Abstract
Antistoffer eller protein av- ledet fra en gitt art kan fremstilles i et utvalgt diskordant beslektet verts- dyr ved dyrkning av en hybridom eller en annen udødelig proteinproduserende cellelinje i vertsdyrets bukhule, der- som både vertsdyrets humorale immun- respons og den cellemedierte immun- respons kompromitteres. Den humorale immunrespons kan kompromitteres kongenitalt; slik at f.eks. C3- manglende dyr kan anvendes. Den celle- medierte immunrespons kompromitteres fortrinnsvis ved administrering av syklosporin A. Dette muliggjør frem- stilling av humanavledete antistoffer in vivo, og tillater generelt oppgra- dering av in vivo-fremstillingsmetoder ved anvendelse av større dyr enn de for tiden vanligvis anvendte mus.
