Description
Foreliggende oppfinnelse vedrører fremstilling av nye pyridyl- og pyrimidylderivater av den art som er angitt i krav l's ingress.
Det er et betydelig behov for effektive medikamenter for behandling av mentale lidelser, medikamenter som er mer effektive og har færre bivirkninger enn de medikamenter som er i klinisk bruk idag. Antipsykotiske medikamenter som er i bruk, medfører en rekke brysomme ekstrapyramidale beve-gelsesforstyrrelser (dvs. akutte dystoniske reaksjoner og tardiv dyskinesia), og de har liten innvirkning på de nega-tive symptomer (dvs. begrenset eller sløvede emosjonelle følelser) ved schizofreni. Den største ulempe med anti-depressiva er at de ikke lindrer depresjonene hos 30 til 40 % av pasientene. Anxiolytika er vanligvis assosiert med avhengighet.
Det er kjent forskjellige pyridyl- og pyrimidylpiperazin-derivater som er farmakologisk aktive i sentralnervesystemet. Det kan nevnes noen representative eksempler. Azaperone, et neuroleptisk medikament i butyrfenon-serien, er et sedativ for griser. Buspirone er et anxiolytisk middel. Den anxiolytiske effekt er antatt å formidles via innvirkning
på 5HT-reseptorene.
Forskjellige andre pyridyl- og pyrimidinylpiperazinderiva-ter med aktivitet i sentralnervesystemet er kjent, f.eks. fra:

Av disse må (1), særlig eks. 4, anses å representere den nærmest beslektede teknikkens stand.
Pyridyl- og pyrimidyl-piperaziner substituert i 4-posisjon på piperazin-ringen med en fenyl-butyl eller en fenoksy-propyl-gruppe har uventet vist seg å ha farmakologiske egenskaper som er overlegne i forhold til kjente forbindelser. 5-HT2-reseptoraffinitets-tester ble anvendt for å vise overlegenheten hos foreliggende oppfinnelses forbindelser sammenlignet med forbindelsen beskrevet i eks. 4 i (1) samt med Buspirone. De representative forbindelser fra foreliggende oppfinnelse var: 2:4 4-[3-(p-fluorfenoksy)propyl]-1-[2-pyridyl]piperazin, 1:1 4-[4-(p-fluorfenyl)butyl]-l-(2-pyridyl)piperazin, 2:19 4-[3-(p-fluortiofenoksy)propyl]-1-[3-karbamyl-2-pyridyl]piperazin,
2:20 4-[3-(p-fluortiofenoksy)propyl]-1-[2-pyridyl]piperazin,
2:9 4-[4-(p-fluorfenyl)butyl]-1-[6-klor-2-pyridyl]piperazin,
2:12 4-[4-(p-fluorfenyl)butyl]-2-[3-etoksy-2-pyridyl]piperazin,
2:17 4-[3-(p-fluorfenoksy)propyl]-1-[3-hydroksy-2-pyridyl]-piperazin,
2:22 4-[4-(p-fluorfenyl)butyl]-l-[3-piperidinokarbonyl-2-pyridyl]piperazin.
Forbindelsen fra eks. 4 i (1) er: N-(fenyl-l-butyl-4)N'-(pyridyl-2)piperazin.
Azaperone er sterkt sedativt, mens forbindelsene fremstilt ifølge foreliggende oppfinnelse ikke viser noen sedativ virkning i trappetrinn-testen.
Som det fremgår av Tabell 1, har forbindelsene fra foreliggende oppfinnelse vesentlig høyere affinitet for 5HT2-reseptoren enn forbindelsen fra eks. 4 i (1), idet de fleste har minst ti ganger bedre verdier.
Foreliggende oppfinnelse vedrører analogifremgangsmåte ved fremstilling av terapeutisk aktive forbindelser med den generelle formel (I) hvor Ri er fluor eller hydrogen, og R2 er fluor, X er enten oksygen, svovel eller metylen, R3 og R4 er like eller forskjellige og valgt fra hydrogen og lavere alkyl, n er 2 eller 3, A er valgt fra følgende pyrimidyl eller pyridyl-grupper :
hvor R5 er valgt fra hydrogen, lavere alkyl eller halogen, R6 og R7 er like eller forskjellige og valgt fra hydrogen, halogen, lavere alkyl, lavere alkoksy, hydroksy, cyano, nitro, trifluormetyl, CONR9R10 eller CO-B, R9 og R10 er like eller forskjellige og valgt fra hydrogen og lavere alkyl;
B er valgt fra
hvor m er 1, 2, 3 eller 4, R1X er valgt fra hydrogen eller lavere alkyl,
og de farmakologisk aktive salter derav.
Begrepet "lavere alkyl" er ment å inkludere rettkjedede og forgrenede, mettede hydrokarbongrupper med 1-5 karbonatomer, begrepet "cykloalkyl" er ment å inkludere cykliske hydrokarbon-grupper med 3-8 karbonatomer, begrepet "lavere alkoksy" er ment å inkludere rettkjedede eller forgrenede, mettede hydrokarbon-grupper med 1-5 karbonatomer, begrepet "halogen" er ment å inkludere fluor, klor og brom.
Det er foretrukket at Ra er hydrogen og R2 er halogen, hvorav fluor er foretrukket.
Med hensyn til R3 og R4 er hydrogen eller metyl foretrukket, spesielt hydrogen.
Med hensyn til R5 er hydrogen, alkyl eller halogen, spesielt fluor, foretrukket.
Med hensyn til R6 er hydrogen, alkyl, alkoksy, amid, nitro, karboksy, trifluormetyl, halogen, hydroksy eller cyano foretrukket^
Det er foretrukket at R7 er hydrogen, alkyl, alkoksy, nitro, karboksy, halogen, hydroksy, cyano eller an amid-gruppe.
Forbindelser hvor A er 2-substituert pyridyl er av spesiell interesse, spesielt de som har en alkoksy-, hydroksy-, alkyl-, amid-, cyano- eller hydrogensubstituent i 3-posi-sj onen.
Forbindelsene med formel (I) har basiske egenskaper og de kan derfor omdannes til sine terapeutisk aktive syre-addi-sjonssalter ved behandling med syre eller organiske syrer som eddiksyre, propansyre, glykolsyre, melkesyre, malon-syre, oksalsyre, ravsyre, fumarsyre, vinsyre, sitronsyre eller embonsyre.
I motsatt fall kan saltformen omdannes til den frie base-form ved behandling med et alkali.
Forbindelsene med formel (I) og deres farmakologisk akseptable salter har verdifulle farmakologiske egenskaper som gjør dem anvendelige ved behandling av mentale forstyrrelser som f.eks. psykoser, depresjoner og nervøsitet. Stress og nervøsitet hos dyr kan også behandles. Forbindelsene i foreliggende oppfinnelsen utviser psykotropiske egenskaper. De viser for eksempel affinitet for 5-HT2 og D2 bindingssteder i hjernen. I atferdsmodellforsøk utviser forbindelsene en limbisk virkningsprofil, dvs. de har kraftig virkning i forsøk med undersøkende atferd, dvs. trappe-testen.
Forbindelser med en kombinert 5-HT2/D2 affinitet, f.eks. clozapin, har anti-psykotisk effekt med en lav grad av ekstrapyramidale bivirkninger. Videre har forbindelsene med affinitet for 5-HT2-bindingsseter vist seg å påvirke de-pressive lidelser og også nervøse tilstander.
Effektive mengder av de tidligere nevnte farmakologisk aktive forbindelsene med formel (I) kan administreres til
mennesker eller dyr for terapeutiske formål i henhold til - vanlige administreringsmåter og i vanlige former, såsom oralt i løsninger, emulsjoner, suspensjoner, piller, ta-bletter og kapsler, i farmasøytisk akseptable bærere og parentalt i form av sterile løsninger. Ved parental administrering av den aktive substans må bæreren av eksipienset være en steril, parentalt akseptabel væske, f.eks. vann, eller en parentalt akseptabel olje f.eks. arachidinolje.
Selv om meget små mengder av de aktive materialene fremstilt i foreliggende oppfinnelse er effektiv når mindre terapi er involvert eller i tilfeller med administrering til individer med forholdsvis lav kroppsvekt, er enhets-dosene vanligvis fra 2 mg og oppover, fortrinnsvis 19, 25 eller 50 mg og til og med høyere, avhengig av forholdet som skal behandles, pasientens alder og vekt samt respons på medisineringen.
Enhetsdosen kan være fra 0.1 til 100 mg, fortrinnsvis fra 10 til 50 mg. Daglig dose bør fortrinnsvis være i området
10 til 200 mg. Den eksakte individuelle dose vil selvføl-gelig bli bestemt i henhold til vanlige medisinske prin-sipper under veiledning av lege eller veterinær. Forbindelsene med generell formel (I) kan fremstilles ved at (i) en forbindelse med den generelle formel (II)

hvor R5, R6 og R7 er som tidligere definert, og Z er en avspaltbar gruppe.
I det følgende illustreres oppfinnelsen ved noen eksempler. Disse forbindelser er betegnet med~kodenummer, a:b, hvor a betyr eksempelets nummer, hvori fremstillingen av forbindelsen det gjelder er beskrevet, og b refererer til de fremstilte forbindelsenes orden i henhold til det eksem-pelet. Forbindelse 1:2 betyr den andre forbindelsen fremstilt i henhold til eksempel 1.
Forbindelsenes struktur er bekreftet ved hjelp av NMR, massespektra og elementanalyse. Når smeltepunktene er angitt, er disse ukorrigert.
Eksempel 1
4- [4-(p-fluorfenyl)butyl]-l-(2-pyridyl)piperazinfumarat.
6.0 g (0.0323 mol) 4(p-fluorfenyl)butylklorid, 5.3 g (0.-0323 mol) pyridyl-piperazin, 5.2 g natriumkarbonat og 0.1 g jod ble oppvarmet sammen med 25 ml xylen til 150 °C (tempe-ratur på oljebad) i 20 timer.
Etter avkjøling av reaksjonsblandingen til ca. 100 °C, ble 50 ml toluen tilsatt og blandingen ble filtrert. 25 ml eter ble tilsatt til filtratet. Den organiske løsningen ble vasket tre ganger med 25 ml vann og til slutt en gang med 25 ml mettet natriumkloridløsning.
Fordamping av løsningsmidler ga et råprodukt som ble krys-tallisert fra cykloheksan. Smeltepunktet for den frie base var 57 - 58 °C.
Den frie basen ble oppløst i etanol/eter og fumaratet ble utfelt med overskudd av fumarsyre i etanol.
Omkrystallisasjon fra etanol ga 4.8 g av tittelforbindelsen (1:1), smp. 160 - 161 °C.
Eksempel 2
4- [4-(p-f luorfenyl )butyl] -1- [2-( 3-karbamyl-pyridyl)] piperazin dihydroklorid.
5.9 g (0.025 mol) l-[4-(p-fluorfenyl)butyl-piperazin, 3.9 g (0.025 mol) 2-klor-icotonsyre amid og 3.1 g natriumkarbonat ble refluksert sammen med 20 ml toluen i 20 timer.
Etter kjøling ble det erholdt en fast blanding som ble løst i etylacetat og vann. Toluen/etylacetat-fasen ble separert og vasket med vann og natriumklorid-løsning og deretter tørket med natriumsulfat.
Fordamping av løsningsmidlene ga en fri base som råprodukt, som ble omkrystallisert fra toluen. Den frie base hadde et smeltepunkt på 135 - 136 °C. 5 g av den frie base ble løst i etanol og dihydrokloridet ble utfelt med et overskudd av saltsyre i etanol. Omkrystallisasjon ga 3.0 g av tittelforbindelsen (2:1), smp.
210 - 213 °C.
Ved å bruke en lignende prosedyre ble følgende forbindelser fremstilt (isolert og renset ved flash-kromatografi som den rene base eller tilsvarende salter) fra de tilsvarende utgangsmaterialer: 2:2 4-[3-(p-Fluorfenoksy)propyl]-1-[6-klor-2-pyridyl]-piperazin hydroklorid, smp. 185 - 186 °C. 2:3 4-[3-(p-fluorfenoksy)propyl]-1-[2-pyrimidyl]piper-azinhydroklorid hemihydrat. smp. 208 - 210 °C. 2:4 4-[3-(p-fluorfenoksy)propyl]-1-[2-pyridyl]piperazin-dihydroklorid, smp. 233 - 235 °C. 2:5 4-[3-(p-fluorfenoksy)propyl]-1-[3-karbamyl-2-pyridyl]-piperazin dihydroklorid, smp.240 - 242 °C. 2:6 4-[4-(p-fluorfenyl)butyl]-1-[2-pyrimidyl]piperazin-hydroklorid, smp. 197 - 198 °C. 2:7 4-[4-(p-fluorfenyl)butyl]-1-[2-pyridyl]piperazin-fumarat, smp. 160 - 161 °C. 2:8 4-[3-(p-fluorfenoksy)propyl]-1-[3-nitro-2-pyridyl]-piperazin hydroklorid, smp. 182 - 182 °C. 2:9 4-[4-(p-fluorfenyl)butyl]-1-[6-klor-2-pyridyl]pipera-zinhydroklorid, smp. 150 - 151 °C. 2:10 4-[3-(p-fluorfenoksy)propyl]-1-[6-metoksy-2-pyridyl]-piperazinfumarat, smp. 185 - 186 °C. 2:11 4-[3-(p-fluorfenoksy)propyl]-1-[3-karbamyl-2-pyridyl]-[1,4-diazacykloheptan]oksalat, smp. 148 - 150 °C (Base smp. 140 - 141 °C). 2:12 4-[4-(p-fluorfenyl)butyl]-1-[3-etoksy-2-pyridyl]piper-azindihydroklorid, hemiisopropanolhemihydrat, smp. 168 - 169 °C.

1. Analogifremgangsmåte ved fremstilling av terapeutisk aktive forbindelser med den generelle formel (I)

5. Fremgangsmåte i henhold til krav 1, 2 eller 3 ved fremstilling av forbindelser hvor A er
karakterisert ved å anvende tilsvarende substituerte utgangsmaterialer.

Fremgangsmåte i henhold til krav 4 ved fremstilling av forbindelser hvor R6 er hydrogen, lavere alkyl, trifluormetyl, lavere alkoksy, CONRgR10 eller CO-B, R9 og R10 er som angitt i krav 1, hydroksy, nitro eller cyano og R7 er hydrogen, lavere alkyl, lavere alkoksy, hydroksy, nitro, halogen, cyano eller CONR9R10 eller CO-B, hvor B, R9 og R10 er som angitt i krav 1, karakterisert ved å anvende tilsvarende substituerte utgangsmaterialer.
7. Fremgangsmåte i henhold til krav 1-6 ved fremstilling av forbindelser hvor X er oksygen eller metylen, karakterisert ved å anvende tilsvarende substituerte utgangsmaterialer.
8. Fremgangsmåte i henhold til krav 5 ved fremstilling av forbindelser hvor R6 er hydrogen, lavere alkyl eller trifluormetyl, og R7 er lavere alkoksy, hydrogen, hydroksy, nitro, halogen, cyano eller CONRgR^ eller CO-B, hvor B, R9 og R10 er som angitt i krav 1, og R7 er plassert i 3-posisjon, karakterisert ved å anvende tilsvarende substituerte utgangsmaterialer.
9. Fremgangsmåte i henhold til krav 1-8 ved fremstilling av forbindelser hvor X er oksygen, karakterisert ved å anvende tilsvarende substituerte utgangsmaterialer.

Fremgangsmåte i henhold til krav 6 ved fremstilling av forbindelser hvor R6 er hydrogen og R7 er hydrogen, cyano, nitro, lavere alkoksy, lavere alkyl, hydroksy eller CONR9R10 eller CO-B, hvor B, R9 og R10 er som angitt i krav 1, karakterisert ved å anvende tilsvarende substituerte utgangsmaterialer.
11. Fremgangsmåte i henhold til krav 7 ved fremstilling av forbindelser hvor R7 er CONR9R10 eller CO-B, hvor B, R9 og R10 er som angitt i krav 1, hydroksy, metyl, cyano eller metoksy, karakterisert ved å anvende tilsvarende substituerte utgangsmaterialer.
