Abstract
Przedmiotem zgłoszenia jest hybryda porfirynowo-cukrowa o wzorze ogólnym I zawierająca ugrupowanie porfirynowe i 2-etynyloglikalowe. Przedmiotem zgłoszenia jest również sposób syntezy hybrydy porfirynowo-cukrowej o wzorze ogólnym I obejmujący a) reakcję pochodnej 2-etynyloglikalowej z porfiryną w mieszaninie rozpuszczalnika organicznego i trietyloaminy, w obecności źródła jonów fluorkowych i katalizatora palladowego oraz ligandu fosfinowego z otrzymaniem związku o wzorze ogólnym II; b) reakcję związku o wzorze ogólnym II otrzymanego w etapie a) z węglanem metalu prowadzoną w mieszaninie rozpuszczalników z otrzymaniem hybrydy porfirynowo-cukrowej o wzorze ogólnym I.The subject of the application is a porphyrin-sugar hybrid of the general formula I containing a porphyrin and 2-ethynylglycal moiety. Pochodne porfiryn są wykorzystywane jako fotouczulacze w nowoczesnej terapii przeciwnowotworowej, tj. terapii fotodynamicznej (PDT) (N. Tsolekile, et al., Molecules, 2019, 24, 2669; J. Kou, et al., Oncotarget, 2017, 8, 81591; A. E. O'Connor, et al., Photochem Photobiol., 2009, 85, 1053). Zdecydowana większość znanych porfiryn cechuje się ograniczoną rozpuszczalnością w polarnych rozpuszczalnikach, co koreluje z ich znikomą rozpuszczalnością w płynach ustrojowych. Wymienione właściwości powodują trudność z usuwaniem fotouczulacza przez organizm; zatem po zakońc zeniu terapii pojawiają się liczne skutki uboczne w przypadku kontaktu pacjenta ze światłem. Jednym ze sposobów funkcjonalizacji porfiryn w kierunku zwiększenia ich polarności i eliminacji niepożądanych efektów terapii, może być wbudowanie w strukturę fotouczulacza fragmentu polarnej cząsteczki, np. pochodnych cukrów (S. Singh, et al., Chem. Rev., 2015, 115, 10261). The vast majority of known porphyrins are characterized by limited solubility in polar solvents, which correlates with their negligible solubility in body fluids. The mentioned properties make it difficult for the body to remove the photosensitizer; therefore, after completing the therapy, numerous side effects occur when the patient comes into contact with light. One way to functionalize porphyrins to increase their polarity and eliminate undesirable effects of therapy may be to incorporate a fragment of a polar molecule, e.g. sugar derivatives, into the structure of the photosensitizer (S. Singh, et al., Chem. Rev., 2015, 115, 10261). . Several such combinations have been described in the literature, but their number is still unsatisfactory and this group should be expanded to include new compounds characterized by good bioavailability resulting from, among others, due to their good solubility in polar solvents.

Na wydajność reakcji ma także wpływ rodzaj rozpuszczalnika, co doskonale widać w przykładach wykonania, w których zastosowano takie rozpuszczalniki jak 1,4-dioksan, toluen, t-BuOH i dimetyloformamid (tabela 1). Kolejnym elementem był uważny dobór katalizatora oraz ligandu (tabela 2). Reagents in the form of glycals functionalized with an ethynyl moiety on the second carbon atom and appropriate bromosubstituted porphyrins, such as, for example, 5,10,15,20-tetrakis (4-bromophenyl)porphyrin, 5,10,15,20-tetrakis-(3-bromophenyl) )porphyrin etc. were selected to prevent the formation of byproducts while obtaining tetrasubstituted porphyrin–sugar hybrids in good yields. Research on the reaction conditions has shown that it is crucial to use equal volumes of solvents in step a), i.e. triethylamine and an appropriate organic solvent. The type of solvent also affects the reaction efficiency, which is clearly visible in the examples where solvents such as 1,4-dioxane, toluene, t-BuOH and dimethylformamide were used (Table 1). The next element was the careful selection of the catalyst and ligand (Table 2). In order to select appropriate conditions, various combinations of catalysts selected from the group including: Pd(PPh3)2Cl2, PdCl2, Pd(OAc)2, Pd-XantPhos-G3, Pd-RuPhosPL 244591 Β1 were tested
-G3, Pd-XPhos-G3 oraz ligandów wybranych z grupy PPh3, XantPhos, RuPhos, XPhos. Najlepszą wydajność uzyskano dla kombinacji Pd-XPhos-G3 i XPhos. Do reaktora dodano 5,10,15,20-tetrak/s(4-bromofenylo)porfirynę (22,0 mg, 0,024 mmol), 3,4,6tri-O-acetylo-2-(2-trimetylosililoetynylo)-D-glukal (69,8 mg, 0,189 mmol, 8,0 eq), Bu4N+F'-3H2O (61,2 mg, 0,194 mmol, 8,2 eq), XPhos (1,7 mg, 0,004 mmol, 0,15 eq), Pd-XPhos-G3 (3,0 mg, 0,004 mmol, 0,15 eq), 1,4-dioksan (2,5 ml) oraz trietyloaminę (2,5 ml). Reaktor szczelnie zamknięto i zabezpieczono przed dostępem światła za pomocą folii aluminiowej. Reakcję mieszano w atmosferze argonu przez 18 godzin w temperaturze 90°C. Następnie mieszaninę poreakcyjną zatężono i produkt oczyszczono chromatograficznie (chlorek metylenu —> chlorek metylenu/octan etylu/trietyloamina 8:2:1 obj./obj./obj.). Otrzymano 5,10,15,20- fefrak/s(4-(3,4,6-tri-O-acetylo-2-D-glukal-2-yloetynylo)fenylo)porfirynę w postaci ciemnofioletowego, krystalicznego ciała stałego (36,1 mg, 85%).5,10,15,20-tetrak/s(4-bromophenyl)porphyrin (22.0 mg, 0.024 mmol), 3,4,6tri-O-acetyl-2-(2-trimethylsilylethynyl)-D- glucal (69.8 mg, 0.189 mmol, 8.0 eq), Bu4N + F'-3H2O (61.2 mg, 0.194 mmol, 8.2 eq), XPhos (1.7 mg, 0.004 mmol, 0.15 eq), Pd-XPhos-G3 (3.0 mg, 0.004 mmol, 0.15 eq), 1,4-dioxane (2.5 ml) and triethylamine (2.5 ml). The reactor was tightly closed and protected from light using aluminum foil. The reaction was stirred under argon for 18 hours at 90°C. The reaction mixture was then concentrated and the product was purified by chromatography (methylene chloride -> methylene chloride/ethyl acetate/triethylamine 8:2:1 v/v/v). 5,10,15,20- fefrac/s(4-(3,4,6-tri-O-acetyl-2-D-glucal-2-ylethynyl)phenyl)porphyrin was obtained in the form of a dark purple crystalline solid (36 .1 mg, 85%).

Do reaktora dodano 5,10,15,20-tetrakis(4-(3,4,6-tri-O-acetylo-2-D-glukal-2-yloetynylo)fenylo)porfirynę (26,4 mg, 0,015 mmol), węglan potasu (24,4 mg, 0,118 mmol, 8,0 eq), metanol (0,5 ml) oraz chlorek metylenu (0,5 ml). Reaktor szczelnie zamknięto. Całość mieszano w temperaturze 40°C przez 3 godziny. Po tym czasie mieszaninę zatężono. Produkt oczyszczono chromatograficznie (chlorek metylenu —> chlorek metylenu/metanol 7:3 (obj./obj.) ^metanol). Otrzymano 5,10,15,20-fefrak/s(4-(2-D-glukal-2-yloetynylo)fenylo)porfirynę w postaci fioletowego proszku (16,2 mg, 86%).5,10,15,20-tetrakis(4-(3,4,6-tri-O-acetyl-2-D-glucal-2-ylethynyl)phenyl)porphyrin (26.4 mg, 0.015 mmol) was added to the reactor , potassium carbonate (24.4 mg, 0.118 mmol, 8.0 eq), methanol (0.5 ml) and methylene chloride (0.5 ml). The reactor was tightly closed. The whole was stirred at 40°C for 3 hours. After this time, the mixture was concentrated. The product was purified by chromatography (methylene chloride -> methylene chloride/methanol 7:3 (v/v) ^methanol). 5,10,15,20-fefrac/s(4-(2-D-glucal-2-ylethynyl)phenyl)porphyrin was obtained in the form of a purple powder (16.2 mg, 86%).
