Abstract
La presente invención proporciona métodos y sistemas de espectroscopía Raman in situ para monitorear y controlar una o más variables de proceso en un cultivo celular de biorreactor para mejorar la calidad y la consistencia del producto. Los métodos y sistemas utilizan espectroscopía Raman in situ y técnicas de modelado quimiométrico para las evaluaciones en tiempo real de los cultivos celulares, combinadas con técnicas de procesamiento de señales, para la retroalimentación continua precisa y control predictivo del modelo de variables de proceso de cultivo celular. Mediante el uso de datos en tiempo real de la espectroscopía Raman, las variables del proceso dentro del cultivo celular pueden monitorearse de forma continua o intermitente y los controladores de retroalimentación automáticos mantienen las variables del proceso en los puntos de ajuste predeterminados o mantienen un protocolo de alimentación específico que administra cantidades variables de agentes al biorreactor para maximizar la calidad del bioproducto.
