Abstract
Foreliggende oppfinnelse tilveiebringer en fremgangsmåte og materialer for å undersøke og isolere differensiert uttrykte gener. Ved fremgangsmåten ifølge foreliggende oppfinnelse blir forskjellige merkede populasjoner av DNAer fra kilder som skal sammenlignes kompeptivt hybridisert med referanse-DNA klonet på fast fasebærere, f.eks. mikropartikler, for å tilveiebringe et differensiert ekspresjonsbibliotek som de foretrukne utførelsesformer, kan bli manipulert ved hjelp av fluoresens-aktivert cellesortering (FACS). Undersøkelse av den relative signalintensiteten av forskjellige fluoresensmerker på mikropartiklene muliggjør kvantitativ analyse av ekspresjonsnivåer i forhold til referanse-DNA. Oppfinnelsen tilveiebringer også en fremgangsmåte for å identifisere å isolere sjeldne gener. Populasjoner av mikropartikler som har relative signalintensiteter av interesse kan bli isolert ved hjelp av FACS og de bundede DNAer identifisert ved sekvensering, slik som ved parallell signatursekvensiering (MPSS) i stort omfang, eller med vanlig DNA-sekvensfremgangsmåter.
