Abstract
Description
도 2는 배액 림프절에서 R-DOTAP 유도된 I형 IFN 유전자 발현을 입증한 그래프를 제공한다. 도 2의 데이터는 C57BL/6J 마우스 (n=2)가 50 ㎕의 6 mM R-DOTAP 또는 S-DOTAP 나노입자를 발바닥에 피하로 주사된 실험에 상응한다. B) FLT3-유도된 BMDC (pDC) 및 GM-CSF/IL-4 유래 BMDC (cBMDC)는 표시된 농도 (6-400 μM)의 양이온성 지질 또는 LPS 양성 대조군 (1-500 ng/mL)으로 24시간 동안 자극되었다. BMDC 분비된 I형 인터페론은 리포터 세포주에서 재조합 마우스 IFN-β 자극된 SEAP 활성을 사용해 생성시킨 표준 그래프를 사용해 정량하였다.
도 5는 I형 IFN 신호 결함 마우스에서 R-DOTAP 기반 백신 효율을 입증하는 그래프를 제공한다. 도 5의 데이터는 Jackson의 C57BL/6J 마우스 (n=6) 또는 IFNAR -/- 마우스가 SIINFEKL 펩티드 (A) (SEQ ID NO: 1) 또는 RF9 펩티드 (B) (SEQ ID NO: 2)를 함유하는 100 ㎕의 백신 제제 (Versamune® 또는 완전 프로인트 보강제)를 0일 및 7일에 피하로 주사된 실험에 상응한다. 2차 백신접종 이후 7일에, 백신접종 및 대조군 마우스로부터의 비장은 야생형 및 IFNAR-/- 마우스에서 백신 제제에 의해 유도된 항원-특이적 CD8 T 세포 반응을 측정하기 위해 처리되었다. 항원-특이적 CD8T 세포 반응은 ELISPOT 어세이에서 IFN-γ의 SIINFEKL 특이적 (A) (SEQ ID NO: 1) 및 RF9 펩티드 특이적 (B) (SEQ ID NO: 2) CD8 T 세포 생산을 측정하여 평가하였다. 데이터는 유사한 결과의 다수 실험을 대표한다. 데이터의 통계적 유의성은 단측 ANOVA를 사용해 추정하였고, 다중 비교는 터키 검정을 사용해 분석하였다. ** WT Versamune® 백신접종 그룹과 비교된 통계적 유의성 (p<0.05).
Claims (25)
- 대상체에서 I형 IFN 신호전달 경로를 변형시키기 위한 방법으로서, 포유동물에게 양이온성 지질을 투여하는 단계를 포함하는 것인 변형 방법.
- 제1항에 있어서, 양이온성 지질은 1,2-디올레오일-3-트리메틸암모늄 프로판 (DOTAP), N-1-(2,3-디올레오일옥시)-프로필-N,N,N-트리메틸 암모늄 클로라이드 (DOTMA), 1,2-디올레오일-sn-글리세로-3-에틸포스포콜린 (DOEPC), 및 이의 조합을 포함하는 것인 변형 방법.
- 제2항에 있어서, 양이온성 지질은 양이온성 지질의 거울상 이성질체를 포함하는 것인 변형 방법.
- 제3항에 있어서, 양이온성 지질은 1,2-디올레오일-3-트리메틸암모늄 프로판 (DOTAP)인 변형 방법.
