Przedmiotem wynalazku jest sposób hodowli komórek macierzystych wyprowadzonych z rosnącego poroża jeleniowatych, zwłaszcza jelenia szlachetnego, w celu przygotowania przeszczepu służącego do regeneracji tkanki chrzestnej i kostnej.The present invention relates to a method of culturing stem cells derived from growing antlers of deer, especially red deer, in order to prepare a graft for the regeneration of cartilage and bone tissue.
W europejskim kręgu kulturowym, piękno twarzy wymaga zachowania symetrii poszczególnych jej elementów. Całkowity lub częściowy brak oraz wrodzone i nabyte zniekształcenia małżowiny usznej i nosa stanowić więc mogą poważny defekt kosmetyczny. W przypadku tych zniekształceń leczniczym postępowaniem z wyboru są skomplikowane, często wieloetapowe zabiegi chirurgiczne, z uż yciem dostę pnych organicznych wszczepów. Poszukiwania idealnego materiał u do rekonstrukcji rusztowania chrzestnego małżowin usznych i nosa mają ponad 100-letnią historię i są wciąż intensywnie prowadzone. Własna żebrowa chrząstka gospodarza była i jest najczęściej używana jako materiał rekonstrukcyjny w chirurgii twarzowej. Niefortunnie czas potrzebny do pozyskania chrząstki utrudnia zabieg, naraża pacjenta na pooperacyjny dyskomfort, przedłuża rekonwalescencję i zwiększa prawdopodobieństwo pojawienia się późniejszych komplikacji w miejscu pobrania materiału. Ponadto u dzieci ilość moż liwej do pozyskania chrzą stki ż ebrowej jest często zbyt mał a dla odtworzenia rusztowania małżowiny usznej lub nosa, a u dorosłych może być ona skostniała i nieprzydatna do formowania. Tych niedogodności pozwala uniknąć zastosowanie chrząstki allogennej - zgodnej gatunkowo, jako postępowanie z wyboru u każdego pacjenta, u którego nie jest dostępna wystarczająca ilość chrząstki własnej. Dla osłabienia odpowiedzi immunologicznej biorcy stosowane są wówczas różne metody jej przygotowania. Do popularnych sposobów należą kilkutygodniowe przechowywanie chrząstki w mertiolacie, wielokrotne płukanie w skoncentrowanym roztworze soli (NaCl) lub sterylizacja naświetlaniem promieniami gamma. Nie wykazująca aktywności metabolicznej chrząstka, pozostaje wówczas jedynie rusztowaniem dla otaczających tkanek i ulega powolnej resorpcji. Za kolejny etap w poszukiwaniu najodpowiedniejszego materiału organicznego do rekonstrukcji mogą być uważ ane badania Vacantiego i wsp. rozpoczęte w latach 90 XX wieku. W pionierskich eksperymentach użyto wówczas syntetycznej, biokompatybilnej, biodegradalnej, porowatej siatki polimerowej, którą impregnowano izolowanymi ludzkimi chondrocytami. In the case of these deformities, the therapeutic treatment of choice is complex, often multi-stage surgical procedures using available organic implants. The search for the perfect material for the reconstruction of the godfather of the auricles and the nose has over 100 years of history and is still intensively conducted. The host's own rib cartilage has been and is most often used as a reconstructive material in facial surgery. Unfortunately, the time needed to obtain the cartilage makes the procedure difficult, exposes the patient to postoperative discomfort, prolongs convalescence and increases the likelihood of later complications at the site of collection of the material. Furthermore, in children the amount of rib cage that can be recovered is often too small to recreate the scaffold of the auricle or nose, and in adults it may be ossified and unsuitable for molding. These inconveniences can be avoided by the use of allogeneic cartilage - compatible with species, as the treatment of choice for every patient who does not have enough own cartilage. In order to weaken the recipient's immune response, various methods of its preparation are then used. Popular methods include storing the cartilage in merthiolate for several weeks, repeated rinsing in concentrated salt solution (NaCl) or sterilization with gamma irradiation. Cartilage, which does not exhibit metabolic activity, then remains only a scaffold for the surrounding tissues and is slowly resorbed. The research by Vacanti et al., Started in the 1990s, may be considered the next stage in the search for the most suitable organic material for reconstruction. In pioneering experiments, a synthetic, biocompatible, biodegradable, porous polymer mesh was used, which was impregnated with isolated human chondrocytes. However, for most tissues, it is extremely difficult to obtain targeted cell lines. The collection of human stem cells is also often associated with ethical problems and the risk of transferring viral and neoplastic diseases. An alternative to human cells could be modified xenograft cells. Searching for their source, attention was drawn to the possible use of deer antlers, which are characterized by a specific, nowhere else, process of renewal and growth.
