Abstract
Otrzymaną warstwę organiczną suszy się w bezwodnym MgSO4, po czym tak otrzymany surowy półprodukt izoluje się na żelu krzemionkowym stosując jako eluent układ chlorek metylenu i metanol w stosunku (5:0,1), uzyskując w wyniku mono - chloro pochodną 9,10-antrachinonu z wydajnością około 80%.The subject of the invention are mono-chloro derivatives of 9,10-anthraquinone containing piperazine systems and a method for obtaining new derivatives of 9,10-anthraquinone containing piperazine systems. The invention concerns a group of new cytostatic antibiotics that inhibit cell division. The mono-chloro derivative of 9,10-anthraquinone is characterized by the fact that it is 1-chloro-4-(piperazin-1-yl)-9,10-anthraquinone in which R1 = Cl, R2 = H, R3 = H with the formula general 1. The method of obtaining the mono-chloro derivative of 9,10-anthraquinone is characterized by the use of 1,4-dichloro-9,10-anthraquinone as the starting substrate, which is reacted in a toluene medium or in an isopropanol medium. or in ethanol, heating the reaction mixture to a temperature of 100°C for about 24 hours, after which the compounds obtained after the reaction are washed with chemically and bacteriologically inactivated process water. The obtained organic layer is dried in anhydrous MgSO4, and then the crude intermediate thus obtained is isolated on silica gel using methylene chloride and methanol in a ratio (5:0.1) as an eluent, obtaining a mono-chloro derivative of 9,10-anthraquinone with an efficiency of approximately 80%.

Cancer has become the leading cause of mortality, especially in the 21st century, mainly because it is very difficult to diagnose and treat.
A., Gorczyca M., Chemia leków, PZWL, Warszawa , 1999).Known compounds with the 9,10-anthraquinone skeleton in their structure belong to the most numerous group of antibiotics called anthracyclines or anthraquinone antibiotics. Anthracyclines (doxorubicin, daunorubicin, idarubicin, epirubicin, aclarubicin, mitoxantrone and their synthetic derivatives) can be distinguished among the anthraquinone derivatives used. These compounds are mainly used as drugs in the treatment of acute leukemias, lymphomas and various solid tumors, including breast, lung, thyroid, ovarian cancer and soft tissue cancer (Zejc A., Gorczyca M., Chemia medicin, PZWL, Warsaw, 1999 ).
Antybiotyki antracyklinowe są najliczniejszą grupą antybiotyków cytostatycznych hamujących podział komórek stosowanych najczęściej w terapii przeciwnowotworowej.Anthracycline antibiotics are the most numerous group of cytostatic antibiotics that inhibit cell division and are most often used in cancer therapy.
W znanej literaturze zaproponowano kilka różnych mechanizmów wyjaśniających biologiczne działanie tych antybiotyków. Pierwszy z nich oparty jest na interkalacji, czyli wtłaczaniu chemioterapeutyku pomiędzy dwie sąsiadujące ze sobą pary zasad w DNA.The known literature proposes several different mechanisms explaining the biological action of these antibiotics. The first one is based on intercalation, i.e. the injection of a chemotherapeutic agent between two adjacent base pairs in DNA.
Interkalacja w helisę DNA może spowodować jej wydłużenie lub pęknięcie oraz zmianę właściwości biochemicznych, co w konsekwencji prowadzi do zahamowania zdolności do replikacji i transkrypcji (Gieseler F., Biersack H., Brieden T., Manderscheid J., Nubler V., Annals of Hematology, 1994, 69, 13-17).Intercalation into the DNA helix may cause it to lengthen or break it, and change its biochemical properties, which in turn leads to inhibition of the replication and transcription capacity (Gieseler F., Biersack H., Brieden T., Manderscheid J., Nubler V., Annals of Hematology , 1994, 69, 13-17).
A., Współczesna onkologia, 2003, 7, 45-53).Anthracycline antibiotics are also inhibitors of topoisomerase I and II. Topoisomerases I and II are a group of homodimeric chromosomal enzymes that play an essential role in maintaining the physiological function of DNA. These enzymes catalyze the relaxation process of DNA molecules from chromosomes by causing periodically repeated so-called cracks, breaks in one or two DNA strands, which have an impact on the correct process of replication and transcription (Omyła-Staszewska J., Deptała A., Współczesna oncologia, 2003, 7, 45-53).
Właściwości redoks antybiotyków antracyklinowych mogą doprowadzić do wytworzenia wolnych rodników lub rodników hydroksylowych, które powodują uszkodzenie DNA, co w konsekwencji prowadzi do zablokowania procesu transkrypcji (Frederick C. A., Williams L. D., Ughetto G., Marel G. A., Boom J. H., Rich A., Wang A. H., Biochemistry, 1990, 29, 2538-2549).In addition, these compounds bound to the cell membrane can have a significant impact on the fluidity of cell membranes, ion transport, and also disturb the balance of biochemical processes in the cell. The redox properties of anthracycline antibiotics can lead to the generation of free radicals or hydroxyl radicals that damage DNA, which in turn leads to blocking the transcription process (Frederick CA, Williams LD, Ughetto G., Marel GA, Boom JH, Rich A., Wang AH , Biochemistry, 1990,29,2538-2549).
Znane i opisane rozwiązania środków o aktywności przeciwnowotworowej i sposobów ich otrzymywania wykazują liczne wady. The known and described solutions of agents with antitumor activity and methods of their preparation show numerous disadvantages.
The disadvantage of anthracycline antibiotics is their cardiotoxicity and the development of resistance in neoplastic cells, occurring alongside other side effects. Circulatory disorders are attributed to products formed during the biotransformation of anthracyclines, especially in the reduction of quinone groups to hydroquinone groups. The resulting oxygen radicals and hydroxyl radicals cause the formation of peroxide structures in lipids, and their gradual decomposition leads to compounds damaging the heart muscle.
