Abstract
Description
도 1은 제대혈 (CB) 또는 인간 태반 관류액 (HPP) 중 CD34 및/또는 CD45를 발현하는 유핵 관류액 세포의 비율을 도시한다.
도 2는 제대혈 (CB) 또는 인간 태반 관류액 (HPP)으로부터 수득된, CD31, CXCR4 및/또는 VEGFR을 발현하는 CD34+ 세포의 비율을 도시한다.
도 3은 HPP CD34+CD45- 및 CD34+CD45+ 세포에서 실시된 qRT-PCR에 의한 유전자 발현 분석 결과를 도시한다. 인간 태반 관류액 CD34+, CD45- 및 CD34+, CD45+ 세포 중의 CD34 및 CD45의 상대적인 발현값은 제대혈로부터 수득된 CD34+ 세포 중의 CD34 및 CD45의 발현값으로 정규화된 값이다. 상대적인 정량값 (RQ) (Y축)은 2-ΔΔCt으로 제시한다.
도 4는 길즈 변형된 헤마톡 실린(Gill's Modified Hematoxylin) 염료로 염색된 CFU-Hill 콜로니 (배율 200X)를 도시한다. 콜로니는 엔도컬트(ENDOCULT)® 배지에서 2주 동안 배양된 인간 태반 관류액 세포 배양물로부터 발생하였다.
도 5는 96-웰 플레이트의 각 웰당 106개의 세포로 EC매트릭스 상에서 18-24시간 동안 배양된 HPP 세포에 의한 혈관 형성을 도시한다. 배율 200X.
도 6은 HPP에 의한 생체내 골 형성 활성을 도시한다.
도 7A, 7B는 이식 후 21일 경과시에 스캐폴드 중심부를 촬영한 영상이다. (A) 비토스(Vitoss) 및 HPP 세포 (B) 세포를 포함하지 않는 비토스. 도 6A에 나타난 바와 같이, 다수의 CD34 양성 세포 (화살표로 표시)는 혈관 인접부에서 관찰할 수 있다. 원래 배율 200X, 축적 막대 100 ㎛; 원 색상: aSMA 적색 (AF594), CD34 녹색 (플루오레세인).
도 8A, 8B는 이식 후 42일 경과시에 스캐폴드 중심부를 촬영한 영상이다. (A) 비토스 및 HPP 세포 (B) 세포를 포함하지 않는 비토스. 도 7A에 나타난 바와 같이, CD34 양성 세포 (화살표로 표시)는 혈관 인접부에서 관찰할 수 없다. 원래 배율 200X, 축적 막대 100 um, aSMA 적색 (AF594).
도 9는 21일째에 두 동물군에서, HPP 세포의 사용으로 혈관형성이 통계학상 유의적으로 증강되었음을 나타내는 영상 분석 결과이다. Y축 - 알파 평활근 액틴 발현율(%).
Claims (11)
- CD34+ 인간 태반 관류액 세포를 개체에서 혈관형성을 촉진시키기에 충분한 양으로 포함하는, 개체에서 혈관형성을 촉진시키기 위한 제약 조성물.
- 제1항에 있어서, 상기 개체가 심장 또는 혈관의 기능부족(insufficiency) 또는 결손(defect)을 앓고 있는 것인 제약 조성물.
- 제2항에 있어서, 상기 심장 또는 혈관의 기능부족 또는 결손이 말초 혈관 질환, 급성 또는 만성 심근경색증, 심근병증, 울혈성 또는 만성 심부전, 심혈관 허혈, 고혈압성 폐 혈관 질환, 말초 동맥 질환, 또는 류마티스 심장병과 연관된 기능부족 또는 결손인 약제학적 조성물.
- 제1항에 있어서, 상기 개체가 뼈 결손을 앓고 있는 것인 제약 조성물.
- 제1항 내지 제4항 중 어느 한 항에 있어서, 상기 인간 태반 관류액 세포가 단일의 태반 관류로부터 단리된 인간 태반 관류액으로부터의 전체 유핵 세포를 포함하는 것인 약제학적 조성물.
- 제1항 내지 제4항 중 어느 한 항에 있어서, 상기 약제학적 조성물이 태반 관류액으로부터 단리된 것 이외의 단리된 CD34+ 세포를 추가로 포함하는 것인 약제학적 조성물.
- 제6항에 있어서, 상기 태반 관류액으로부터 단리된 것 이외의 CD34+ 세포가 태반으로부터 단리된 것인 약제학적 조성물.
- 제6항에 있어서, 상기 태반 관류액으로부터 단리된 것 이외의 CD34+ 세포가 제대혈, 태반 혈액, 말초 혈액, 또는 골수로부터 단리된 것인 약제학적 조성물.
- 제1항 내지 제4항 중 어느 한 항에 있어서, 상기 CD34+ 인간 태반 관류액 세포가 제대혈로부터 수득된 같은 개수의 CD34+ 세포보다 더 높은 수준으로 CD31, CXCR4 또는 VEGFR을 발현하는 것인 약제학적 조성물.
- 제1항 내지 제4항 중 어느 한 항에 있어서, 상기 CD34+ 인간 태반 관류액 세포 및/또는 태반 관류액으로부터 단리된 것 이외의 CD34+ 세포가 CD34+CD45- 세포인 약제학적 조성물.
- 제1항 내지 제4항 중 어느 한 항에 있어서, 상기 조성물이 인간 태반 관류액으로부터의 전체 유핵 세포를 포함하는 것인 약제학적 조성물.
