Podstawowymi gatunkami z rodzaju Lactobacillus zasiedlającymi ludzki przewód pokarmowy są między innymi: Lactobacillus rhamnosus, Lactobacillus plantarum, Lactobacillus fermentum, Lactobacillus casei, Lactobacillus acidophilus, zaś z rodzaju Bifidobacterium od ludzi izolowane zostały: Bifidobacterium infantis (z dwoma podgatunkami), Bifidobacterium longum, Bifidobacterium breve, Bifidobacterium bifidum, Bifidobacterium adolescentis, Bifidobacterium dentium, Bifidobacterium catenulatum i Bifidobacterium angulatum.The basic species of the genus Lactobacillus inhabiting the human gastrointestinal tract are, among others: Lactobacillus rhamnosus, Lactobacillus plantarum, Lactobacillus fermentum, Lactobacillus casei, Lactobacillus acidophilus, while from the genus Bifidobacterium from humans were isolated: Bifidobacterium Bifunkimus, Bifidobacterium longobacterium, , Bifidobacterium bifidum, Bifidobacterium adolescentis, Bifidobacterium dentium, Bifidobacterium catenulatum and Bifidobacterium angulatum.

Właściwości adherencyjne szczepu Lactobacillus plantarum PL02 zostały przeprowadzone na ludzkiej linii komórek jelitowych HT-29MTX. Działanie metotrexatu (MTX) indukuje tkankę HT-29 do wydzielania zwiększonej ilości śluzu, dzięki czemu ten model tkankowy bardziej przypomina naturalne właściwości jelita grubego zdrowego człowieka. The mixture prepared in this way was added to the wells containing Ht-29MTX cells. The cells of the tissue line with the test strain were incubated at 37 ° C in an atmosphere enriched with 10% CO2 for 30 minutes. After incubation, cells were washed twice with PBS to remove non-attached bacteria, then fixed with 3.7% formaldehyde at room temperature for 1 hour. After fixation, cells were washed again with PBS and stained with crystal violet for 3 minutes. The preparation was examined microscopically at 1000 x magnification. Lactobacillus plantarum PL02 cells were counted and adhered to the tissue line in 20 fields of view of the preparation. Uzyskane w ten sposób pojedyncze kolonie zawieszano w 1 ml płynnego podłoża MRS i inkubowano w tych samych warunkach przez 24 godz. Nastę pnie przy wykorzystaniu soli fizjologicznej doprowadzano gęstość hodowli do 0,5 według skali McFarland'a. W celu oznaczenia oporności badanego szczepu Lactobacillus plantarum PL02 na sole żółci posłużono się metodą Dashkevicz'a i Feighner'a. Metoda ta polegała na dodaniu do agaru MRS barwnika wskaźnikowego - purpury bromokrezolowej w ilości 0,17 g/l oraz soli żółci w pięciu kolejnych stężeniach: 1 g/l, 2 g/l, 5 g/l, 10 g/l, 20 g/l. Na tak przygotowane, stałe podłoża MRS posiewano metodą redukcyjną 100 μΐ hodowli szczepu Lactobacillus plantarum PL02, który następnie inkubowano w standardowych warunkach beztlenowych przez 48 godz. Do badania antagonistycznego działaniem Lactobacillus plantarum PL02 na wzrost Candida albicans wybrano 4 grzyby z których 3 pochodziły z przewodu pokarmowego, zaś jeden był szczepem kolekcyjnym (Candida albicans ATCC 10231). Ponadto jako kontrolę antygrzybiczego działania bakterii kwasu mlekowego na grzyby zastosowano również szczep Lactobacillus rhamnosus GG. Do badania posłużono się zmodyfikowaną metodą półilościową według Fitzsimmons i Berry. Metoda polegała na zastosowaniu podłóż dwuwarstwowych. Pierwszą warstwę stanowiło podłoże MRS agar (Oxoid) na którym posiewano w formie paska o szerokości 2 cm badane szczepy probiotyczne z rodzaju Lactobacillus. Po wymazaniu płytki inkubowano przez 48 godzin w warunkach beztlenowych, a po upływie tego czasu na wierzch wylewano drugie, lekko przestudzone, płynne podłoże MYPG agar (Bacto YM Agar, Difco) w kierunku wzrostu grzybów wskaźnikowych. Po zastygnięciu na podłoże wysiewano grzyby z rodzaju Candida o gęstości 0,5 MacFarlanda. Następnie płytki były wkładane na 4 godziny do lodówki na 4°C, a następnie dalszą inkubacje prowadzono już w warunkach tlenowych w 37°C przez 24 godziny.
