 5.8~6.6의 완충액을 포함하는 제1 시약을 포함하는, TAT 측정 시약.
- 생체로부터 분리된 시료 중에 존재하는 시료 중의 TAT를 측정하는 방법으로써, 제6항 또는 제7항에 기재된 TAT 측정 시약을 이용하여 라텍스 면역 응집 반응을 pH가 5.8~6.6인 조건에서 실시함으로써 TAT를 측정하는 것을 특징으로 하는 방법.
