Abstract

Fremgangsmåte for fremstilling av en vaksine mot dental karies som er kjennetegnet ved trinnet for dyrkning av en variant som oppnås ved å inte- grere et proteinantigen (PAc)-utrykk-, ende gen inn i det kromosomale gen til en Streptococcus mutans GS-5-stamme for å oppnå proteinantigenet, idet proteinantigenet fremstilles på celle- overflaten til oral Streptococcus mutans eller det fremstilles extra- cellulært av mikroorganismen og har en molekylvekt som varierer fra omkring 170.000 til 220.000. Streptococcus mutans GS-5 (K-3) hvori et protein- antigen-uttrykkende gen er integrert i det kromosomale gen, har evnen til å fremstille proteinantigenet på over- flaten av cellene eller extracellulært. En forebyggende vaksineblanding mot dental karies i form av nesedråper omfatter proteinantigenet som således er fremstilt av stammen: Streptococcus mutans, idet vaksinen administreres intranasalt. Fremgangsmåten gjør det mulig å øke utbyttet av PAc og å for- enkle fremgangsmåtene for rensing av PAc. Vaksineblandingen gjør det mulig å absorbere proteinantigenet, PAc, lett internt, med høy effektivitet, og gjør det også mulig å effektivt øke antistofftiteren observert etter administreringen av denne.
