Abstract
Description
도 2A 내지 2D는 겔화제에 따른 CS/TOCNF 하이드로겔의 37 ℃에서의 겔화시간, 점도, 유동 거동을 나타낸 것이다.
도 3은 겔화제에 포함된 중탄산나트륨의 농도에 따른 CS/TOCNF 하이드로겔의 푸리에 변화 적외선(FT-IR) 스펙트럼을 나타낸 것이다.
도 4A 및 4B는 겔화제에 포함된 중탄산나트륨(0M, 0.1M, 0.25M, 0.5M)의 농도에 따른 CS/TOCNF 하이드로겔을 리소자임 효소(1 mg/㎖)를 함유한 PBS에 침지된 경우에 분석한 프로파일로 잔존량과 광학사진을 나타낸 것이다.
도 5는 배양 1일, 3일, 5일 및 7일 후 전조골세포 MC3T3-E1의 증식에 미치는 fk506(5 nM, 15 nM, 25 nM, 50 nM 및 100 nM)의 효과를 MTT 분석한 것이다.
도 6은 겔화제 중 0.5M 중탄산나트륨을 포함하는 CS/TOCNF 하이드로겔의 fk506의 누적 방출 속도를 UV 분광법으로 나타낸 것이다.
도 7은 임계 크기(Critical- sized)의 쥐 두개골 결손부에 fk506이 포함된 CS/TOCNF 하이드로겔을 이식한 모습을 나타낸 것이다.
도 8은 동물 모델에 fk506이 포함된 CS/TOCNF 하이드로겔 이식 한 후 3주, 6 주 후의 마손 트리크롬 (Masson's Trichrome)으로 염색 결과를 나타낸 것이다.
Claims (14)
- fk506, 키토산, 겔화제, 및 템포 산화 셀룰로오스 나노 섬유를 포함하는, 골치료를 위한 fk506을 포함하는 키토산/템포 산화 셀룰로오스 나노 섬유 하이드로겔.
- 제1항에서,
상기 겔화제는,
0.1~1.0 M 중탄산나트륨(NaHCO3) 및 β-글리세로포스페이트(β-Glycerophosphate)를 포함하는 것을 특징으로 하는 키토산/템포 산화 셀룰로오스 나노 섬유 하이드로겔. - 제1항에서,
상기 하이드로겔은, 생체 내에서(In vivo) 겔화되는 것을 특징으로 하는 키토산/템포 산화 셀룰로오스 나노 섬유 하이드로겔. - 제2항에서,
상기 하이드로겔은 상기 중탄산나트륨의 함량이 증가하면 졸에서 겔로 변환되는 속도가 증가되는 것을 특징으로 하는 키토산/템포 산화 셀룰로오스 나노 섬유 하이드로겔. - 제2항에서,
상기 하이드로겔은 상기 중탄산나트륨의 함량과 하이드로겔의 분해 속도가 반비례하는 것을 특징으로 하는 키토산/템포 산화 셀룰로오스 나노 섬유 하이드로겔. - 제1항에서,

상기 중탄산나트륨은, 하이드로겔의 분해 시간 및 약물 방출 시간을 지연시키는 것을 특징으로 하는 키토산/템포 산화 셀룰로오스 나노 섬유 하이드로겔. - (a) 키토산을 락트산 수용액에 투입하여 키토산 용액을 제조하는 단계,
(b) 템포 산화 셀룰로오스 나노 섬유(Tempo-oxidized cellulose nanofiber) 용액을 제조하는 단계,
(c) 상기 키토산 용액과 상기 템포 산화 셀룰로오스 나노 섬유(TOCNF) 용액을 혼합하여 1차 혼합액을 제조하는 단계,
(d) fk506 용액을 제조한 후 상기 1차 혼합액에 혼합하여 2차 혼합액을 제조하는 단계,
(e) 겔화제를 제조하는 단계, 그리고
(f) 상기 2차 혼합액에 겔화제를 적하하는 단계를 포함하는, 골치료를 위한 fk506을 포함하는 키토산/템포 산화 셀룰로오스 나노 섬유 하이드로겔의 제조방법. - 제10항에서,
상기 (c) 단계는,
상기 TOCNF 용액과 상기 키토산 용액을 1:2~4 부피비(v/v)로 혼합하는 것을 특징으로 하는 키토산/템포 산화 셀룰로오스 나노 섬유 하이드로겔의 제조방법. - 제10항에서,
상기 (e) 단계는,
0.1~1 M 농도의 중탄산나트륨(NaHCO3) 수용액에 60 %(w/v)의 β-글리세로포스페이트(β-Glycerophosphate) 파우더를 용해하여 제조하는 것을 특징으로 하는 키토산/템포 산화 셀룰로오스 나노 섬유 하이드로겔의 제조방법. - 제10항에서,
상기 fk506 용액은 에탄올에 fk506 분말을 용해한 것임을 특징으로 하는 키토산/템포 산화 셀룰로오스 나노 섬유 하이드로겔의 제조방법. - 제10항에서,
상기 (f)단계는,
