Description
Opis wynalazkuDescription of the invention
The subject of the invention is a liposome preparation containing zinc and copper, a preparation in the form of liposome powder, a hard capsule containing the same, and a method of producing the same.
Niedobór miedzi w organizmie obniża wchłanianie żelaza, co z kolei prowadzi do niedokrwistości, a niedobór cynku powoduje zaburzenia układu immunologicznego, rozrodczego i nerwowego [M. Kraus i in., Farmakoterapia, vol. 25, nr 05’15 (288)]. Microelements, including zinc and copper, are elements that occur in the human body in trace amounts, but are essential for its proper functioning. Microelement deficiency can lead to many disorders and diseases. Copper deficiency in the body reduces iron absorption, which in turn leads to anemia, and zinc deficiency causes disorders of the immune, reproductive and nervous systems [M. Kraus et al., Farmakoterapia, vol. 25, no. 05'15 (288)]. Therefore, sometimes it is necessary to supplement microelements, either in the form of a single microelement preparation or in the form of preparations containing many microelements or combinations of microelements and vitamins. Various forms of microelement preparations are known. Publication P.426183 describes a dietary supplement in the form of powder, tablets or capsules containing magnesium, chromium, zinc, iron and copper. Publication P.403126 discloses a method for obtaining a zinc or copper supplement in a liquid by dissolving their salts in an alcohol solution or a mixture of alcohol and water. Publication P.425971 discloses a method for producing a zinc-containing microelement supplement in the form of nanoparticles placed in capsules or suspended in water, physiological saline or ethanol.
Encycl Life Sci, 2010, 1-8]. Pierwsze doniesienia o wykorzystaniu zawierającego liposomy preparatu farmaceutycznego do wstrzyknięć można znaleźć w publikacji patentowej GB417715 zgłoszenia dokonanego 29 grudnia 1932. W publikacji tej wskazano, że preparaty farmaceutyczne, zawierające lecytynę, cholesterol i wodę, mogą być wykorzystane do podawania dowolnej dawki leku przy kontrolowanym jej uwalnianiu (preparat typu „depot”).Liposomes are synthetic vesicles consisting of one or more phospholipid bilayers, used as a delivery system for drugs, genes and vaccines. Depending on the composition of the phospholipid layer and the method of their production, it is possible to control the size and charge of liposomes and their stability, which translates directly into the release of active substances contained in them [G. Gregoriadis, Y. Perrie, Liposomes. Encycl Life Sci, 2010, 1-8]. Additionally, they are an ideal carrier for both water-soluble and insoluble substances. Therefore, they are a particularly desirable form of drug delivery. The first reports on the use of a pharmaceutical preparation containing liposomes for injection can be found in patent publication GB417715, an application filed on December 29, 1932. This publication indicates that pharmaceutical preparations containing lecithin, cholesterol and water can be used to administer any dose of a drug with controlled release (a "depot" preparation).
2009 6(1):71-89].However, liposome technologies are now much more advanced. For example, liposomes can be used to actively target active substances to the appropriate site of action, thereby improving specificity and reducing toxicity, by attaching appropriate ligands to their surface or by modifying the composition of the phospholipid bilayer. Liposomal preparations in the form of powders are also known, which are used to administer active substances by inhalation [A. Misra, Expert Opin Drug Deliv. 2009 6(1):71-89].
The disadvantage of liquid preparations and emulsions containing liposomes is their instability in the acidic environment of the stomach, where they aggregate or even precipitate. As a result, the bioavailability of active substances contained in aggregated liposome conglomerates is significantly reduced. The liquid form of liposome preparations also poses an additional technological problem, because it requires the use of special sachets or bottles filled with nitrogen, which must be stored in a refrigerator after opening and their contents consumed within 3 months.
Twórcy niniejszego wynalazku opracowali preparat liposomowy kombinacji cynku i miedzi w postaci proszku, który dzięki liposomowej strukturze znacząco poprawia dostępność tych mikroelementów z przewodu pokarmowego. In the prior art, there was a need for a pharmaceutical preparation in the form of a powder containing zinc and copper encapsulated in liposomes, which preparation could be administered orally. The present inventors have developed a liposome preparation of a combination of zinc and copper in the form of a powder, which, thanks to its liposome structure, significantly improves the availability of these microelements from the gastrointestinal tract. Furthermore, the liposome preparation according to the invention is stable under gastric acid conditions. Moreover, it is also significant that liposome preparations of zinc and copper in the form of a powder are obtained according to the invention, in which the content of microelements is suitably high, i.e. the liposomes are loaded with microelements in an effective manner, which is reflected in the weight ratio of the microelements present in the liposomes to the lipid envelope.
The subject of the invention is a liposome preparation of a combination of zinc and copper in the form of a powder, in which the lipid bilayer of the liposomes contains lecithin, which preparation contains inulin as a stabilizer, wherein the weight ratio of the combination of zinc salt and copper salt to lecithin is 1:100, and preferably 1:1, while the weight ratio of lecithin to inulin is in the range of 1:0.1 to 1:5, and preferably 1:0.89. An advantage of the solution according to the invention is that the presence of inulin stabilizes the liposome preparation and prevents the formation of liposome aggregates, especially in conditions corresponding to gastric acid conditions.
In a preferred embodiment, the zinc in the preparation according to the invention is in the form of a salt selected from the group consisting of zinc acetate, zinc L-ascorbate, zinc L-aspartate, zinc bisglycinate, zinc chloride, zinc citrate, zinc gluconate, zinc lactate, zinc L-lysinate, zinc malate, zinc mono-L-methionine sulfate, zinc oxide, zinc carbonate, zinc L-pidolate, zinc picolinate, zinc sulfate, or a mixture thereof, and most preferably the salt is zinc gluconate. In a further preferred embodiment, the copper in the preparation according to the invention is in the form of a salt selected from the group consisting of copper (II) carbonate, copper (II) citrate, copper (II) gluconate, copper (II) sulfate, copper L-aspartate, copper bisglycinate, copper-lysine complex, copper (II) oxide or a mixture thereof, and the most preferred salt is copper (II) gluconate. Also preferably, the lecithin in the preparation according to the invention is selected from the group consisting of sunflower lecithin, soy lecithin, rapeseed lecithin and egg yolk lecithin, and the lecithin is most preferably sunflower lecithin.
In a preferred embodiment, the liposomal zinc and copper preparation according to the invention is supplied in an acid-resistant hard capsule (hard enteric capsule), whereby the liposomes are released only in the intestine, where the absorption of the microelements takes place. Thus, the subject of the invention is a hard enteric gel capsule containing the liposomal zinc and copper preparation according to the invention. When zinc and copper in liposomal form are in an acid-resistant capsule, their delivery to the body of a person takes place in the most effective manner, because these microelements are released in the digestive tract only near the place where they are absorbed. Thanks to this, the liposomes are not subjected to the unfavorable effects of the acidic environment of the stomach, which additionally prevents their aggregation into larger conglomerates. In a preferred embodiment, the hard capsule is a gelatin, cellulose, pullulan or gellan capsule, and most preferably is an enteric-coated capsule made from a combination of hydroxypropylmethylcellulose and gellan gum.
The subject of the invention is also a method for obtaining a liposome preparation of zinc and copper according to the invention. Preferably, in the method according to the invention, the mixture formed after dissolving inulin in the liposomal suspension of zinc and copper salt contains 5.10% by weight of zinc salt, 0.44% by weight of copper salt, 5.60% by weight of lecithin, 4.96% by weight of inulin and 83.90% by weight of water, based on the total weight of the mixture.
Należy również nadmienić, że dzięki wykorzystaniu sposobu według wynalazku możliwe jest bardzo efektywne ładowanie liposomów, tj. stosunek wagowy substancji czynnej, którą stanowi kombinacja cynku i miedzi, do tworzącej otoczkę lipidową lecytyny odpowiada stosunkowi wagowemu mieszaniny, w której formowane są zawierające te mikroelementy liposomy. Co więcej, sposób według wynalazku umożliwia zamknięcie stosunkowo dużej ilości mikroelementów w liposomowym preparacie, nawet w stosunku kombinacji soli cynku i soli miedzi do lecytyny wynoszącym 1:1.It should also be noted that by using the method according to the invention, it is possible to load liposomes very effectively, i.e. the weight ratio of the active substance, which is a combination of zinc and copper, to the lecithin forming the lipid envelope corresponds to the weight ratio of the mixture in which liposomes containing these microelements are formed. Moreover, the method according to the invention enables the encapsulation of a relatively large amount of microelements in a liposome preparation, even in a ratio of the combination of zinc salt and copper salt to lecithin of 1:1.
Przykład 1. Sposób wytwarzania preparatu cynku i miedzi w postaci proszku liposomów (suszenie rozpyłowe)Example 1. Method of producing a zinc and copper preparation in the form of liposome powder (spray drying)
Liposomes containing a combination of zinc and copper are obtained as follows. 51.25 kg of sunflower lecithin SF100 are dissolved in 136.8 kg of purified water at a temperature of 70°C +/- 5°C, and a combination of zinc gluconate and copper gluconate in a total amount of 50.53 kg are dissolved in 476.0 kg of purified water at a temperature of 45°C +/- 5°C. Then, the above-described aqueous solutions of sunflower lecithin and the combination of zinc gluconate and copper gluconate are combined with each other under intensive stirring, the temperature being maintained at 45°C +/- 5°C during stirring, and the suspension thus obtained is homogenized in portions of 20 I for 5 minutes. Independently, at a temperature of 45°C +/- 5°C, 45.3 kg of inulin is dissolved in 152.93 kg of purified water, and the aqueous solution thus obtained is added with intensive mixing to the suspension resulting from the combination of aqueous solutions of sunflower lecithin and a combination of zinc gluconate and copper gluconate. After mixing, the liposome suspension is spray-dried at a temperature of 175°C +/- 5°C to obtain liposome powder.
Przykład 2. Sposób wytwarzania preparatu cynku i miedzi w postaci proszku liposomów (liofilizacja)Example 2. Method of producing a zinc and copper preparation in the form of liposome powder (lyophilization)
The zinc and copper preparation in the form of liposome powder is obtained as described in Example 1, with the difference that after mixing, the liposome suspension is dried by lyophilization in a shelf lyophilizer at a temperature of -50°C+/-5°C.
Przykład 3. Sposób wytwarzania preparatu cynku i miedzi w postaci liposomów - ocena efektywności enkapsulacjiExample 3. Method of producing a zinc and copper preparation in the form of liposomes - evaluation of encapsulation efficiency
W ten sposób otrzymano przesącz bez liposomów, w którym określano ilość kombinacji glukonianu cynku i miedzi. Pozostałości gulkonianiu cynku i glukonianu miedzi w przesączu były śladowe, co oznacza dużą efektywność enkapsulacji tych mikroelementów w liposomach.Different batches of zinc and copper preparation (zinc gluconate plus copper gluconate) in the form of liposome powder were prepared as described in Example 1, except that the amounts of reagents and the process temperatures were shown in Table 1 below. After the liposomal preparation was formed, samples were taken from each batch. From each sample batch, liposomes were separated on a polycarbonate filter with a pore diameter of 80 nm. In this way, a liposome-free filtrate was obtained, in which the amount of the combination of zinc gluconate and copper was determined. The residues of zinc gulconate and copper gluconate in the filtrate were trace, which indicates a high efficiency of encapsulation of these microelements in liposomes.

Przykład 4. Analiza wielkości liposomów w preparatach według wynalazkuExample 4. Analysis of liposome size in preparations according to the invention
Pomiar wielkości liposomów otrzymanych w Przykładzie 1 i 2 wykonano z wykorzystaniem urządzenia NanoSizerZS (Malvern Instruments, Wielka Brytania) w funkcji „volume”. 10 μΙ próbki liposomów (po dokładnym wymieszaniu preparatów z Przykładu 1 i 2) rozcieńczano wodą dejonizowaną do objętości 1000 μΙ i umieszczano w kuwecie przyrządu. The size measurement of liposomes obtained in Examples 1 and 2 was performed using the NanoSizerZS device (Malvern Instruments, UK) in the "volume" function. 10 μΙ liposome samples (after thorough mixing of the preparations from Examples 1 and 2) were diluted with deionized water to a volume of 1000 μΙ and placed in the instrument's cuvette. At least three independent measurements of the size distribution were then recorded. A mixture of 140.1 nm liposomes for a volume of 62% and 742.3 nm for a volume of 38% was obtained.
In order to determine the stability of liposomes in the preparation according to the invention, in particular their resistance to aggregation under strong acid conditions, the size of liposomes obtained in Examples 1 and 2 was measured in a manner analogous to that described above, however after prior incubation of the liposomes for two hours in 0.1 M HCl acid. The measurement resulted in the values obtained for liposomes not incubated in 0.1 M HCl, which confirms the stability of the liposomes.
Przykład 5. Badanie stabilności liposomowego preparatu cynku i miedzi w postaci proszku według wynalazkuExample 5. Stability study of the liposomal zinc and copper powder preparation according to the invention
Próbki przechowywano w temperaturze 25°C, a ich stabilność oceniano na podstawie wyglądu, zapachu i pomiaru pH.The stability study of the liposomal zinc and copper preparation obtained in Example 1 was carried out for 3 months, with the condition of preserved samples of the preparation being monitored every month. The samples were stored at 25°C, and their stability was assessed based on appearance, odor, and pH measurement.
After three months, the appearance, smell and pH of the samples did not change. The test results therefore indicate full stability of the samples in the tested range.
