Description
Oppfinnelsen vedrører ekstrahering og rensning av to typer antineoplastiske stoffer som er til stede i human urin og fremgangsmåte for ekstrahering og rensning herav. Spesielt vedrører oppfinnelsen to typer antineoplatiske fraksjoner et lavmolekylærvekt peptid og høymolekylærvekt protein som er ekstraherbar , fra human urin ved adsorbsjon på en syntetisk magnesiumsiliciumoksydgel og etterfølgende eluering derifra.
nærværet av antineoplastiske peptider i human urin ble omtalt av 3urzynski et al i en serie publikasjoner J<p>hysiol.Chmem. Phys., 5, 436 (1973); 6_, 457 ( 1974); 8_, 13 (1976); 8, 275 ( 1976) 3 . Disse peptider viste antineoolastisk .aktivitet
mot forskjellig humane tumorce1le 1injer og påvirket ikke veksten av normale celler. Disse urinpeptider ble frem-
stilt ved 80%etanolekstrahering av urin etterfulgt av en bistrømningselektroforese, papirkromatografi og ende prepar-ativ tynnsjiktkromatografi.
Florisil, et syntetisk magnesiums i 1ikat bestilt av Floridin Company i Pittsburgh, Pennsylvania, er blitt benyttet som adsorbent for biologisk aktive materialer i human urin som omtalt av Sloane i US-patent nr. 3.711.377,og 3.884.760.
Disse patenter omtaler adsorbsjon av et fibrinalyttisk protein-enzyra fra human urin med Florisil og etterfølgende eluering av dette protein enzym fra Florisil adsorbentet.
Generelt overensstemmende med oppfinnelsen føres human urin gjennom en adsorbentlagring av syntetisk magnesiumsilikat som ekstraherer to fraksjoner,.en.peptidfraksjon og en proteinfraksjon sammen med mange andre ikke identifiserte forbind-elser. Adsorbentlagringen vaskes deretter med kaldt destillert vann for å fjerne gjenværende urin. Kver av de ønskede antineoplastiske fraksjoner, peptidfraksjonen og proteinfraksjonen elueres deretter fra magnesiumsilikatet. Den lavmolekylærvekt peptidfraksjon elueres først med vandig aceton med svakt alkalisk pH. Deretter elueres høymolekyIvekt prote infraksjonen rr.ed en blanding av vann, aceton og glycerol ved' svakt alkalisk pil. Peptidf raks j onen konsentreres i vakuum for å fjerne aceton og den gjenværende vandige oppløsning av peptidet renses ytterligere ved membranfi 1trering. Etter dialyse for å fjerne aceton og glycerol renses proteinfraksjonen ved membranfiItrering og gelfiltrering.
Det er en hensikt med oppfinnelsen å tilveiebringe et materiale ekstraherbart fra human urin og som har tydelig antinetoplast-isk aktivitet. Det er også en hensikt å tilveiebringe en metode for å ekstrahere fra human urin et materiale med tydelig anti-ne top1as tisk aktivitet. Ytterligere hensikter og fordeler vil framgå av følgende beskrivelse.
Overensstemmende med oppfinnelsen adsorberes både en antineoplastisk peptidfraksjon og en antineoplastisk proteinfraksjon fra human urin ved et adsorbent som Florisil.. Hver av disse antineoplastiske fraksjoner elueres separat fra Florisil. Det antineoplastiske peptid elueres først den
vandig acetonoppløsning ved svakt alkalis!; pK, det antineoplastiske protein elueres deretter en aceton-glycerol-vann-blanding ved svakt alkalisk pH. Hver av de human urinanti-necplastiske fraksjoner inhiberer veksten av human tumor cellelinjer uten å innvirke på normale humane diploide celler. Videre inhiberer proteinfraksjonen veksten av visse humant tumorceller implantert i en naken mus.
De kjemiske og biologiske egenskaper av den ekstraherte og rensede antineoplastiske peptidfraksjon isolert fra human urin er følgende: XI) Molekylvekten av peptidet er omtrent 600 bestemt ved filtrering gjennom Amicon Diaflo membraner.
(2) Peptidet er varmestabilt ved 90°C i 10 minutter.
(3) Peptidets antineoplastiske aktivitet ødelegges omtrent totalt ved bwhandling av tørrfraksjonen med enten vannfri metanol eller absolutt etylalkohol. Etter hver av disse alkoholbehandlinger er den tørkede fraksjon i det vesentlige inaktiv som antineoplastisk middel.
(4) Den aktive peptidfraksjon viser en RF-verdi mellom 0,4
og 0,5 ved papirkromatografi i et n-butano1:eddiksyre: pyridin:vannsystem (30 : S : 20 :24). (5) Den aktive peptidfraksjon viser antineoplastisk aktivitet mot følgende humane tumorce1le linjer: bryst, melanoma, pancreas, blære og lunge. Disse antineoplastiske aktiviteter måles in vitro ved vevkulturteknikk.
De kjemiske og biologiske egenskaper på det ekstraherte og rensede antineoplastiske urin (ANU) protein er følgende: 1) Molekylvekten av AMU-proteinet er mellom 20.000 og 40.000 daltons. 2) AMU-proteinet inneholder karbohydrat, er varmestabil ved 90°C i 20 minutter, oppløselig i fenol mettet med vann og stabil overfor syre ved pH 2 ved 4°C i 48 timer. 3) AITU-proteinf raks j onen inhiberer veksten av følgende human tumor cellelinje: bryst, melanoma, blære, cervix og lunge. 4) AITU-prote inet er ikke-toksisk til humane dip loidce 1 le-linjer. 5) ANU-proteinet er et spesifikt antineoplastisk middel for humane tumorce1le 1injer da hverken mus-tumorce1le linjer eller normal museceller påvirkes av AI-TU-protein. 6) AiTU-protein undertrykker veksten og bevirker regresjon av humane tumorce1le linjer, f.eks. SEP-2, human lungetumor-
cellelinjer, implantert i naken mus.
7) AITU-proteinet inhiberer ikke human pancreas tumorcelle-linjer PAi:c/3 eller SV-40 overført WI-38 human lungeceller.
8) AMU-proteinet utøver ikke antiviral aktivitet.
I en utførelse av oppfinnelsen fremstilles Florsil i to mesh størrelser, 15-30 mesh og 30-60 mesh for adsorbsjon først ved flotteringsfjerning av det fine, deretter suspensjon i fortynnet askorbinsyre (pK 3-4) ved vare Ise temperatur i flere timer før bruk.
Uttømt human urin samles i en bulk og borsyrekrystaller og butylesteren av p-hydroksybenzosyre has i beholderen for å hindre bakteriell vekst. Ca. 1 liter av den samlede urin ved værelsetemperatur helles gjennom en stabel av to jersey tekstilbagger inneholdende Florisil. Den øvre bag inneholder ca. 0,2 kilo av 15 - 30 mesh Florisil og den nedre bag inneholder ca. 0,2 kilo 30 - 60 mesh Florisil. 3ruk av to bagger med forskjellig mesh Florisil hindrer klumping. Hår uttømt human urin samles in situ ved en urinal og Florisil granuler holdes på et trau som angitt i US-patent nr. 3.711. 377, plasseres en jersey tekstilbag av pellets av borsyre med p-hydroksybenzosyrebutylester på toppen av de to Florisil-holaige bagger.
Etter at urinen har fullstendig berørt Florisilet for å mulig-gjøre adsorbsjon av de ønskede materialer, utvaskes Florisilet med ca. 3 liter kaldt destillert vann (ca. 4°C). All etter-følgende eluering og rensningsprosesser utformes ved ca. 2-4°C.
Antineoplastisk peptidfraksjonen elueres med ca. 1500 ml
kald vandig aceton (15%); ved pH 9-9,5. Denne fraksjon justeres deretter til pK 7-7,5 med 2 normal kald HC1 og konsentreres ved
å fjerne aceton i vakuum. Konsentratet renses ved å føres gjennom en "Amicon Diaflo UM2" membran som holder tilbake materialet som har molekylvekt større enn ca. 1000, deretter gjennon en "Amicon Diaflo UM05" membran som holder tilbake materiale som har molekylvekt større enn ca. 600. Denne filtrede fraksjon er ytterligere renset ved standard papirkromatografi teknikk.
Etter at peptidfraksjonen er blitt først eluert, elueres antineoplastikk fraksjonen fra Florisilet ved vandig aceton (15%) inneholdende 30% glycerol ved pH 9-9,5. Prote ineluatet justeres til p'I 7-7,5 med 2 normal KCL og konsentreres deretter ved filtrering gjennom en "Amicon Diaflo" membran UM20 som holder tilbake materialet som har molekylvekter over ca. 20.000. Det ønskede antineoplastikk protein passerer ikke gjennom membranen. Proteinoppløsningen dialyseres deretter mot destillert vann og lyofiliseres. Det lyofiliserte protein renses ytterligere ved "Sephacryl" S-200 kromatografi ved nøytral pl-:, f os f atpuf f ret ved 0,1 molar.
Det rensede antineoplastiske protein utbytte er omtrent 300 yg/liter urin, ved en konsentrasjon omtrent 500 ganger urinens konsentrasjon. Det rensede ANO-prote in utøver anti-neoplastisk aktivitet ved konsentrasjoner på mindre enn lyg protein/
ml medium.
Nakne museksperimenter ble utført- på følgende måte:
4 til 6 uker gamle i!IH athymic (nu/nu) mus ble transplantert subkutant med human carcinoma cellelinje KEP 2 (10 7 celle/ mis). Fire dager etter transplantasjonen ble separate grupper av mus behandlet subkutant en gang hver 3 dag med 1 yg av ren eller rå AiTU-protein i medium. Kontrollgrupper mottok 0,1 ml av medium inneholdende 10%kalve fosterserum. Tumorvekst ble fulgt i 3 - 4 uker. Ved slutten av denne periode ble tumorvolum målt og det ble underkastet histologi. Et minimum på 8 mus ble benyttet i hver gruppe. Ekstra kontrollgrupper mottok ASU-prote in alene i mediet uten tumor-transplant. Behandling med rått AKU-protein resulterte i en tydelig reduksjon i tumorvolum (4.1 cm 3 til 1,8 cm 3gjennom-snittlig) . Behandling med halvrent ANU-protein resulterte i total regresjon av tumor i fire av åtte tilfeller og tydelig reduksjon i tumorstørre Ise i de gjenværende dyr.
In vitro forsøk utført på følgende måte:
Omtrent IO<5>celler av hver cellelinje som skulle tuderes ble inokkulert med 5 ml av mediet i cellekultur f lasker ("Falcon" 3013 flasker, 25 cm 2 areal), prote infraksjonene ble oppløst i fosfatpuffret saltoppløsning og satt til cellen i mediet. Kulturene ble inkubert ved 37° med 5%C02i 96-144 timer, kontrollflaskene inneholdt ekstra puffer. Ved slutten av forsøksperioden ble cellene talt etter fjerning fra flasken av EDTA-trypsinoppløsning (Gibco).
AMU-proteinet inhiberte veksten av human bryst, melanoma, blære, cervix og lungetumorcelie linjer.
Det kan selvsagt foretas modifikasjoner uten å overskride oppfinnelsens ramme slik den fremgår av kravene.
Claims (1)
1. Fremgangsmåte for ekstrahering av en glykoprote infraksjon innbefattende et glykoprotein som har en molekylvekt mellom ca. 20.000 dalton og ca. 40.000 dalton, idet proteinet utøver, antineoplastisk aktivitet fra human urin inneholdende glykoproteinfraksjonen, karakterisert ved at et magnesiumsilikatadsorbent bringes i berøring med human urin, ikke adsorbert urin vaskes fra adsorbentet og glyko--proteinfraksjonen ble eluert fra adsorbentet ved en blanding av vandig aceton og glycerol.
2. Fremgangsmåte ifølge krav 1, karakterisrt ved at den ikke adsorberte urin vaskes fra adsorbentet med vann.
3. Fremgangsmåte ifølge krav 1, karakterisert ved at en antineoplastisk peptidfraksjon elueres fra adsorbentet før eluering av glykoprote infraksjonen.
4. Fremgangsmåte ifølge krav 3, karakterisert ved at peptidfraksjonen elueres med vandig aceton.
5. Fremgangsmåte ifølge krav 3, karakterisert ved at peptidfraksjonen elueres mee ca. 15% vandig aceton ved en pK mellom ca. 8,5 og ca. 9,5.
6. Fremgangsmåte ifølge krav 1, karakterisert ved at glykoproteinfraksjonen elueres med ca. 15% vandig aceton inneholdende ca. 30% glycerol ved en pil mellom ca. 8,5 og ca. 9,5.
-7. -Fremgangsmåt e ifølge krav 1, karakterisert ved at adsorbentet omfatter granuler med en størrelse mellom ca. 15 mesh og ca. 60 mesh.
8. Fremgangsmåte ifølge krav 1, karakterisert ved at den eluerte glykoprote infraksjon renses og konsentreres ved hjelp av memforan fi 1trer ing.
9. Fremgangsmåte ifølge krav 8, kara k;.t.e:.r i s e r t ved ytterligere dialysering av nevnte fraksjon mot destillert vann.
10. Fremgangsmåte ifølge krav 9, karakterisert ved ytterligere lyofi1iser ing av glykoproteinfraksjonen.
11. Fremgangsmåte ifølge krav 1, karakterisert ved at silicagel bringes i berøring med urinen ved ca. vsre Ise temperatur.
12. Fremgangsmåte ifølge krav 1, karakterisert ved at glykoproteinfraksjonen elueres ved en temperatur på mellom ca. 2°C og 4°C.
13. Glykoprotsin som har en molekylvekt på mellom ca. 20.000 dalton og ca. 40.000 dalton ekstrahert fra human urin ved å bringe magnesiumsi 1ikatadsorbent i kontakt med human urin, utvasking av ikke-adsorbert urin fra adsorbentet og eluering av glykoproteinet fra adsorbentet med en blanding av vandig aceton og glycerol, idet glykoproteinet utøver antineoplastisk aktivitet på humane tumorce1le 1 injer.
