Opis wynalazkuDescription of the invention
Przedmiotem wynalazku jest sposób otrzymywania olejku eterycznego z surowców roślinnych metodą destylacji z zastosowaniem pary wodnej. The subject of the invention is a method of obtaining essential oil from plant materials by steam distillation.
Znany w stanie techniki sposób otrzymywania olejków eterycznych z zastosowaniem pary wodnej polega na umieszczeniu substancji roślinnej w kolbie destylacyjnej, dodaniu wody i ogrzewaniu mieszaniny w urządzeniu grzewczym z płynną regulacją temperatury. W dotychczasowym sposobie destylacji z parą wodną olejek eteryczny otrzymuje się poprzez doprowadzenie mieszaniny surowca z wodą do wrzenia. Powstają cy destylat jest zbierany w rurce kalibrowanej aparatu do destylacji, gdzie olejek eteryczny przechodzi do warstwy ksylenowej, a warstwa wodna zawracana jest automatyczne do kolby destylacyjnej. The method of obtaining essential oils with the use of steam known in the art consists in placing the plant substance in a distillation flask, adding water and heating the mixture in a heating device with smooth temperature control. In the prior art steam distillation process, the essential oil is obtained by bringing a mixture of the raw material and water to the boil. The resulting distillate is collected in the tube of the calibrated distillation apparatus, where the essential oil passes into the xylene layer and the aqueous layer is automatically returned to the distillation flask.
As a result of the tests carried out, it has surprisingly turned out that it is possible, by using ultrasonic waves, to obtain an additional amount of essential oil obtained in a traditional way in the distillation process. The use of ultrasonic waves has a positive effect on the process of the release of volatile compounds from the secretory structures present in plant raw materials.
The method of obtaining the essential oil according to the invention is carried out in two stages, in the first stage the macerate prepared as a result of maceration of plant raw material with water is immersed together with a distillation flask connected with an apparatus for distilling essential oils in a bath filled with a high-boiling liquid, preferably with vegetable or mineral oil, and heated at the boiling point of the macerate for a minimum of 30 minutes by heating the oil in a bathtub, keeping its temperature at a level of at least 120 ° C, and after the distillation process is completed, the essential oil is obtained in a calibrated tube, and then, after cooling the macerate in the flask, a second a distillation stage in which the flask with macerate is subjected to ultrasound by switching on the ultrasonic apparatus, the elements of which being the source of ultrasound are placed in the bath, the distillation being carried out at the temperature and time specified in the first stage, and after completing the distillation, d an additional amount of essential oil.
The ultrasonic action is preferably started after the first drop of distillate has been obtained.
Preferably, the effect of ultrasound is sustained for the duration of the distillation.
Korzystnie jest, gdy temperatura oleju w pierwszym i w drugim etapie destylacji wynosi 140°C.Preferably, the oil temperature in the first and second distillation stages is 140 ° C.
Preferably, the high-boiling liquid is a polar organic solvent, for example glycerin or propylene glycol.
Zastosowanie ultradźwięków powoduje zwiększenie efektywności procesu destylacji, poprzez degradację roślinnych struktur wydzielniczych, a tym samym, uwolnienie dodatkowej ilości olejku eterycznego w etapie redestylacji. In the method according to the invention, through the use of ultrasound, it is possible to obtain from the macerate remaining after the first distillation step, an additional amount of essential oil, which increases the total content of active compounds extracted from the plant raw material. The proposed method of distillation is particularly advantageous for those substances that are present in the raw material in special secretion structures. As a result of classical distillation, the frequently released amount of essential oil is not complete. The use of ultrasound increases the efficiency of the distillation process by degrading the plant secretion structures, and thus releasing an additional amount of essential oil in the redistillation stage.
The use of ultrasound in the method according to the invention is possible by using a vegetable or mineral oil as the medium for the propagation of ultrasonic waves. In addition, the use of oil as a medium with high-boiling properties makes it possible to adjust the oil boiling point, thanks to which it is possible to obtain the desired temperature inside the flask.
P r z y k ł a d I. Proces destylacji przeprowadzono dwuetapowo, wykorzystując koszyczki rumianku (Chamomilla recutita (L.) Rauschert).Example I. The distillation process was carried out in two stages, using chamomile baskets (Chamomilla recutita (L.) Rauschert).
W pierwszym etapie odważ ono 20 g wysuszonych i rozdrobnionych koszyczków rumianku. Po upływie 10 min. odczytanoIn the first stage, weigh out 20 g of dried and crushed chamomile baskets. The raw material was transferred to a ground glass flask with a capacity of 1000 ml and 400 ml of distilled water were poured over, the flask was closed with a ground glass stopper. The raw material was macerated on an RS 10 control mechanical shaker for 30 minutes with continuous stirring at room temperature (at the speed of 130 cycles per minute), then the macerate flask was connected to the oil distillation apparatus according to FP VII. The whole apparatus was placed in a UM-4 ultrasonic bath filled with oil as a medium. Heating was carried out at the temperature of 120 ° C without the ultrasounds turned on, for 30 minutes from the receipt of the first drop of distilled oil in the receiver to which 0.3 mL of m-xylene had been previously added. The heating of the oil in the bath and the control of the temperature increase was carried out using a heater and a thermal controller with a sensor. After the predetermined time had elapsed, the distillation apparatus with the flask was raised above the oil line in the ultrasonic bath. After 10 min. read

Then the oil was quantitatively transferred to a special container, anhydrous sodium sulfate was added to remove the residual water and sealed.
The percentage of essential oil components and their content were analyzed by GC-MS, GC-FID chromatography. The analysis was repeated 3 times.
In order to compare the hydrodistillation efficiency, the distillation of the oil was performed analogically for the tested raw material in accordance with the standards described in Polish Pharmacopoeia VII. The oil was also analyzed by GC-MS and GC-FID gas chromatography. The analysis was repeated 3 times.
Tak przygotowaną mieszaninę analizowano metodą GC-MS i GC-FID.The analytical procedure was that the essential oil obtained in the previous steps was diluted 1000-fold with n-hexane by adding 100 μΐ of the mixture of internal standards C 12 , C19, C24 (1 mg / mL). The mixture prepared in this way was analyzed by GC-MS and GC-FID.
Analizę GC-MS prowadzono przy użyciu aparatu Varian 4000 GC-MS/MS (Varian, USA), wyposażony w automatyczny dozownik CP-8410, kolumnę VF-5ms (Varian, USA) o długości 30 m; średnicę 0,25 mm i grubość fazy stacjonarnej 0,25 μ^ι, gaz nośny Hel - przepływ stały 0,5 mL/min. Temperatura dozownika i detektora (pułapka jonowa) odpowiednio 250°C i 200°C, Split 1:100. Dozowano 5 μL próby. Program temperatury dozownika: 50°C przez 1 min., narost do 250°C z szybkością 4°C/min., 250C przez 10 min. Energia jonizacji 70 eV, zakres mas 40-870 Da, czas skanowania 0,80 sek.GC-MS analysis was performed using a Varian 4000 GC-MS / MS (Varian, USA) equipped with an automatic injector CP-8410, a VF-5ms column (Varian, USA) 30 m long; diameter 0.25 mm and thickness of the stationary phase 0.25 μ ^ ι, carrier gas Hel - constant flow 0.5 mL / min. Temperature of the dispenser and detector (ion trap) 250 ° C and 200 ° C respectively, Split 1: 100. 5 μL of the sample was dispensed. Dispenser temperature program: 50 ° C for 1 min., Increase to 250 ° C at a rate of 4 ° C / min., 250C for 10 min. Ionization energy 70 eV, mass range 40-870 Da, scan time 0.80 sec.
Analizę GC-FID prowadzono stosując układ GC Varian 3800 wyposażony w automatyczny dozownik CP-840, kolumnę DB-5 o długości 30 m, średnicy 0,25 mm i grubości fazy stacjonarnej 0,25 μm, gaz nośny Hel - przepływ stały 0,5 mL/min., temp. dozownika i detektora (FID) były ustalone na 260°C, split 1:100, objętość dozowanej próby: 5 μL. Program temperaturowy kolumny: 50°C przez 1 min., narost do 250°C z szybkością 4°C/min., 250°C przez 10 min.The GC-FID analysis was carried out using the GC Varian 3800 system equipped with an automatic CP-840 dispenser, a DB-5 column 30 m long, 0.25 mm in diameter and 0.25 μm in the stationary phase thickness, Helium carrier gas - constant flow 0.5 mL / min, dispenser and detector (FID) temperatures were set to 260 ° C, split 1: 100, sample volume dispensed: 5 μL. Column temperature program: 50 ° C for 1 min., Rise to 250 ° C at 4 ° C / min., 250 ° C for 10 min.
The quantitative analysis was carried out on the basis of the previously determined dependences of the ratio of the surface area of the tested compounds standards to the surface area of the corresponding internal standard (C12 for compounds with IR <1450, C19 for compounds with 1450 <IR <2150, C24 for compounds with IR> 2150).

Jak przedstawiono w tabeli I.2. ilość składników olejku eterycznego otrzymanego w wyniku destylacji z parą wodną, zarówno w wannie ultradźwiękowej (bez włączonych ultradźwięków) jak i metodzie farmakopealnej były niemal równe 183,70 μg/g surowca (etap I) i 184, 66 μg/g surowca (FP VII). Ponowna destylacja z zastosowaniem ultradźwięków pozwoliła na destylację dodatkowej ilości olejku eterycznego 89, 05 μg/g surowca. As shown in Table I.2. the amount of essential oil components obtained by steam distillation, both in the ultrasonic bath (without the ultrasound on) and in the pharmacopoeial method, was almost equal to 183.70 μg / g of raw material (stage I) and 184.66 μg / g of raw material (FP VII ). Re-distillation with the use of ultrasound allowed the distillation of an additional amount of essential oil 89.05 μg / g of raw material.
P r z y k ł a d II. Olejek eteryczny otrzymywano z korzeni arcydzięgla. W tym celu odważono 20g wysuszonego i rozdrobnionego korzenia arcydzięgla (Archangelica officinalis Hoffm.). Proces destylacji i otrzymywania olejku eterycznego prowadzono jak w przykładzie I. Ocenę analityczną prowadzono stosując sprzęt, aparaturę i procedury analityczne jak opisano w przykładzie I.P r z x l a d II. The essential oil was obtained from the roots of angelica. For this purpose, 20 g of dried and shredded angelica root (Archangelica officinalis Hoffm.) Was weighed. The process of distillation and obtaining the essential oil was carried out as in Example I. Analytical evaluation was carried out using the equipment, apparatus and analytical procedures as described in Example I.
Poniżej w tabelach podano procentową zawartość olejku eterycznego z korzeni arcydzięgla wg. FP VII oraz sposobem według wynalazku (tabela II. 1) oraz udział procentowy poszczególnych składników olejku eterycznego otrzymanego z korzeni arcydzięgla (tabela II.2.).The tables below show the percentage of essential oil from angelica roots by FP VII and the method according to the invention (Table II. 1) and the percentage of the individual components of the essential oil obtained from angelica roots (Table II.2.).

Jak przedstawiono w tabeli II.2 ponowne wykorzystanie surowca do destylacji z parą wodną, wspomaganej ultradźwiękami pozwoliło otrzymać dodatkową porcję olejku eterycznego 22,05 μg/g surowca. As shown in Table II.2, the reuse of the feedstock for ultrasonic assisted steam distillation resulted in an additional portion of essential oil of 22.05 μg / g of feedstock. In the known pharmacopoeial method, the raw material is treated as waste after steam distillation. In the proposed methodology with the use of ultrasound, it is possible to redistill it, as a result of which we can double their content for some components. Essential oil in plants can be found in numerous products, e.g. in the secretory ducts. Simply heating the raw material may not be sufficient to release the entire oil content. Therefore, as shown in example 2, it is advisable to use ultrasound in order to increase the use of the raw material to isolate more oil.
