Claims (17)
- a. 에틸 (S)-7-메톡시-9-옥소-11,12,13,13a-테트라하이드로-9H-벤조[e]이미다조[5,1-c]피롤로[1,2-a][1,4]디아제핀-1-카르복실레이트 (L-655,708);
b. 3-브로모-10-(디플루오로메틸)-9H-벤조[f]이미다조[1,5-a][1,2,4]트리아졸로[1,5-d][1,4]디아제핀 (RO4938581);
c. N-벤질-6-에톡시-4-옥소-1H-1,5-나프티리딘-3-카르복스아미드 (CP-457,920);
d. 3-tert-부틸-7-(5-메틸이속사졸-3-일)-2-(1-메틸-1H-1,2,4-트리아졸-5-일메톡시)피라졸로(1,5-d)(1,2,4)트리아진 (MRK-016); 및
e. (1,1-디옥시도티오모르폴리노)(6-((3-(4-플루오로페닐)-5-메틸이속사졸-4-일)메톡시)피리딘-3-일)메탄온 (RG-1662)로 이루어진 군에서 선택되고,

(b) 단계 (a)의 생성물을 6-클로로니코티노니트릴 또는 메틸 6-클로로니코티네이트에 첨가하는 단계;
(c) 단계 (b)의 생성물을 카르복실산으로 가수분해하는 단계; 및
(d) 단계 (c)의 생성물을 티오모르폴린 1,1-디옥사이드 또는 이의 염과 아미드 커플링시키는 단계로 합성된 청구항 1의 중수소화된 바스미사닐 화합물. - 제4항에 있어서, 여기서 상기 중수소 공여체는 D2O 또는 CD3OD인 화합물.
- 6-((3-(4-플루오로페닐)-5-메틸이속사졸-4-일)메톡시)니코티노니트릴;
6-((3-(4-플루오로페닐)-5-메틸이속사졸-4-일)메톡시)니코틴아미드; 및
6-((3-(4-플루오로페닐)-5-메틸이속사졸-4-일)메톡시)니코틴산으로 이루어진 군으로부터 선택된 화합물을 중수소-함유 용매의 존재하의 염기로 처리하는 단계를 포함하는 중수소화된 RG-1662 화합물을 생성하는 방법. - 제7항에 있어서, 여기서 상기 중수소-함유 용매는 D2O인 방법.
- 같은 구조의 비-중수소화된 화합물보다 포유동물에 투여되어 더 긴 생물학적 반감기를 가지는 청구항 1의 중수소화된 GABAA5-NAM 화합물.
- 대상에 청구항 1 또는 청구항 4의 중수소화된 GABAA5-NAM 화합물의 치료적으로 유효한 양을 투여하는 단계를 포함하는, 이를 필요로 하는 인간 대상에서 우울증-관련 장애를 치료하는 방법.
