Description

Opis wynalazkuDescription of the invention

Przedmiotem niniejszego wynalazku jest sposób otrzymywania 10,10'-di(mezytylo)-9,9'-bisantracenu (inaczej: 10,10'-bis(2,4,6-trimetylofenylo)-9,9'-bisantracenu) w reakcji sprzęgania 10,10'-dibromo-9,9'-bisantracenu z kwasem mezytyloboronowym, w obecności katalizatora palladowego (palladowego układu katalitycznego). The subject of the present invention is a method for obtaining 10,10'-di(mesityl)-9,9'-bisanthracene (also: 10,10'-bis(2,4,6-trimethylphenyl)-9,9'-bisanthracene) in the reaction coupling of 10,10'-dibromo-9,9'-bisanthracene with mesitylboronic acid, in the presence of a palladium catalyst (palladium catalytic system).

W literaturze opisano zaledwie kilka pochodnych bisantenu z podstawnikami w pozycjach 7 i 14, to jest fenylami, 3,5-di-tert-butylofenylami, p-trifluorometylofenylami, grupami -C=C-Si(i-pr)3 [J. Li, K. Zhang, X. Zhang, K-W. Huang, C. Chi, J. Wu, meso- Substituted Bisanthenes as Soluble and Stable Near-infrared Dyes, J. Org. Chem., 2010, 75(3), 856-863]. Jednakże najważniejszą pochodną bisantenu jest pochodna z grupami mezytylowymi - szczególnie w kontekście dalszej pi-ekspansji układu bazowego. The compound subject to the invention is described in the literature as a substrate for the synthesis of 7,14-di(mesityl)bisanthene. Bisantene belongs to polyaromatic compounds, important due to their photophysical properties and, above all, because its derivatives are precursors of many nanographenes. However, due to its partially diradical character, unsubstituted bisanthene is unstable and is easily subject to oxidation and other undesirable transformations. To be useful as a building block in the synthesis of nanographenes, it requires the presence of sterically extended substituents at the 7 and 14 positions. Only a few bisanthene derivatives with substituents at the 7 and 14 positions have been described in the literature, i.e. phenyls, 3,5-di-tert-butylphenyls, p -trifluoromethylphenyls, -C=C-Si(i-pr)3 groups [J. Li, K. Zhang, X. Zhang, K.-W. Huang, C. Chi, J. Wu, Meso-Substituted Bisanthenes as Soluble and Stable Near-infrared Dyes, J. Org. Chem., 2010, 75(3), 856-863]. However, the most important bisanthene derivative is the derivative with mesityl groups - especially in the context of further pi-expansion of the base system.

Wszystkie, znane reakcje cykloaddycji do wnęk bisantenu opisano w pracy przeglądowej [A. Kurpanik, M. Matussek, P. Lodowski, G. Szafraniec-Gorol, M. Krompiec, S. Krompiec, Diels-Alder cycloaddition to bay region of perylene and its derivatives as an attractive APEX strategy for PAHs’ core expansion: theoretical and practical aspects, Molecules, 2020, 25(22), 5373]. Z kolei, gdy chodzi o znane metody syntezy 10,10'-di(mezytylo)-9,9'-bisantracenu to jest to skomplikowana i mało efektywna droga od antronu, poprzez bis-antron, di-mezytylo-bis-antracenodiol do oczekiwanego produktu [E. H. Fort, P. M. Donovan, L. T. Scott, DielsAlder Reactivity of Polycyclic Aromatic Hydrocarbon Bay Regions: Implications for Metal-Free Growth of Single-Chirality Carbon Nanotubes, J. Am. Chem. Soc., 2009, 131(44), 16006-16007; A. Konishi, Y. Hirao, K. Matsumoto, H. Kurata, T. Kubo, Facile Synthesis and Lateral n-Expansion of Bisanthenes, Chem. Fett., 2013,42(6), 592-594].As mentioned, after introducing sterically expanded substituents into the bisanthene structure, it is possible to use this compound for further development (expansion) of the pi-electron system - by Diels-Alder cycloaddition to one or both cavities. All known cycloaddition reactions into bisanthene cavities are described in the review [A. Kurpanik, M. Matussek, P. Lodowski, G. Szafraniec-Gorol, M. Krompiec, S. Krompiec, Diels-Alder cycloaddition to bay region of perylene and its derivatives as an attractive APEX strategy for PAHs' core expansion: theoretical and practical aspects, Molecules, 2020, 25(22), 5373]. In turn, when it comes to known methods of synthesizing 10,10'-di(mesityl)-9,9'-bisanthracene, it is a complicated and ineffective path from anthrone, through bis-anthron, di-mesityl-bis-anthracenediol to the expected product [E. H. Fort, P. M. Donovan, L. T. Scott, DielsAlder Reactivity of Polycyclic Aromatic Hydrocarbon Bay Regions: Implications for Metal-Free Growth of Single-Chirality Carbon Nanotubes, J. Am. Chem. Soc., 2009, 131(44), 16006-16007; A. Konishi, Y. Hirao, K. Matsumoto, H. Kurata, T. Kubo, Facile Synthesis and Lateral n-Expansion of Bisanthenes, Chem. Fett., 2013,42(6), 592-594].

Znane ze stanu techniki jest jedynie sprzęganie 10,10'-dibromo-9,9'-bisantracenu z kwasem fenyloboronowym prowadzące do pochodnej difenylowej [H. S. Jang, K. H. Lee, S. J. Fee, Y. K. Kim, S. S. Yoon, Blue Organic Light-Emitting Diodes Containing Anthracene Derivatives With End-Capping Phenyl Group, Mol. Cryst. Liq. Cryst., 2012, 563(1), 173-184]. Sprawdzono to w trakcie opracowywania niniejszego rozwiązania syntetycznego. The aim of the creators of this invention was to develop a one-step, efficient method for obtaining 10,10'-di(mesityl)-9,9'-bisanthracene from commercially available - 10,10'-dibromo-9,9'-bisanthracene and 2,4 acid, 6-trimethylphenylboronic acid (mesitylboronic acid). This type of method is not known from the prior art. Only the coupling of 10,10'-dibromo-9,9'-bisanthracene with phenylboronic acid leading to a diphenyl derivative is known from the state of the art [H. S. Jang, K. H. Lee, S. J. Fee, Y. K. Kim, S. S. Yoon, Blue Organic Light-Emitting Diodes Containing Anthracene Derivatives With End-Capping Phenyl Group, Mol. Cryst. Liq. Cryst., 2012, 563(1), 173-184]. However, the palladium catalyst used in this reaction, i.e. [Pd(PPhs)4], turned out to be practically completely ineffective in the reaction of the present invention. This was verified during the development of this synthetic solution. This is certainly due to steric hindrance that occurs during the reaction when the reactant is 2,4,6-trimethylphenylboronic acid (mesitylboronic acid) instead of phenylboronic acid. Despite a number of attempts, including increasing the reaction temperature and several times increasing the concentration of the catalytic system based on [Pd(PPhs)4], it was not possible to achieve a conversion greater than 5%.

Li, Y. Yamamoto, N. Miyaura, Synthesis of Tetra-ortho-Substituted Biaryls Using Aryltriolborates, Synlett, 2011,2011(12), 1769-1773].The coupling of simple 9-bromoanthracene leading to 9-(mesityl)anthracene is also known [patent description US2018/155375; G-Q. Li, Y. Yamamoto, N. Miyaura, Synthesis of Tetra-ortho-Substituted Biaryls Using Aryltriolborates, Synlett, 2011,2011(12), 1769-1773].

Wynika to z kilku powodów. Po pierwsze obecność dwóch atomów bromu może prowadzić do niepożądanego debromowania, a obecność podstawnika antracenylowego naprzeciwko podstawianego atomu bromu zdecydowanie utrudnia reakcję. Eksperymenty prowadzące do opracowania metody według niniejszego wynalazku potwierdziły powyższe obawy i trudności. Do generowania - in situ - rzeczywistego katalizatora testowano różne fosfiny, w tym trifenylofosfinę, trifenoksyfosfinę, dppf (difenylofosfinoferrocen), 2-dicykloheksylofosfino-2',6'-dimetoksybifenylu (SPhos), 2-dicykloheksylofosfino-2',4',6'-triizopropylbifenylu (XPhos), 4-(Antracen-9-yl)-3-(t-butyl-2,3-dihydrobenzo[d][1,3]oksafosfolu (AntPhos). Ponadto, kluczowymi parametrami gdy chodzi o wydajność i czystość produktu okazały się być: bezwodna i beztlenowa atmosfera, proporcje molowe 10,10'-dibromo-9,9'-bisantracen/kwas mezytyloboronowy oraz prekursor palladowy/fosfina, temperatura i czas reakcji. However, the synthesis according to the present invention is much more demanding and cannot be considered a simple analogy of the coupling leading to 9-mesityanthracene. This is due to several reasons. First, the presence of two bromine atoms may lead to undesirable debromination, and the presence of an anthracenyl substituent opposite the substituted bromine atom significantly complicates the reaction. Experiments leading to the development of the method of the present invention confirmed the above concerns and difficulties. However, after performing a number of experiments, an effective procedure was developed that allowed obtaining 10,10'-di(mesityl)-9,9'-bisanthracene in one step, in the coupling reaction of 10,10'-dibromo-9,9'-bisanthracene with commercially available mesitylboronic acid with high efficiency, over 90%. The most important thing turned out to be the use of an appropriate catalytic system, i.e. an appropriate palladium precursor and an appropriate phosphine. A number of simple palladium(II) compounds were tested, including palladium(II) acetate, palladium(II) chloride, and palladium(II) acetylacetonate. Various phosphines were tested for generating - in situ - the actual catalyst, including triphenylphosphine, triphenoxyphosphine, dppf (diphenylphosphinoferrocene), 2-dicyclohexylphosphine-2',6'-dimethoxybiphenyl (SPhos), 2-dicyclohexylphosphine-2',4',6' -triisopropylbiphenyl (XPhos), 4-(Anthracen-9-yl)-3-(t-butyl-2,3-dihydrobenzo[d][1,3]oxaphosphole (AntPhos). Moreover, the key parameters when it comes to efficiency and the purity of the product turned out to be: anhydrous and oxygen-free atmosphere, molar proportions of 10,10'-dibromo-9,9'-bisanthracene/mesitylboronic acid and palladium precursor/phosphine, temperature and reaction time.

It should be emphasized that the synthesis method according to the invention is simple (based on commercially available substrates) and very effective - over 90% efficiency is achieved. As mentioned earlier, commenting on the state of the art, the only known method of synthesizing 10,10'-di(mesityl)-9,9'-bisanthracene is a complicated and ineffective route from anthrone, through bis-anthrone, di-mesityl-bis-anthracenediol to expected product.

Sposób otrzymywania 10,10'-di(mezytylo)-9,9'-bisantracenu według wynalazku zostanie bliżej objaśniony na podstawie poniższych przykładów oraz na schemacie ogólnym reakcji (schemat 1), gdzie [Pd] = katalizator palladowy generowany in situ z prekursora i liganda fosfinowego; M = K lub Na.The method of obtaining 10,10'-di(mesityl)-9,9'-bisanthracene according to the invention will be explained in more detail on the basis of the examples below and in the general reaction scheme (Scheme 1), where [Pd] = palladium catalyst generated in situ from the precursor and phosphine ligand; M = K or Na.

W naczyniu Schlenka o pojemności 100 ml umieszczono 1,0 mmol 10,10'-dibromo-9,9'-bisantracenu, 4,0 mmol kwasu 2,4,6-trimetylofenyloboronowego, 5,0 mmol K3PO4, 0,1 mmol prekursora katalizatora w postaci octanu palladu(II) - [Pd(OAc)2], 0,3 mmol liganda fosfinowego w postaci dialkilobiarylo fosfiny tj. 2-dicykloheksylfosfino-2',6'-dimetoksybifenylu (SPhos), oraz 0,1 g wysuszonych sit molekularnych 4A. Dane spektroskopowe otrzymanego produktu były zgodne z literaturowymi [E. H. Fort, P. M. Donovan, L. T. Scott, Diels-Alder Reactivity of Polycyclic Aromatic Hydrocarbon Bay Regions: Implications for Metal-Free Growth of Single-Chirality Carbon Nanotubes, J. Am. Chem. The Schlenk vessel and its contents were then dried under vacuum for 2 hours at 60°C. After this time, 60 ml of anhydrous toluene were added under an argon atmosphere and the mixture was heated in a closed system at 110°C in a strictly oxygen-free and anhydrous atmosphere for 24 hours. After this time, the mixture was cooled to 22°C and the solid residues were filtered off on a celite funnel, and then the volatile fractions were evaporated on a vacuum evaporator. The product was isolated from the solid residue by silica gel column chromatography using a 10:1 volume ratio of toluene:ethyl acetate as eluent. The obtained crude product was purified by crystallization from a mixture of hexane/methylene chloride (v/v = 1/1) to finally obtain 0.92 mmol of 10,10'-di(mesityl)-9,9'-bisanthracene with a purity of greater than 97% ( NMR), which is 92% of the theoretical yield in relation to the starting 10,10'-dibromo-9,9'-bisanthracene. Spectroscopic data of the obtained product were consistent with those in the literature [E. H. Fort, P. M. Donovan, L. T.
