Description

Do realizacji sposobu wedlug wynalazku stosuje sie uklad sluzacy do automatycznego pomiaru i regulacji, którego elementami sa: zegar czasu rzeczywistego, miernik ilosci pozywki doprowadzonej do fermentora, miernik zawartosci tlenu w gazach odlotowych, komputer i ele¬ ment wykonawczy np. pompa.Sposób wedlug wynalazku umozliwia intensyfikacje produkcji biomasy drobnoustrojów oraz pozwala uzyskac wysoka wydajnosc i produktywnosc hodowli.Przyklad. Pozywke dozowano dwoma pompami typu 335 prod. Unipan. Jedna pompka pracujaca niezaleznie od ukladu sterujacego dozowala nieprzerwanie pozywke do bioreaktora w ilosci 0,3 cm3/min. Praca drugiej pompki sterowal natomiast minikomputer zgodnie z wlasciwym programem sterujacym. Pracownia Poligraficzna UP PRL. There is a method of cultivating microorganisms which consists in controlling the supply of nutrient solution depending on the rate of oxygen consumption by microorganisms, e.g. from Ohashi's publication in J. Chem. Soc. Japan, 1958, vol. 61. The principle of this cultivation method is to supply the bioreactor with nutrient proportional to the rate of oxygen consumed by the microorganisms. This method does not prevent the introduction of excess nutrient into the fermentor, which leads to a reduction in the efficiency of the process and the formation of ethanol. The method according to the invention consists in the fact that the nutrient inflow rate increases or decreases when the ratio of the rate of oxygen uptake by microorganisms increases or decreases. the rate of consumption of the nutrient medium or the source of carbon contained in it or the feed of nutrient is regulated in proportion to the value of the quotient of the rate of consumption of the nutrient or the source of carbon contained in it by microorganisms to the rate of oxygen uptake by them. To implement the method according to the invention, a system is used for automatic measurement and regulation, the elements of which are: a real-time clock, a meter of the amount of nutrients supplied to the fermentor, a meter for the oxygen content in exhaust gases, a computer and an actuator, e.g. a pump. The method according to the invention enables the intensification of the production of microbial biomass and leap high yield and productivity of breeding. Example. The cultivation was started with the volume of the substrate equal to 2 dm / which was inoculated with 23.25 g of yeast in terms of dry mass. The control parameter was to maximize the expression d02 / dC. The patch was dispensed with two pumps, type 335 prod. Unipan. One pump, working independently of the control system, continuously dosed the nutrient to the bioreactor in the amount of 0.3 cm3 / min. The work of the second pump was controlled by a minicomputer in accordance with the appropriate control program. It provided for starting or stopping the pump depending on the value of the expression d02 / dC / the ratio of the rate of oxygen uptake by microorganisms to the rate of their nutrient consumption /. The increase in the value of dVdC was accompanied by the start of the pump, and the decrease in the value of the quotient d02 / dC was accompanied by the shutdown of the pump. This pump, like the first one, dosed food in the amount of 0.3 cm3 / min. In the 2-hour cultivation, the bioreactor-2 139 537 trough 60.2 cm3 of nutrient was supplied to the bioreact, which, with the efficiency coefficient equal to Y = 0.66 g / g, ensured productivity at the level of rx = 1.69 g / dm3 h and the specific growth rate fx = 0.131 h "1.Patent claim A method of cultivating microorganisms, especially baker's yeast, in which the nutrient is introduced into the fermentor depending on the rate of oxygen consumption by the microorganisms, characterized by the fact that the nutrient feed rate increases or decreases at the time of growth or decrease. the value of the quotient of the rate of oxygen uptake by microorganisms to the rate of consumption of nutrient medium or the carbon source contained in it or the feed of nutrient medium is regulated proportionally to the value of the quotient of the rate of consumption of the medium or the carbon source contained in it to the rate of oxygen uptake by them. 100 copies Price PLN 100 PL
