Description

Okres dzialania preparatu moze byc wydluzony róznymi metodami jak: — chemiczna modyfikacja substancji czynnej — tworzenie kompleksów — dodatek specjalnych srodków pomocniczych hamujacych farmaceutyczne dzialanie i/albo — odpowiednie postepowanie farmaceutyczne. Potwierdza to badanie radiologiczne i badanie krwi. Preparaty oddaja calosc substancji czynnej i zapewniaja staly terapeutycznie obraz krwi. Opóznione uwalnianie substancji czynnej prowadzi w terapii do oszczednego stosowania srodków technicznych. The subject of the invention is a method for producing frame tablets made of a porous inert polymer carrier with a prolonged action in terms of releasing active substances in the form of water-soluble inorganic and organic salts, such as those containing potassium, zinc, iron, magnesium and codeine. In medical practice, preparations are used pharmaceuticals with uniform and delayed secretion of active substances, and the therapeutic effect of the blood can be ensured for a longer period by the first unit dose. The period of action of the preparation can be extended by various methods, such as: - chemical modification of the active substance - formation of complexes - addition of special auxiliaries inhibiting pharmaceuticals action and/or - appropriate pharmaceutical procedure. From the Swedish patent description No. 808 014, a method is known for producing preparations with delayed release of the active substance, in which the active substance is pressed before or after wet granulation with a neutral plastic powder such as polyvinyl chloride, polyethylene or polyvinyl acetate.Direct pressing only produces a looser skeleton from which the active substance is released in a shorter time.Delayed release is only achieved with a low concentration of active substance and a high concentration of the skeleton substance.Wet granulation leads to a more compact structure and therefore requires less amount of skeletal substance. The release of the active substance is initially too fast and, in the end, not always complete and depends on wetting and pressing pressure. According to United States Patent No. 3,065,143, hydrophilically formed raw materials such as sodium alginate, alginic acid or sodium carboxymethyl cellulose are used as framework substances. , from which the active substance is released partly by diffusion and partly by slow dissolution of the hydrophilic framework substance. The rate of release of the active substance incorporated into the hydrophilic matrix depends on the properties of the framework substance, the forming method, pressing pressure and the conditions in the gastrointestinal tract. The release of the active substance can be delayed by coating with a poorly soluble or permeable polymer. By using polymers with pH-dependent solubility, it is possible to achieve pH-dependent secretion of the active substance in the stomach or intestines. In this case, the release rate of the active substance depends on the travel time. A small amount of active substances has a crystalline structure that can be easily coated. For this purpose, an appropriate grain size must first be created, which increases costs. US patents Nos. 3,035,985, 3,037,911 and 3,087,860 describe obtaining a delay in the release of active substances from wax matrices. The active substances are dispersed in molten waxes. and/or in other lipid-like substances and granulate after cooling. The release rate depends on the mass of the dispersed substance, the grain size of the active substance and the type of wax and/or lipid-like substance. For therapeutic purposes, only preparations in which the direction of action of the active substance remains and unchanged can be used, a therapeutic picture of the blood is achieved in a short time, the secretion of the active substance over a long period of time is uniform and in which, even with long-term application, none of the active substances shows any side effects. Regulating the release of the active substance in known solutions is very difficult, the quality and repeatability of the preparations is not always satisfactory, or the production costs are too high. It was found that that for the preparation of such preparations, water-insoluble, thermoplastic and practically non-toxic polyvinyl butyral can be successfully used, producing so-called filled tablets which do not disintegrate in water and which contain an internal polymer only in such an amount as to ensure appropriate strength. The release of the active substance is regulated by reducing the porosity of the tablet by heat treatment and, optionally, by coating with a permeable coating. Polyvinyl butyral (Movital B 30T), polyvinyl alcohol butyroacetal, is a plastic, film-forming polymer. It is highly soluble in alcohols, esters, ketones and chlorinated hydrocarbons, however, insoluble in water and digestive juices, it does not decompose in the body, is not adsorbed and is not toxic. Its permeability is relatively low and can be regulated by the addition of water-soluble substances. When using polyvinyl butyral, the release of the entire active substance content at the desired rate can be achieved by an appropriately formed skeleton or by a skeleton and a permeable outer coating, under the conditions release properties determine the properties of an appropriately formed skeleton or shell and coating. The method according to the invention consists in the active substance and auxiliary agents being evenly moistened with a solution of polyvinyl butyral in a suitable solvent containing polyvinyl butyral, taking into account the average weight of the tablet in the amount of 2 - 8%, granulated using a known method and then, after homogenization, the granules are pressed into tablets with filling and/or the granules are pressed into tablets with filling, which are heat treated at a temperature of 50-53°C and their surfaces are optionally coated with a film of permeable polyvinyl butyral in embedded method, regulating the permeability of the polymer coating by the addition of water-soluble polyethylene glycol with a molar weight of approximately 6000. The method according to the invention is suitable for the production of frame tablets with a slow action of active substances of various solubility. The release of water-soluble substances is independent of the pH conditions. , time of movement/ prevailing in the digestive tract (stomach and intestines). The release, depending on the therapy, can be changed within wide limits. The preparation passes through the stomach and intestines without any obstacles or inhibitions and releases the active substance continuously. This is confirmed by radiological examination and blood tests. The preparations contain all the active substance and provide a constant therapeutic picture of the blood. During the secretion of the active substance, the filling shrinks and, when the active substance is completely released, it forms a kind of sponge that does not irritate the mucous membranes and does not cause any damage.145 663 3 The production of frame tablets with a slow action of active substances using a small amount of auxiliaries is simple and economical. The delayed release of the active substance leads to economical use of technical measures in therapy. The method according to the invention, without limiting it, is described in more detail by the following examples. Example: 1.50.00 kg of potassium chloride and 0.05 kg of magnesium stearate, after homogenization, were moistened with a solution of 1.75 kg of polyvinyl butyral, 0.20 patent blue. A and 0.75 kg of methyl alcohol in a mixture of 87.30 g of distilled water and 4.912 kg of 96 percent ethanol. The wet mixture was granulated, dried at 40°C and granulated again. The resulting dry granulate (weighing 52.55 kg) was moistened with a solution of 1.125 kg of polyvinyl butyral in 5.0 kg of 96 percent ethanol and the above granulation was repeated. The prepared granulate (weighing 53.65 kg) was homogenized with a mixture of 0.100 kg of cetyl alcohol and 0.400 kg of talc and then pressed into tablets with an average weight of 1.0835 mg and processed in a polishing drum (driller) for 20 minutes at a temperature of 51-53 °C.Example II. 502.0 g of dry zinc sulfate were moistened with a solution of 25.0 g of polyvinyl butyral in 70.0 g of 96 percent ethanol. The wet mixture was granulated, dried at 40°C and granulated again. The obtained dry granulate (weight 527.0 g) was homogenized with 380.0 g of mannitol, moistened with a solution of 35.0 g of polyvinyl butyral and 8 g of pigment in 100.0 g of 96 percent ethanol and 20.0 g of cetyl alcohol, and the above was repeated. granulation. The prepared granulate (weighing 970.0 g) was homogenized with a mixture of 24.0 g of talc and 6.0 g of cetyl alcohol and then pressed into tablets with an average weight of 250 mg and processed in a polishing drum for 20 minutes. minutes at a temperature of 51 - 52°C. Example III. After homogenization, 502.0 g of zinc sulfate and 380.0 g of milk sugar were moistened with a solution of 45.00 g of polyvinyl butyral in 20.00 g of cetyl alcohol and 140.00 g of 96 percent ethnol. The wet mixture was granulated, dried at 40°C and granulated again. The resulting granulate (weighing 947.0 g) was moistened with a solution of 15.0 g of polyvinyl butyral and 8.0 g of pigment in 150.0 g of 96 percent ethanol and the above granulation was repeated. The resulting granulate (weighing 970.0 g) was homogenized with a mixture of 24 0.0 g of talc and 6.0 g of cetyl alcohol and finally compressed into tablets with an average weight of 250 mg and processed in a polishing drum for 20 minutes at a temperature of 51 - 52°C. Example IV. 50.00 kg of potassium chloride was moistened with a solution of 1.75 kg of polyvinyl butyral and 0.75 kg of cetyl alcohol in a mixture of 2.50 kg of chloroform and 3.50 kg of isopropanol.
