Description

Przedmiotem wynalazku jest sposób wytwarzania trwalego preparatu karotenoidowego z surowców naturalnych: pomidorów, marchwi,jarzebiny a zwlaszcza z odpadów powstajacych przy przerobie dyni.Barwniki karotenoidowe sa naturalnymi barwnikami stosowanymi do barwienia produktów spozywczych, farmaceutycznych i kosmetycznych.Znane sa sposoby otrzymywania barwników karotenoidowych w postaci ekstraktów lub preparatów z roslin~(opis patentowy PRL nr 89 816), zwierzat (opis patentowy PRL nr 113 328) oraz z mikroorganizmów. Preparaty karotenoidowe dostepne sa w postaci roztworów olejowych, wodnych lub jako suche proszki (opis patentowy St. Zjedn. Am. nr 3 110598).Znane metody otrzymywania barwników karotenoidowych polegaja na ich ekstrakcji rozpu¬ szczalnikami organicznymi niepolarnymi: eterem naftowym, benzyna, chloroformem, benzenem, dwusiarczkiem wegla lub nie mieszajacymi sie ze soba rozpuszczalnikami takimi jak alkohol i eter naftowy. Surowiec zmydla sie w roztworach lekko alkalicznych, a nastepnie odpedza sie rozpu¬ szczalnik i rozpuszcza karotenoidy w oleju roslinnym lub krystalizuje.Znane sa równiez metody polegajace na bezposredniej ekstrakcji karotenu olejem roslinnym ze zmydlonego surowca.Wada podanych wyzej metod jest wytwarzanie barwników z dosc drogich surowców oraz stosowanie olejowego roztworu karotenu. Taki preparat jest nietrwaly wskutek zachodzacej hydrolizy oleju (jelczenia), szczególnie szybko przebiegajacej w obecnosci tlenu atmosferycznego i swiatla. W wyniku procesu oksydacji karotenoidów nastepuje utrata charakteru prowitaminy A.Uzycie oleju roslinnego do ekstrakcji sprzyja takze równoczesnemu przejsciu zanieczyszczen do preparatu. Stosowanie toksycznych rozpuszczalników (CS2, eter naftowy, chloroform) powoduje, ze musza byc one dokladnie usuniete z produktu. Wytwarzanie preparatów proszkowych naraza karotenoidy na ich utlenienie w czasie procesu emulgowania i suszenia.Sposobem wedlug wynalazku prowadzi sie ekstrakcje materialu roslinnego acetonem w czasie 10-24 godzin, przy czym uzywa sie na 1 czesc substratu 1 -5-5 czesci acetonu.Nastepnie dodaje sie eteru etylowego w ilosci od 1 do 2 czesci objetosciowych na 1 objetosc acetonu. Calosc przemywa sie woda, po czym oddziela sie faze wodno-acetonowa od fazy eterowej2 133 342 z barwnikiem. W czasie tej operacji karotenoidy przechodza z acetonu do eteru, natomiast aceton przechodzi do roztworu wodnego. Dzieki temu nastepuje oddzielenie wiekszosci zanieczyszczen przechodzacych wraz z barwnikiem do acetonu w czasie procesu ekstrakcji. Eter etylowy z barwnikiem dozuje sie do rozpuszczonego tluszczu w temperaturze wyzszej do temperatury wrze¬ nia eteru, korzystnie okolo 50°C. Nastepnie ciekly preparat wlewa sie do formy, w której zachodzi jego krzepniecie. Eter etylowy i aceton odzyskuje sie poprzez skroplenie par lub destylacje i skroplenie oraz zawraca sie z powrotem do procesu ekstrakcji. Jako tluszcz, do którego wprowadza sie eter z barwnikiem uzywa sie tluszcz cukierniczy.W wyniku stosowania sposobu wg wynalazku otrzymuje sie preparat karotenoidowy o stosunkowo duzej trwalosci. Zmiany pod wplywem swiatla i tlenu atmosferycznego sa w takim preparacie wielokrotnie wolniejsze niz w oleju roslinnym. Uzywa sie nietoksycznego eteru etylo¬ wego, który calkowicie jest usuwany w czasie jego dozowania do tluszczu z barwnikiem przy produkcji np. ciasta makaronowego, margaryny itp. Odparowanie eteru etylowego przy wprowadzaniu tluszczu odbywalo sie w tempera¬ turze 50°C, która to temperatura utrzymywana w trakcie zabiegu powodowala jednoczesne odpa¬ rowanie eteru. Carotenoid dyes are natural dyes used for dyeing food, pharmaceutical and cosmetic products. There are known methods of obtaining carotenoid dyes in the form of extracts. or preparations from plants (PRL Patent No. 89,816), animals (PRL Patent No. 113,328) and from microorganisms. Carotenoid preparations are available in the form of oily, aqueous solutions or as dry powders (US Patent No. 3,110,598). The known methods of obtaining carotenoid dyes consist in their extraction with non-polar organic solvents: petroleum ether, gasoline, chloroform, benzene, carbon disulfide or incompatible solvents such as alcohol and petroleum ether. The raw material is saponified in slightly alkaline solutions, and then the solvent is stripped off and the carotenoids are dissolved in vegetable oil or crystallized. There are also methods involving the direct extraction of carotene with vegetable oil from the saponified raw material. The disadvantage of the above methods is the production of dyes from quite expensive raw materials and the use of an oil solution of carotene. Such a preparation is unstable due to the hydrolysis of (deer) oil, which proceeds particularly rapidly in the presence of atmospheric oxygen and light. As a result of the carotenoid oxidation process, the nature of provitamin A is lost. The use of vegetable oil for extraction also favors the simultaneous transfer of contaminants to the preparation. The use of toxic solvents (CS2, petroleum ether, chloroform) causes that they must be carefully removed from the product. The production of powdered preparations exposes the carotenoids to their oxidation during the emulsification and drying process. The method according to the invention extracts the plant material with acetone for 10-24 hours, using 1-5-5 parts of acetone for 1 part of the substrate. 1 to 2 parts by volume of ethyl ether to 1 volume of acetone. The mixture is washed with water, and the water-acetone phase is separated from the dye-ether phase. During this operation, carotenoids are transferred from acetone to ether, while acetone is transferred to aqueous solution. Thanks to this, most of the impurities passing along with the dye to acetone are separated during the extraction process. The dyed ethyl ether is dosed into the dissolved fat at a temperature higher than the boiling point of the ether, preferably around 50 ° C. The liquid preparation is then poured into a mold where it solidifies. The ethyl ether and acetone are recovered by vapor condensation or by distillation and condensation and recycled back to the extraction process. A confectionery fat is used as the fat to which the dye ether is introduced. The method according to the invention gives a carotenoid preparation with a relatively long stability. Changes under the influence of light and atmospheric oxygen are many times slower in such a preparation than in vegetable oil. The non-toxic diethyl ether is used, which is completely removed during its dosing into the fat with the dye in the production of e.g. pasta, margarine, etc. The obtained preparation is characterized by high purity due to the application of multistage extraction. 100 g of dried pumpkin waste was ground in a grinder and then extracted with acetone in a Soxklet apparatus in a 1: 2 volume ratio of the material and acetone for 12 hours, without light. The extract was vacuum-filtered and diluted 1: 1 by volume with ethyl ether. Distilled water was washed to remove acetone and water-soluble compounds. An etheric extract of carotenoids, of which the carotenoid content was 15%, was added dropwise with vigorous stirring to the melted confectionery fat (melting point approx. 38 ° C), simultaneously receiving the ether condensation in the receiver. Evaporation of the diethyl ether during the introduction of the fat took place at a temperature of 50 ° C., which temperature was maintained during the treatment and caused the ether to evaporate simultaneously. A dark orange liquid preparation, free of ether, was poured into a mold in which it solidified. 1 g of the solid preparation contained 144-720 mg of carotene. With respect to the carotenoids in the raw material, the process efficiency was 88%.
