Opis wynalazkuDescription of the invention
Przedmiotem wynalazku są nowe sacharyniany i acesulfamiany 1-alkoksymetylo-3-hydroksypirydyniowe oraz sposób wytwarzania sacharynianów i acesulfamianów 1-alkoksymetylo-3-hydroksypirydyniowych.The present invention relates to new 1-alkoxymethyl-3-hydroxypyridinium saccharinates and acesulfamates and a process for the preparation of 1-alkoxymethyl-3-hydroxypyridinium saccharinates and acesulfamates.
Smak ten ogranicza stosowanie tych wielofunkcyjnych związków. Chlorki pirydyniowe zawierające podstawnik oktylowy, decylowy, dodecylowy, tetradecylowy lub heksadecylowy mogą przedostawać się do środowiska wodnego, w którym trudno ulegają, biodegradacji. Ciągle poszukuje się nowych soli pirydynowych, które zachowywałyby wysoką aktywność wobec bakterii, a jednocześnie łatwo ulegałyby biodegradacji bądź były podatne na jakąś inną metodę utylizacji.Pyridinium salts containing a pyridine ring and a quaternary nitrogen atom with an alkyl group are frequently cited in Chemical Abstract. The group of these compounds has its keyword pyridinium compounds. The best known are 1-alkylpyridinium chlorides and bromides, which are highly hygroscopic. They are multifunctional salts belonging to cationic surfactants: they are strongly active against bacteria and fungi, they have anti-electrostatic and anti-caking properties. Aqueous solutions of pyridine chlorides have a bitter taste. This taste limits the use of these multifunctional compounds. Pyridinium chlorides containing an octyl, decyl, dodecyl, tetradecyl or hexadecyl substituent may be released into the aquatic environment where they are difficult to biodegrade. There is a constant search for new pyridine salts that would retain high activity against bacteria and at the same time would be easily biodegradable or would be susceptible to some other method of disposal.
Also known and described is sodium saccharinate (systematic name: 1,2-benzisothiazoline-3-one-1-sodium 1-dioxide). It is a popular sweetener as a sugar substitute. It is in the form of white crystals or white fine crystalline powder. It is characterized by an intense sweet taste, even 300 to 500 times sweeter than sucrose in dilute solutions.
Also known and described is acesulfame-K, a low calorie sweetener approved as a food additive in 1988. It is a synthetic organic salt (systematic name - 5,6-dimethyl-1,2,3-oxathiazin-4 (3H) -one [2,2-dioxide] potassium salt), 200 times sweeter than sucrose. It works synergistically with other synthetic sweeteners, is resistant to high temperatures and has a long shelf life. It is excreted from the human body unchanged and therefore does not provide any calories.
Znane są chlorki 1-alkoksymetylo-3-hydroksypirydyniowe (J. Pernak, M. Branicka, The Properties of 1-Alkoxymethyl-3-hydroxypyridinium and 1-Alkoxymethyl-3-dimethylaminopyridinium Chlorides, Journal of Surfactants and Detergents, 6, 119-123, 2003). Chlorki 1-alkoksymetylo-3-hydroksypirydyniowe jako nieliczne czwartorzędowe sole amoniowe ulegają rozkładowi pod wpływem ozonu (J. Pernak, M. Branicka, Synthesis and Aqueous Ozonation of Some Pyridinium Salts with Alkoxymethyl and Alkylthiomethyl Hydrophobic Group, Ind. Eng. Chem. Res., 43, 1966-1974, 2004).1-Alkoxymethyl-3-hydroxypyridinium chlorides are known (J. Pernak, M. Branicka, The Properties of 1-Alkoxymethyl-3-hydroxypyridinium and 1-Alkoxymethyl-3-dimethylaminopyridinium Chlorides, Journal of Surfactants and Detergents, 6, 119-123 , 2003). They are obtained from 3-hydroxypyridine and chloromethylalkyl ethers in the quaternization reaction, which follows the SN1 mechanism. This is a special case of Menschutkin's reaction, which follows the SN2 mechanism. 1-Alkoxymethyl-3-hydroxypyridinium chlorides as a few quaternary ammonium salts decompose under the influence of ozone (J. Pernak, M. Branicka, Synthesis and Aqueous Ozonation of Some Pyridinium Salts with Alkoxymethyl and Alkylthiomethyl Hydrophobic Group, Ind. Eng. Chem. Res. , 43, 1966-1974, 2004).

Acesulfamian 3-hydroksy-1-undecyloksymetylopirydyniowy3-Hydroxy-1-undecyloxymethylpyridinium acesulfamate
Do kolby okrągłodennej o pojemności 100 cm3 wprowadzono 0,01 mola chlorku 3-hydroksy-1-undecyloksymetylopirydyniowego rozpuszczonego w 40 cm3 wody destylowanej o temperaturze 333 K. Następnie przy ciągłym mieszaniu dodawano stechiometryczną ilość acesulfamu K rozpuszczonego w wodzie. Otrzymano sól krystaliczną z wydajnością 90%, o temperaturze topnienia 351-353 K.0.01 mole of 3-hydroxy-1-undecyloxymethylpyridinium chloride dissolved in 40 cm 3 of distilled water at a temperature of 333K was introduced into a 100 cm 3 round bottom flask. Then, a stoichiometric amount of acesulfame K dissolved in water was added with continuous stirring. The whole was stirred vigorously at 333 K. 40 cm 3 of chloroform were added, after mixing and separation of the layers, the lower layer was separated and washed several times with water until there was no longer any chloride ions in the effluent. The presence of chloride ions was monitored with an aqueous solution of silver nitrate. Chloroform was distilled off on a rotary evaporator. A crystalline salt was obtained with a yield of 90%, m.p. 351-353 K.
