Abstract

Det beskrives sekvenser av B-avhengige dehydrataser med forbedret reaksjonskinetikk, og således redusert hastighet for enzymets dreperinaktivering i nærvær av glycerol og 1,3-propandiol.Enzymene skapes ved bruk av feiltilbøyelig PCR,. og en oligonukleotidrettet mutagenese for å sikte på DhaBl-genet, kodende en oc-subenhet av glyceroldehydratase.Mutanter med forbedret reaksjonskinetikk ble hurtig identifisert ved bruk av høygjenomløpsanalyser.
