Przedmiotem wynalazku jest zastosowanie aktywnego białka o następującej sekwencji:

Aktywność białka pl4 przeciwko wirusowi HIV demonstrowano przy użyciu surowicy zwierzęcej lub ludzkiej, uprzednio immunizowanej p14. W tym celu, białko podawano podskórnie w dawce 1-2 mg, co siedem dni w czasie 4-6 tygodni, do uzyskania znacznego miana przeciwciał anty-p14. Otrzymaną surowicę inkubowano w komórkach linii E (pochodne ludzkiej T-limfocytowej linii komórkowej HuT78 przewlekle zakażonej wirusem HiV-1sf2) lub w normalnych, niezakażonych komórkach HuT78.
Wymienione komórki mieszano w stosunku 1:1000 (1 komórka linii E wytwarzająca wirus na 1000 normalnych komórek Hut78), nanoszono na 24-dołkowe płytki w stężeniu 1 x 105 komórek/dołek w 1 ml pożywki hodowlanej RPMI 1640, zawierającej 5% bydlęcej surowicy płodowej i antybiotyki. Do każdego dołka dodawano następnie badaną surowicę przy końcowym stężeniu 5%, tak aby końcowe stężenie surowicy ogółem wyniosło 10%. Próbki kontrolne zawierały pożywkę hodowlaną RPMI 1640 z 5% bydlęcej surowicy płodowej i 5% normalnej surowicy ludzkiej.
Jako kontrolę pozytywną stosowano króliczą wysokoodpomościową surowicę przeciwko HIV-1, która jest zdolna do hamowania zakażenia wirusem o około 3 log.
Po 5 dniach inkubacji, pożywki hodowlane odwirowano i wytworzony wirus oznaczono ilościowo w oznaczeniu wychwytu antygenu rdzenia p24 HTV-1.
Badane surowice wykazywały wysoki odsetek procentowy hamowania, sugerując terapeutyczną aktywność białka p14, które może być stosowane jako immunogenny antygen. Można stosować również przeciwciała wytworzone przeciwko temu białku.
Taką aktywność, potwierdzono również, jakkolwiek w ograniczonej liczbie przypadków, także w warunkach in vivo u pacjentów HIV-pozytywnych i pacjentów klinicznie chorych na AIDS.

Po każdej iniekcji i przed podaniem mierzono następujące parametry:

- ogólną liczbę limfocytów;

Przy końcu leczenia parametry serologiczne wykazywały tendencje do poprawienia, zaś cechy hematologiczne - normalizację.
Kilka lat po leczeniu pacjenci nadal żyli i ich stan był zadowalający.

Zgodnie z niniejszym wynalazkiem, białko pl4 można podawać w odpowiedniej postaci, zwykle w postaci iniekcji, ewentualnie zawierającej konwencjonalne adiuwanty, takie jak, wodorotlenek glinu, polisacharydy, białka nośnikowe i tym podobne.
Sposób podawania (dawki, częstotliwość podawania i tym podobne) określany będzie zgodnie z warunkami, zależnie od różnych czynników, takich jak, stan pacjenta, stan zaawansowania choroby, parametry hematologiczne i serologiczne. Miano przeciwciała anty-p14 można wykorzystać w celu śledzenia postępu terapii, wraz z parametrami zwykle stosowanymi do oceny właściwego funkcjonowania immunologicznego. Zastosowanie według zastrz. 1, znamienne tym, że stosowane białko wykazuje przynajmniej 80% homologii do sekwencji przedstawionej w zastrzeżeniu 1.Przedmiotem niniejszego wynalazku jest zastosowanie białka o masie cząsteczkowej około 14 kDa, ekstrahowanego z organów ssaków, do wytwarzania leku o aktywności przeciwko retrowirusom, w szczególności przeciwko wirusowi HIV, wirusowi ludzkiego niedoboru odpornościowego.Dobrze znane są terapeutyczne problemy związane z zakażeniem retrowirusem HIV. W szczególności problemy związane z rozwojem zakażenia i ostrego przebiegu choroby wywołanej przez wirus, znanej jako nabyty zespół niedoboru odpornościowego lub AIDS, spowodowały podejmowanie olbrzymiej ilości badań, które dostarczyły wyników pozwalających na ograniczenie choroby i leczenie zakażenia. Na przykład ostatnio, inhibitory proteazy łączono z inhibitorami odwrotnej transkryptazy, takimi jak, azydotymidyna.
