PL PLDescription of the invention The subject of the invention is a method for obtaining material for test fields, especially dry tests, which is a cellulose substrate covered with a semi-permeable polymer membrane made of a non-reactive polymer that forms pores that do not collapse without filling, despite repeated immersion and drying in water and/or other solvents. There are many types of test field materials known that allow for the separation of structured elements contained in the analytical matrix on the test surface. For example, US patent 3092465 describes the preparation of a test for determining glucose in whole blood, where the filter paper is saturated with a solution of reagents containing gelatin. A similar solution was used in the description of Polish patent PL 166404, where a layer containing a set of reagents suspended in gelatin is applied on the surface of the filter paper, creating a swelling, reactive and semi-permeable indicator layer after drying. US patent 4258001 describes a solution consisting in covering a polyethylene glycol terephthalate carrier with a reaction layer consisting of reagents reacting with with the analyte suspended in a membrane-forming mixture based on high-molecular-weight natural polymers. A similar solution is presented in U.S. Patent 4,486,537, where a semi-permeable layer composed of a high-molecular-weight synthetic polymer is deposited on a polyethylene glycol terephthalate carrier, creating a semi-permeable film on an impermeable, but highly light-transmitting, chemically resistant carrier layer. These tests are used to analyze the content of various components in whole blood, such as glucose, urea, or cholesterol in whole blood. There is also a more general solution developed by Boehringer Mannheim, which involves separating erythrocytes and other shaped elements from whole blood using a nonwoven fabric. An example of such a solution is presented in German patent DE 3130749 for a cholesterol test. The principle of this test is that separation occurs by both sieve filtration and chromatogram development. The blood is then placed on a glass nonwoven fabric substrate, where lower molecular weight elements are separated from the shaped elements of the blood. This principle has also been used in other solutions described, for example, in German patents: DE Patent No. 4001155; DE Patent No. 4005-4021. The principle of operation of this type of solution was also described by Vogel et al. (Vogel P., Rittersdorf W., Thym D., Bartl K.: - Development of a potassium assay on the Reflotron. - Clin. Chem. 36(1990)107). This principle was used to develop several whole blood tests currently available on the market. However, there is currently no universal method for preparing tests for determinations in a wide variety of media, not only whole blood, but also fruit and vegetable juices and purees, water contaminated with sludge and various suspensions, liquid manure, suspended industrial wastewater, and many other media where the substances contained therein require preliminary separation of contaminants by filtration or centrifugation before analytical procedures can begin. Patent description GB2217717 describes a method in which a membrane-forming solution consisting of at least two incompatible polymers, which form two different phases in solution, is applied to a dry, non-porous polymer substrate. Fillers are added to this solution and gelled in water or another polymer non-solvent. Patent description EP0274911 describes a method in which a membrane made of synthetic polymers such as polyamide, polydifluorovinylidene, nylon or modified polyamide is saturated with reagent solutions and itself serves as a test field, which is then (example 1) applied to a substrate made of cellulose acetate (a cellulose derivative - a non-porous foil), to which it is bonded using an adhesive, by pressure (an additional, troublesome activity that may lead to undesirable reactions with the set of reagents in the membrane), and in addition, the adhesive is an additional load. Patent descriptions DE4438381 and US2002/0119577 describe a membrane made of a non-porous mixture of nitrocellulose and cellulose acetate applied to a polyester foil substrate. As a result, the membranes and test fields are several millimeters thick. Unexpectedly, it turned out that the method according to the invention makes it possible to develop a material for test fields with non-collapsing pores, in which a porous carrier made of pure cellulose has a very thin layer of a semi-permeable membrane bonded directly to this substrate, without an additional layer of glue, with appropriate porosity and permeability, intended for the production of test fields, especially dry tests. PL 203 559 B1 3 The method according to the invention fills the gap that has existed so far, allowing the production of a cellulose base covered with a semi-permeable membrane, which is a raw material for obtaining test field material, from which tests for determining n in aqueous suspensions, blood as well as in juices and vegetable purees. The method according to the invention consists in applying, by known methods, to a cellulose support saturated with a polymer non-solvent, preferably chromatographic quality paper, a thin layer of a membrane-forming polymer solution consisting of a polymer or a mixture of polymers selected from the group comprising synthetic polymers formable by the phase inversion method, preferably selected from the group comprising polyamides, polysulfones or polyethersulfones, in a polymer solvent, of controlled thickness. The solution of the membrane-forming polymer or membrane-forming polymers, optionally in addition to the membrane-forming polymer, may contain one or more pore-forming agents, preferably selected from the group comprising low-molecular-weight polyvinyl alcohol, polyethylene and propylene glycols and polyvinylpyrrolidones. The membrane is then formed using the phase inversion method, by drying the applied thin membrane-forming layer, or preferably by precipitation with a non-solvent. The non-solvent can be a single chemical entity or a solution of multiple substances. After rinsing the pore-forming agent from the resulting membrane and drying, the resulting paper coated with a semi-permeable membrane is ready for preparing test fields. In the invention, a homogeneous solution consisting of one polymer or a mixture of compatible polymers, which allows for obtaining a semi-permeable membrane with a thickness of several to several dozen micrometers, is applied to a cellulose (porous) substrate previously saturated with a polymer non-solvent (the substrate must not be dry), without the addition of insoluble fillers. The material for test fields obtained by the method according to the invention is characterized in that it has pores that do not collapse during repeated drying, and it is a material made of pure cellulose, previously saturated with a non-solvent or a mixture of non-solvents of a polymer or polymers, with a very thin layer of a membrane-forming polymer selected from the group consisting of polyamides, polysulfones and polyethersulfones, with an optional addition of pore-forming agents selected from the group consisting of low-molecular polyvinyl alcohols, polyethylene and propylene glycols or polyvinylpyrrolidones, which at the same time constitutes a connection with a substrate made of pure cellulose. In our method, the test field consists solely of a layer of pure cellulose, previously saturated with a non-solvent. This prevents the membrane-forming mixture from penetrating the substrate and ensures that the layer binding the cellulose membrane is several micrometers thick, while the semi-permeable layer, depending on needs, is several to several dozen micrometers thick, rather than millimeters thick as in previously known solutions. The solution we used has the advantage that the membrane applied to the cellulose substrate does not tend to collapse its pores during repeated wetting and drying, unlike membranes made of cellulose acetate, cellulose nitrate, and their blends, where a single wetting and drying causes complete pore collapse. As mentioned above, in our case, the substrate is cellulose with a previously applied semi-permeable layer. This layer is several microns thick, not several millimeters, and does not constitute a reaction layer as in the known solution. The reaction layer only constitutes the cellulose substrate, and only after saturation with the appropriate reaction mixture. These mixtures can vary greatly depending on the analyte being determined. This results in the test field, after the reagents have been applied, being several microns thick, because the polymer only penetrates to this depth in cellulose saturated with the antisolvent of the polymer used. The essence of our invention is a method of covering a cellulose substrate with a semi-permeable membrane so that the membrane forms a durable bond with the cellulose, but does not penetrate it any deeper than a few micrometers. The semi-permeable membrane itself has a thickness ranging from several to several dozen micrometers, depending on the needs, and does not exhibit pore collapse during repeated saturation with aqueous solutions and drying. The material and method according to the invention are presented in the examples below.
