Six independent primary transformants were thus obtained and all transgenic lines were self-crossed. From each independent transgenic line, 40 distinct seeds were germinated in the soil and checked for altered phenotypes compared to wild-type plants. Four of the six lines showed an altered flowering time phenotype. The seeds from these plants did not germinate well, and if this did, the plants stopped at the seedling stage. Both effects could be reversed by transferring the plants to a culture medium containing 10-5 M GA3 and culturing them in this medium for three days. After recovery and transfer to soil, plants developed normally except for the late flowering phenotype. Under short-day conditions, transgenic plants produce significantly more rosette leaves (vegetative amounts) than wild-type plants (about 40 rosette leaves and 100 days after germination and the plants are still not flowering, compared to about 30 rosette leaves before flowering for wild-type plants) ). Under LD conditions, most of these plants experience a partial reversal of generative to vegetative growth, evidenced by the formation of overhead rosettes (vegetative leaves) on the inflorescence stem. Plants (C24 ecotype) containing an extra copy of the ATH1 cDNA under the control of the Hspl8.2 heat shock gene promoter (HspHiB plants) also showed a late-flowering phenotype.
