Abstract

Denne oppfinnelsen tilveiebringer en fremgangsmåte for fremstilling av nye polymorfe hemifumaratsalter av 8-klor-6,11-dihydro-11-(4-piperidyliden)-5H-benzo[5,6]cyklohepta[1,2-b]pyridin, heretter kalt "desloratadin". Disse polymorfe saltformene viser mye høyere løselighet i vann og også i protiske organiske løsningsmidler sammenlignet med mor-desloratadin. Fremgangsmåten for fremstilling av de polymorfe formene omfatter: (a) å blande den etanoliske løsningen av desloratadin og fumarsyre ved en temperatur på fra omtrent 55°C til 70°C, og å røre i 30 til 45 minutter etter blanding, og deretter filtrere det faste stoffet som derved er fremstilt under varme betingelser; for å gi den polymorfe formen 2 med en DSC på 232°C± 2°C; eller (b) å blande den etanoliske løsningen av desloratadin og fumarsyre ved en temperatur på fra omtrent 15°C til romtemperatur (25°C) og å røre ved denne temperaturen i 30 til 45 minutter, og deretter å filtrere ved romtemperatur; til å gi den polymorfe formen 1 med en DSC på 224°C ± 2°C.
