Description

Graf. Radom — 1539/86 80 egz. A4 Cena 100 zl PL PL PL The subject of the invention is a method of extracting oil seeds to obtain edible protein flours of appropriate microbiological purity. The method applies to oil seeds with a high oil content, such as sunflower, peanut and cotton seeds, as well as to seeds with a low oil content, such as soybean, grape, sesame, carthamum and other seeds. The invention relates in particular to a method for producing sunflower meal for human consumption from ordinary or dehulled sunflower seeds. Oilseed extraction is generally carried out to obtain edible oil and valuable high-protein feed. Production of ingredients and semi-finished products for food The management of finished food products requires maintaining their nutritional properties during the processing and processing of raw materials, as well as compliance with hygienic and sanitary regulations for products in order to protect the consumer's health. Food quality is a function of many different factors, such as chemical composition and quality of raw materials, production methods and materials used in installations, hygienic and sanitary environmental conditions. production and the method of preserving the finished food product (Nickerson J. T. and Sinskey A, J., Microbiology of 25 Food and Food Processing American Elsevier Publishing Co., New York, 1972), Nowadays, sanitization operations are expected in installations adapted to the production of foodstuffs or sterilization in the final stages or even in each of the intermediate stages. The above-mentioned treatment is particularly necessary in the case of protein products obtained from plant seeds, in which the initial number of bacteria is many times higher than the permissible standards established by various legal provisions. To meet such requirements, in industrial practice there is a need to carry out sterilization, which is sometimes performed under such drastic conditions that it causes structural changes or deterioration of the biological nutritional properties of the final product (Tateo F ., Detergenza e sanificazione nell' Industria Alimenatre, AEB Brescia 1977, Italy), The invention concerns the industrial processing of ordinary or dehulled oilseeds in order to produce protein meals with microbiological characteristics compliant with the requirements for human food. According to The invention's method of extracting oil seeds, especially sunflower seeds, using a mixture of solvents, leading to the preparation of food protein flours with an acceptable microbiological purity, consists in using a mixture of solvents consisting of a hydrocarbon solvent and an agent - a nizing agent capable of being homogeneously miscible with a hydrocarbon solvent, such as an organic compound having an alkyl chain of 1-5 carbon atoms and at least one polar group of an alcoholic nature, and the extraction is carried out in this ¬ perature 20 and above, but not exceeding 60°C at a solid-liquid ratio of 1:1-1:40. The solvent to be used is therefore a mixture consisting of a hydrocarbon compound selected selectively from petrochemical products, which is usually hexane, and an organic compound with an alkyl chain containing 1-5 carbon atoms and at least bottom of the polar group of an alcoholic nature, such as, for example, ethanol, which is completely miscible with the previously selected solvent. The use of, for example, ethanol is justified by its sanitizing properties, which are explained by the disruption of cell walls due to the dehydration effect. effects of alcohol (Verona O. and Picci G., Microbiologia degli Alimenti, 1966, UTET). Extraction is carried out using a mixture consisting of a hydrocarbon solvent, hexane, and an organic polar solvent, ethanol, the percentage of which is 1-50% by volume, preferably 1-5% by volume. Extractions solid - the liquid is preferably carried out in multiple stages, each stage lasting from half an hour to 4 hours. The solid-liquid weight-volume ratio is from 1:1 to 1:40, and the operating temperature is from 20-60°C, preferably 20-25°. The solid phase is then dried for 1-60 hours. In the case of sunflower seed extraction, preferred conditions include a temperature in the range of 20-22°C, an ethanol concentration in hexane of 3-5% and a solid-liquid ratio in the range 1—5 and 1:20. The examples relate to a process for producing edible protein meals from sunflower seeds using a mixture of solvents. These examples cannot, however, be interpreted as limiting the invention, which is applicable to all oilseeds. In the examples below, ethanol and n-hexane from Carlo Erba, Milan, Italy, were used as the RPE (Erba Pure Reagent) solvent. The culture media used for microbiological analyzes were Plate Count Agar, Mycological Agar, E. E. Mossel Broth, Tryptic Soy Broth, Violet Red Bilem Bacto Brillant Green Bile Broth, and McConkey Agar, supplied by DIFCO, and Clostiridum perfrigens Selective Agar, supplied by Merck. Determinations of chemical components, moisture, lipids, ash, proteins and crude fiber were performed according to the methods of A. O. A. C. (Association Official Analytical Chemdste, 11th ed., 1970). 10 13 20 30 35 40 45 50 55 60 The content of phenols was measured using a gas chromatograph using the direct silylation method. Soluble sugars were determined using the colorimetric method of M. Dubois et al. (Anal. Chem. 28, no. 3), 350 (1056). Microbiological analyzes were performed according to the procedures described by D. A. A. Mossel, J. Brechet and R. Lambion in "La prevention des infections et des toxi-infections alimentaires", CEPIA, Brussels 1/962. Example I. This example concerns the preparation of deoiled flour of appropriate microbiological purity by extraction of oil from dehulled sunflower seeds using n-hexane containing ethanol in various volume concentrations ranging from 1 to 5%, with a weight-volume ratio. solid-liquid ratio of 1:20 and at a temperature of 22°C. The drying time was 48 hours at ambient temperature. Table 1 Chemical composition of dehulled sunflower seeds, expressed as dry matter, in percent. Table 2 shows the results of microbiological analyzes for various ethanol concentrations. These results show different behavior of microorganisms towards the same sanitizing agent: the latter is very active against vegetative bacterial strains, yeasts and molds, while spore-forming strains are highly resistant. Before being analyzed, the flour was dried at ambient temperature for 48 hours. Example II. This example concerns the preparation of deoiled flour of appropriate microbiological purity by extracting oil from dehulled sunflower seeds with n-hexane containing 5% ethanol by volume, with a weight-volume solid-liquid ratio of 1:20, in temperature 22°C. The flour was dried at ambient temperature for 12 hours or 48 hours. Dehulled sunflower seeds whose degree of contamination was known and which had the composition HI? 514 chemical of the type given in Table 1, was stripped with a mixture of n-hexane and ethanol (5% by volume of the latter) at different contact times after plating and after two stages of batch extraction with n-hexane lasting 1 hour each. The temperature was 22°C, the weight-volume solid-liquid ratio was 1:20. Drying times at ambient temperature ranged from 12 to 48 hours. The results presented in Table 3 show that the sanitizing effect of 5% ethanol is significant already after the first 30 minutes of contact. Drying flour at ambient temperature with different contact times turned out to have no effect on the bacterial content in the tested products. Example III. This example concerns the preparation of deoiled flour of appropriate microbiological purity by extracting oil from dehulled sunflower seeds with n-hexane containing 5% ethanol by volume, at a solid-liquid weight-volume ratio of 1:5 and at a temperature of 22 °C. The chemical composition was as given in Table 1. Oil extraction from dehulled seeds was carried out in two solid-liquid extraction batches in a ratio of 1:20 with n-hexane, after. followed by a third extraction step using 5% ethanol in n-hexane at a solid-liquid weight/volume ratio of 1:5 and at various contact times at a temperature of 22°C. The flour thus obtained was allowed to dry under cover at ambient temperature for 48 hours. The results summarized in Table 4 show the sanitization; ational effect of the ethanol-hexane mixture. also for solid-liquid weight-volume ratios of 1:20 and below, but the effect of this action is slightly weaker compared to the data in Table 3 for Example II. ; *_ Example IV. This example concerns the preparation of deoiled flour of appropriate microbiological purity from dehulled sunflower seeds by extracting the oil with n-hexane, containing 2 or 3% ethanol, respectively, at solid-liquid volume ratio of 1:20 and at a temperature of 22°C. The chemical composition of dehulled sunflower seeds was as given in Table 1. Oil extraction from dehulled seeds was carried out in two extraction batches with n-hexane 1: 20 (solid:liquid); followed by a third extraction step using 2% ethanol in n-hexane or 3% vol. ethanol in n-hexane at a solid-liquid weight/volume ratio of 1:20 and at a temperature of 22°C. The flour obtained in this way was dried at ambient temperature for 48 hours. The results presented in Table 5 show that the sanitizing effect of the ethanol-II-hexane mixture is significant at a concentration of 3% ethanol in n-hexane at a contact time of 3 hours. Example V. This example concerns the preparation of deoiled flour of appropriate microbiological purity by extracting oil from dehulled sunflower seeds with n-hexane containing 3% by volume. ethanol, at a solid-liquid weight/volume ratio of 1:20, at contact times of 1, 2 or 3 hours and at a temperature of 6°C. The chemical composition of the dehulled seeds was as given in Table 1. Extraction of the oil from defoliated seeds were carried out in two extraction batches with n-hexane 1:20 (solid: liquid), followed by a third batch and extraction using 3% vol. ethanol in n-hexane at a solid-liquid weight/volume ratio of 1:20, at a temperature of 60°C and at contact times of 1, 2 or 3 hours. The flour thus obtained was dried at room temperature for 48 hours. As can be observed from the data used in Table 6, the use of a higher temperature, i.e. 60°C, did not improve the sanitizing effect of the ctanol-n-hexane mixture.
