Synteza związku porównawczego - izonikotynianu 2-metyloheptylu (wzór 2)Synthesis of reference compound - 2-methylheptyl isonicotinate (formula 2)
W kolbie trójszyjnej zaopatrzonej w chłodnicę zwrotną, mieszadło magnetyczne, termometr, łaźnię grzejną i absorber gazów (SO2, HCl) umieszczono kwas izonikotynowy (1,0 g, 0,008 mola) i chlorek tionylu (10 ml). Zawartość kolby mieszano w temperaturze wrzenia w czasie 0,5 godziny (do zakończenia wydzielania się SO2). Następnie do kolby podłączono nasadkę destylacyjną, chłodnicę destylacyjną, odbieralnik i pod zmniejszonym ciśnieniem (pompka wodna, 20 mm Hg) oddestylowano SOCI2 do sucha (pozostał osad o barwie jasnokremowej). Następnie kolbę reakcyjną ponownie zaopatrzono w chłodnicę zwrotną, połączoną z absorberem HCl, dodano 15 ml CH2CI2 i wkroplono roztwór alkoholu 2-metyloheptylowego (1,006 g, 0,0078 mola) w CH2CI2 (5,5 ml). Zawartość kolby mieszano w czasie 3 godzin w temperaturze wrzenia, do zakończenia wydzielania się chlorowodoru. Mieszaninę reakcyjną doprowadzono do pH 12 przy pomocy wodnego nasyconego roztworu K2CO3. Po rozdzieleniu warstw, wysuszeniu warstwy organicznej nad bezwodnym MgSO4 i odparowaniu rozpuszczalnika otrzymano surowy izonikotynian 2-metyloheptylu (1,718 g, wydajność 93,9%, olej o barwie jasnobrązowej). Produkt (1,5 g) oczyszczono metodą chromatografii kolumnowej stosując jako eluent: mieszaninę heksanu i octanu etylu w stosunku 9:1 i następnie mieszaninę heksanu - octanu etylu w stosunku 4:1. Otrzymano oczyszczony izonikotynian 2-metyloheptylu (1,32 g, wydajność 72,1%, olej o barwie jasno-żółtej).Isonicotinic acid (1.0 g, 0.008 mol) and thionyl chloride (10 mL) were placed in a three-neck flask equipped with a reflux condenser, magnetic stirrer, thermometer, heating bath, and a gas absorber (SO 2, HCl). The flask contents were stirred at reflux for 0.5 h (until SO 2 evolution ceased). Then, a distillation head, a distillation condenser, and a receiver were connected to the flask and SOCI 2 was distilled to dryness under reduced pressure (water pump, 20 mm Hg) (a pale cream-colored precipitate remained). The reaction flask was then re-fitted with a reflux condenser connected to an HCl absorber, 15 mL of CH 2 Cl 2 was added, and a solution of 2-methylheptyl alcohol (1.006 g, 0.0078 mol) in CH 2 Cl 2 (5.5 mL) was added dropwise. The contents of the flask were stirred for 3 hours at reflux until the evolution of hydrogen chloride had ceased. The reaction mixture was adjusted to pH 12 with an aqueous saturated solution of K 2 CO 3 . After separating the layers, drying the organic layer over anhydrous MgSO 4 and evaporating the solvent, crude 2-methylheptyl isonicotinate (1.718 g, 93.9% yield, light brown oil) was obtained. The product (1.5 g) was purified by column chromatography using a 9: 1 mixture of hexane and ethyl acetate as eluent and then a 4: 1 mixture of hexane-ethyl acetate. Purified 2-methylheptyl isonicotinate (1.32 g, 72.1% yield, light yellow oil) was obtained.
Kontrolę reakcji prowadzono za pomocą TLC, eluując mieszaniną benzen - metanol w stosunku 9:1, UV; benzen - octan etylu w stosunku 3:2, wywoływanie: UV, H2SO4 stężony z waniliną, 140°C.The reaction was monitored by TLC, eluting with a mixture of benzene-methanol 9: 1, UV; benzene - ethyl acetate in the ratio 3: 2, developing: UV, H 2 SO 4 concentrated with vanillin, 140 ° C.

Zawartość kolby mieszano w temperaturze wrzenia w czasie 2 godzin (do zakończenia wydzielania SO2). Następnie do kolby podłączono nasadkę destylacyjną, chłodnicę destylacyjną, odbieralnik i pod zmniejszonym ciśnieniem (pompka wodna, 20 mm Hg) oddestylowano SOCl2 do sucha (pozostałość to osad o barwie białej). Kolbę reakcyjną ponownie zaopatrzono w chłodnicę zwrotną połączoną z absorberem HCl, dodano 50 ml CH2Cl2 i wkroplono roztwór odpowiedniego alkoholu 2-metyloalkilowego (0,025 mola) w CH2Cl2 (10 ml). Zawartość reaktora mieszano w czasie 2 godzin w temperaturze wrzenia, do zakończenia wydzielania HCl. Mieszaninę reakcyjną doprowadzono do pH 12 przy pomocy wodnego, nasyconego roztworu K2CO3. Po rozdzieleniu warstw, wysuszeniu warstwy organicznej nad bezwodnym CaCl2 i odparowaniu rozpuszczalnika otrzymano surowy produkt. Jeżeli analiza TLC wykazała jednorodność surowego produktu, nie oczyszczano go. Natomiast w przypadku uzyskania mieszaniny, surowy produkt oczyszczano metodą chromatografii kolumnowej stosując jako eluent kolejno: heksan, heksan - octan etylu w stosunku 9:1, heksan - octan etylu w stosunku 4:1.Nicotinic acid (3.08 g, 0.025 mol) and thionyl chloride (12.5 ml) were placed in a three-neck flask equipped with a reflux condenser, magnetic stirrer, thermometer, heating bath, and a gas absorber (SO 2, HCl). The contents of the flask were stirred at reflux for 2 hours (until SO 2 evolution ceased). Then, a distillation head, a distillation condenser, and a receiver were connected to the flask and SOCl 2 was distilled to dryness under reduced pressure (water pump, 20 mm Hg) (the residue was a white solid). The reaction flask was re-fitted with a reflux condenser connected to an HCl absorber, 50 mL of CH 2 Cl 2 was added and a solution of the appropriate 2-methylalkyl alcohol (0.025 mol) in CH 2 Cl 2 (10 mL) was added dropwise. Badanie aktywności przeciwko Ascosphaera apisTest for activity against Ascosphaera apis
Skuteczność działania związku obliczono za pomocą wzoru Abbota:The acetone solutions of the compound were poured 1 ml each onto solidified Sabouraud agar medium and allowed to evaporate under aseptic conditions. On the prepared surface, discs of Ascosphaera apis mycelium 0.5 cm in diameter, cut out with a corkbore from a homogeneous culture grown in a Petri dish on a Sabouraud medium, were placed. The control was acetone. The plates were incubated at 25 ° C for 5 days. After this time, the diameter of the grown mycelia was measured. The efficacy of the compound was calculated using the Abbot formula:
Skuteczność Sg (%) = (Kg - Fg) x 100/kg,Efficiency Sg (%) = (Kg - Fg) x 100 / kg,
Gdzie: Kg - średnica kultury grzyba w kombinacji kontrolnej (średnia z 3 pomiarów),Where: Kg - diameter of the mushroom culture in the control combination (average of 3 measurements),
Fg - średnica kultury grzyba w kombinacji z badanym związkiem (średnia z 3 pomiarów).Fg - diameter of the mushroom culture in combination with the test compound (average of 3 measurements).
Miarą skuteczności działania jest procent zahamowania wzrostu grzybni. The measure of effectiveness is the percentage of inhibition of mycelial growth.

After 4-8 days, depending on the growth of the mycelium in the control combination, the linear growth of the mycelial colony was measured. The fungicidal activity is expressed as a percentage of mycelial inhibition compared to a control combination with the same composition but without the test substance.
Metoda B - test „in vivo przeprowadzono przy użyciu grzyba Blumeria graminis.Method B - In vivo test was carried out with Blumeria graminis fungus.
Wyniki ob ydwu testów podano w procentach zahamowania wzrostu grzybni. Wyniki przedstawiono w tablicy 3 i tablicy 4.The test was carried out under controlled conditions on wheat seedlings cv. Koksa. In the unfolded first leaf phase, plants were sprayed with a solution of the test compound at a concentration of 1000 mg / l. The solvent used was a mixture of acetone and distilled water in the ratio 1.5: 1, with the addition of Tween 20. Two hours after application, inoculation was performed (assessment of the preventive effect). Further vegetation was carried out in the phytotron under the following conditions: day length 14 hours, air temperature: 20 (± 1) ° C / day, 16 (± 1) ° C / night; relative air humidity 80 (± 5)%. Assessment of infestation was performed after a minimum of six days. The results of both tests are given as percent inhibition of mycelial growth. The results are shown in Table 3 and Table 4.
