Abstract
Przedmiotem zgłoszenia jest emulsja kosmetyczna typu olej/woda, według wynalazku, zawierająca oleje roślinne, składniki nawilżające, przy czym zawiera fazę wodną w ilości 52,2 do 90% wagowych, fazę olejową w ilości 9,5 do 46,8% wagowych, charakteryzuje się tym, że zawiera od 0,1 do 3% wagowych rozdrobnionych suchych tkanek roślin lawendy wąskolistnej namnażanej w kulturach tkankowych in vitro na pożywce MS wzbogaconej o od 0,5 do 10 mg dm-3 nanocząstek srebra o średnicy od 13 do 30 nm. Fazę olejową stanowi lanolina w ilości od 6 do 15% wagowych, wosk pszczeli w ilości 3 do 10% wagowych, masło shea w ilości od 3 do 10% wagowych i olej jojoba w ilości od 20 do 50% wagowych. Fazę wodną stanowi D —pantenol w ilości od 1 do 5% wagowych, gliceryna w ilości od 1 do 5% wagowych oraz woda w ilości od 20 do 40% wagowych. Przemysł kosmetyczny jest jedną z najszybciej rozwijających się gałęzi branży higieny osobistej na świecie. Bezpieczeństwo mikrobiologiczne produktów kosmetycznych to jeden z ważniejszych aspektów branych pod uwagę podczas produkcji kosmetyków. Rozwój drobnoustrojów w produkcie kosmetycznym prowadzi do jego degradacji, a bliski kontakt mikroorganizmów z podrażnioną lub uszkodzoną skórą zagraża zdrowiu konsumenta oraz przyczynia się do rozprzestrzenia i rozwoju różnego rodzaju infekcji i chorób skóry. Skład nowoczesnych preparatów kosmetycznych i obecność w nich wody są idealnym środowiskiem do rozwoju różnego rodzaju mikroorganizmów. Z tych właśnie powodów niemożliwa jest masowa produkcja kosmetyków wolnych od substancji konserwujących. Za sprawą regulacji światowych organizacji zdrowia, lista dozwolonych substancji syntetycznych używanych podczas produkcji kosmetyków znacząco zmniejsza się. Dlatego też, w ostatnich latach wyraźnie wzrasta zainteresowanie kosmetykami naturalnymi, a wielu producentów poszukuje alternatyw dla chemicznych komponentów. Wiele badań naukowych potwierdza szkodliwość chemicznych konserwantów zabezpieczających produkty kosmetyczne. Wiele z nich uznaje się za powód alergii i chorób skórnych. Innym przypisuje się kancerogenność czy mutagenność (Robert i in., 2005). The stage of production of cosmetics and their daily use by the consumer contribute to the occurrence of contamination in them. For these reasons, it is impossible to mass-produce preservative-free cosmetics. Due to the regulations of world health organizations, the list of permitted synthetic substances used in the production of cosmetics is significantly reduced. Therefore, interest in natural cosmetics has been clearly growing in recent years, and many manufacturers are looking for alternatives to chemical components. Many scientific studies confirm the harmfulness of chemical preservatives protecting cosmetic products. Many of them are considered to be the cause of allergies and skin diseases. Others are believed to be carcinogenic or mutagenic (Robert et al., 2005). The widespread use of chemical preservatives in the form of isothiazolinones often leads to the development of eczema and other allergic skin conditions that are difficult to treat (Lorente-Lavirgen et al., 2019).

Consumers want to have access to effective, safe and natural products, the production of which does not have a negative impact on the environment. For these reasons, the cosmetics sector is still looking for new solutions leading to the acquisition and use of extracts and biologically active substances from isolated plants.
Roślinne kultury in vitro dają możliwość szybkiej hodowli roślin leczniczych w kontrolowanych warunkach laboratoryjnych, które są wolne od zanieczyszczeń i niezależne od pór roku. Technika ta zapobiega nadmiernej eksploatacji naturalnych populacji oraz nie wpływa negatywnie na ekosystem, umożliwia produkcję roślin bez stosowania pestycydów, czy herbicydów (Eibl i in., 2018). Wiele z nich jak np. szikonina, skolopamina, kwas rozmarynowy, czy polisacharydy, których produkcja prowadzona jest w roślinnych kulturach in vitro, zostały wprowadzone do produktów kosmetycznych między wczesnymi latami 80-tymi, a późnymi 90-tymi (Sato i Yamada, 1984; Deno i in. 1987; Hess, 1992). Szeroko rozwinięty trend produkcji kosmetyków w naturalny i zrównoważony sposób rozpoczął nową erę w zastosowaniu i popularyzacji technologii roślinnych kultur in vitro. W ciągu ostatnich 10 lat do przemysłu kosmetycznego trafiło ponad 50 produktów opartych na ekstraktach pochodzących z roślinnych kultur in vitro, przy czym większość z nich pozyskiwana jest z kultur zawiesinowych (Eibl i in., 2018). Many of them, such as shiconine, scolopamine, rosemary acid, or polysaccharides produced in in vitro plant cultures, were introduced into cosmetic products between the early 1980s and late 1990s (Sato and Yamada, 1984; Deno et al., 1987; Hess, 1992). The widely developed trend of producing cosmetics in a natural and sustainable way has started a new era in the use and popularization of in vitro plant culture technology. In the last 10 years, more than 50 products based on extracts from plant in vitro cultures have entered the cosmetics industry, with most of them obtained from suspension cultures (Eibl et al., 2018). Ingredients of plant origin play the role of active ingredients in a cosmetic product (e.g. moisturizing substances, sulfactants, substances with a rejuvenating effect). Some of them are described below.

It is an attractive product that contains natural ingredients that are safe for the user. Lavender tissues grown in in vitro cultures can be added to cosmetic emulsions after grinding, because they are made of tissues without large amounts of phloem and lignified parts.

W celu przygotowania medium hodowlanego użyto podstawową pożywkę MS (Murashige i Skoog, 1962) zawierającą składniki mineralne w postaci: azotan amonu (NH3NO3) w ilości 1650 mg/dm3, azotan potasu (KNO3) w ilości 1900 mg/dm3, siarczan magnezu (MgSO4-7H2O) w ilości 370 mg/dm3, diwodorofosforan potasu w ilości 170 mg/dm3 (KH2PO4), chlorek wapnia (CaCl2-2H2O) w ilości 440 mg/dm3, jodek potasu (KJ) w ilości 0,83 mg/dm3, kwas borowy (H3BO3) w ilości 6,2 mg/dm3, siarczan manganu (II) (MnSO4-4H2O) w ilości 22,3 mg/dm3, siarczan cynku (ZnSO4-7H2O) w ilości 8,6 mg/dm3, molibdenian (VI) sodu (Na2MoO4-2H2O) w ilości 0,25 mg/dm3, siarczan żelaza (II) (FeSO4-7H2O) w ilości 27,8 mg/dm3, wersenian disodu (Na2EDTA-2H2O) w ilości 37,3 mg/dm3, siarczan miedzi (II) (CuSO4-5H2O) w ilości 0,025 mg/dm3, chlorek kobaltu (II) (CoCh6H2O) w ilości 0,025 mg/dm3. Dodatkowe składniki organiczne zawarte w pożywce były następujące: niacyna w ilości 0,5 mg/dm3, pirydoksyna HCl w ilości 0,5 mg/dm3, tiamina HCl w ilości 0,1 mg/dm3, glicyna w ilości 2 mg/dm3. Składniki po odważeniu zostały rozpuszczone w wodzie destylowanej. Następnie dodano roślinne regulatory wzrostu takie jak: cytokininę: kinetynę (KIN) w ilości 2 mg-dm-3, auksynę: kwas indolilooctowy (IAA) w ilości 0,2 mg-dm-3 oraz nanocząstki srebra o wielkości cząsteczki 13 nm w ilości 1 mg-dm-3. Odczyn pożywek ustalono na poziomie 5,7 za pomocą stężonego HCl oraz NaOH. Składniki stałe w postaci inozytolu w ilości 100 mg/dm3, sacharozy w ilości 30 g/dm3 i agaru w ilości 8 g/dm3 dodano do wcześniej przygotowanej bazy. Całość podgrzewano w wysokiej temperaturze do momentu zagotowania pożywki. Pożywkę w odmierzonej ilości 30 ml umieszczano w naczyniach szklanych i zakręcano plastikowymi nakrętkami. Naczynia z pożywkami umieszczono w autoklawie i sterylizowano przez 19 minut w temperaturze 121°C przy ciśnieniu 1 atm. (0,1 MPa). Po wysterylizowaniu pożywki naczynia hodowlane wyjęto z autoklawu i pozostawiono do momentu zestalenia pożywki. Na przygotowane pożywki wykładano fragmenty roślin (ekslpantaty). Zabieg ten odbył się w sterylnych warunkach laboratoryjnych przy użyciu komór laminarnych. Kultury roślinne umieszczono w fitotronie w temperaturze 24°C oraz wilgotności względnej powietrza 70-80%. Oświetlane były światłem fluorescencyjnym o natężeniu 40 PAR (μEm2s1) przez 16 h dziennie. Po 4 tygodniach wzrostu rośliny zebrano. Additional organic components contained in the medium were as follows: niacin in the amount of 0.5 mg/ dm3 , pyridoxine HCl in the amount of 0.5 mg/ dm3 , thiamine HCl in the amount of 0.1 mg/ dm3 , glycine in the amount of 2 mg/dm3 dm 3 . After weighing, the ingredients were dissolved in distilled water. Then, plant growth regulators such as: cytokinin: kinetin (KIN) in the amount of 2 mg-dm -3 , auxin: indole acetic acid (IAA) in the amount of 0.2 mg-dm -3 and silver nanoparticles with a particle size of 13 nm in the amount of 1 mg-dm -3 . The pH of the media was set at 5.7 with concentrated HCl and NaOH. Solid components in the form of inositol in the amount of 100 mg/dm 3 , sucrose in the amount of 30 g/dm 3 and agar in the amount of 8 g/dm 3 were added to the previously prepared base. The whole was heated at high temperature until the medium boiled. The medium in a measured amount of 30 ml was placed in glass vessels and closed with plastic caps. Vessels with media were placed in an autoclave and sterilized for 19 minutes at 121°C and 1 atm pressure. (0.1 MPa). After the medium was sterilized, the culture vessels were removed from the autoclave and left until the medium solidified. Plant fragments (expandates) were placed on the prepared media. The procedure was carried out in sterile laboratory conditions using laminar chambers. Plant cultures were placed in a phytotron at a temperature of 24°C and relative air humidity of 70-80%. In creams to which plant material obtained from plants grown on media enriched with silver nanoparticles with a particle size of 30 nm in the amount of 10 mg/dm 3 was added, bacterial colonies appeared after 6 weeks of storage, and fungal colonies only after 8 weeks of storage. The number of bacterial colonies was only 4.9 and fungal colonies 1.1.
