Abstract
Przedmiotem zgłoszenia jest sposób otrzymywania ekstraktu z wrośniaka różnobarwnego (Trametes versicolor) bogatego w białka i peptydy, charakteryzuje się tym, że przed ekstrakcją owocniki przygotowuje się w ten sposób, że oczyszczone i podzielone na mniejsze fragmenty mrozi się w temperaturze -20°C do -80°C, po czym zamrożony materiał zalewa się ciekłym azotem korzystnie trzykrotnie i rozciera się na proszek lub alternatywnie poddaje się liofilizacji w temperaturze od -50°C do -40°C, korzystnie -45°C przy ciśnieniu wynoszącym od 4 do 6 Pa, optymalnie 5 Pa, aż do uzyskania stałej masy liofilizatu, następnie zliofilizowane lub rozdrobnione (roztarte) owocniki poddaje się ekstrakcji zalewając wodą w proporcjach w:w od 1:15 do 1:30, korzystnie 1:20, proces prowadzi się w temperaturze od 50°C do 80°C przez 1,5 do 2-ch godzin, po czym chłodzi do temperatury 30°C, odwirowuje supernatant i poddaje koncentracji, korzystnie do uzyskania dziesięciokrotnie zatężonego ekstraktu. The application also covers an extract of the variegated tarsi (Trametes versicolor), rich in proteins and peptides, for use in supporting the prevention and treatment of diseases caused by staphylococci and streptococci.

Przedmiot wynalazku stanowi sposób otrzymywania wodnego ekstraktu z wrośniaka różnobarwnego (Trametes versicolor) bogatego w białko oraz jego hydrolizatu, w którym zastosowano ciekły azot lub proces liofilizacji i dobór optymalnej temperatury ekstrakcji na etapie przygotowywania surowca do rozdrobnienia oraz ekstrakcji białek endogennych oraz zastosowanie wodnego ekstraktu z wrośniaka różnobarwnego (Trametes versicolor) oraz jego hydrolizatu do wspomagania profilaktyki i leczenia chorób wywołanych przez gronkowce i paciorkowce.The subject of the invention is a method for obtaining an aqueous extract of Trametes versicolor rich in protein and its hydrolysate, in which liquid nitrogen or a lyophilization process is used and the selection of the optimal extraction temperature at the stage of preparing the raw material for fragmentation and extraction of endogenous proteins, as well as the use of an aqueous extract of Trametes versicolor and its hydrolysate to support the prevention and treatment of diseases caused by staphylococci and streptococci.

Temat hydrolizy enzymatycznej białek grzybowych został przedstawiony w publikacji And i Ismail-Fitry14 dotyczącej przydatności peptydów otrzymywanych na bazie hydrolizy bromelainą. W tej pozycji literaturowej, 0,1% (w/w) roztwór proteazy ProtamexTM był wykorzystywany do pozyskiwania peptydów z podstawczaka pochwiaka wielkopochwowego (Volvariella volvaceae). Reakcję prowadzono w trzech różnych temperaturach i różnych czasach trwania reakcji, co dało ostatecznie stopień hydrolizy sięgający 25%. The digestion efficiency of proteinase K, pepsin, and trypsin at various enzyme concentrations (0.05%, 0.10%, 0.15%), temperatures (30°C, 40°C, and 50°C), and time intervals (60, 90, and 120 min) was also studied by Goswami et al. 16 . The material for the study conducted by this research group were dried fruiting bodies of the oyster mushroom Pleurotus ostreatus, grown on rice straw. The data presented in this publication on the optimization of the enzymatic hydrolysis process clearly showed that the most suitable reaction in terms of the obtained DH value was 0.15% proteinase K at 50°C for 2 hours. The product obtained by Goswami et al. 16 by hydrolysis of fungal proteins was characterized by high foaming efficiency, foam stability, and emulsifying properties. Similar DH values obtained during trypsin digestion were obtained by the team of Farzaneh et al. 17 for two basidiomycetes - the two-spore mushroom (Agaricus bisporus) and the ascomycete - the desert truffle (Terfezia clavery). In the aforementioned article, the DH values obtained ranged from 7.50 to 20.10%, and the preparation of the tested samples differed in the presence or absence of a blanching step.

Zgłaszający prowadzili badania mające na celu uzyskanie nieznanych dotychczas ekstraktów oraz hydrolizatów, nie opisanych w literaturze fachowej, które posiadają właściwości bioaktywne. Istotą wynalazku jest otrzymywanie i zastosowanie uzyskanego w wyniku procesu prowadzonego zgodnie z zaprojektowanym schematem wysokobiałkowego ekstraktu oraz hydrolizatu z wrośniaka różnobarwnego (T. versicolor) do wytwarzania preparatów o działaniu przeciwdrobnoustrojowym, zwłaszcza przeciwbakteryjnym oraz do wytwarzania preparatu o działaniu antyoksydacyjnym. The essence of the invention is the production and use of a high-protein extract and hydrolysate from the variegated taraxacum officinale (T. versicolor), obtained through a process conducted according to a designed protocol, for the production of preparations with antimicrobial, particularly antibacterial, activity, and for the production of a preparation with antioxidant activity. The extracts and hydrolysates obtained according to the invention can be used to obtain preparations with antioxidant activity, including bioactive ingredients in cosmetics and supplements with antiaging properties or as natural preservatives. The extracts obtained from the variegated tarsiers (T. versicolor) according to the invention also demonstrate potent antibacterial activity. Freeze-dried extracts possess species-dependent bactericidal activity against staphylococci, including methicillin-resistant Staphylococcus aureus (MRSA), and bacteriostatic activity against streptococci. The extract obtained without freeze-drying exhibits strain-dependent bactericidal activity against streptococci and bacteriostatic activity against staphylococci. Therefore, the tested mixture may find application in the production of new pharmaceutical products (products) with potential use in the treatment of diseases caused by these bacteria in humans and animals. Similarly, it may be used in disinfectants, both antiseptics, disinfectants, and preservatives. The obtained extracts and hydrolysates are safe to use, because the fungus is a well-known fungus in medicine, used as an adjuvant in the treatment of cancer.
Aktywność antyoksydacyjną ekstraktów i hydrolizatów będących przedmiotem wynalazku oceniano metodami badającymi procent wygaszenia wolnych rodników: kationorodnika ABTS*+ [siarczan 2,2’azyno-di-(3-etylobenztiazoliny)] oraz rodnika DPPH* [2,2-difenylo-1-pikrylohydrazyI]. Jak wynika z uzyskanych danych, powyższy ekstrakt i hydrolizat charakteryzowała zróżnicowana zdolność wygaszania wobec kationorodnika ABTS oraz rodnika DPPH* wahająca się od 50% do niemal 100%.The antioxidant activity of the extracts and hydrolysates being the subject of the invention was assessed using methods examining the percentage of quenching of free radicals: the ABTS*+ cation radical [2,2'azino-di-(3-ethylbenzthiazoline) sulfate] and the DPPH* radical [2,2-diphenyl-1-picrylhydrazine]. As can be seen from the obtained data, the above extract and hydrolysate were characterized by a varied quenching capacity towards the ABTS cation radical and the DPPH* radical, ranging from 50% to almost 100%.

Pomiar absorbancji wykonano po 6-minutowej inkubacji w ciemnym miejscu dla długości fali 734 nm. Określano procent inhibicji rodnika (Rys. 1.) i porównywano go z wartościami uzyskanymi dla liofilizowanych owoców o powszechnie znanych właściwościach przeciwutleniających. Badano wpływ będących przedmiotem wynalazku ekstraktów i hydrolizatów otrzymanych zmodyfikowaną metodą hydrolizy enzymatycznej z wrośniaka różnobarwnego (Trametes versicolor) na wzrost bakterii z rodzaju Staphylococcus i Streptococcus. Przed wykonaniem oznaczeń hodowlę bakterii prowadzono in vitro w warunkach tlenowych na stałym podłożu Mueller-Hinton (MHA, Biomaxima, Polska) w temperaturze 35 ± 2°C przez 18 ± 2 godziny. Do badań wykorzystywano izolaty kliniczne i szczepy wzorcowe z Amerykańskiej Kolekcji Kultur Typowych (ang. American Type Culture Collection, ATCC): S. aureus ATCC 29213, S. aureus ATCC 25923, S. aureus ATCC 6538, S. aureus ATCC 43300 metycylino-oporny (MRSA, ang. methicillin-resistant Staphylococcus aureus), S. aureus ATCC 1707 MRSA, Streptococcus sobrinus ATCC 33478, Streptococcus mutans ATCC 25175, Streptococcus pyogenes ATCC 19615, będące w kolekcji muzealnej Katedry i Zakładu Mikrobiologii Farmaceutycznej Uniwersytetu Medycznego w Lublinie. Aktywność badanych ekstraktów i hydrolizatów otrzymanych w wyniku hydrolizy enzymatycznej oceniano metodą seryjnych mikrorozcieńczeń w podłożu płynnym, zgodnie z rekomendacjami Europejskiego Komitetu ds. Oznaczania Lekowrażliwości (EUCAST) z 2023 roku18. W tym celu zastosowano płytki titracyjne 96-dołkowe. Do świeżej hodowli bakterii dodawano badany ekstrakt wrośniaka różnobarwnego (Trametes versicolor) w postaci homogennej zawiesiny oraz dostępny komercyjnie lek - chloramfenikol (kontrola dodatnia).The effect of the extracts and hydrolysates obtained by a modified enzymatic hydrolysis method from Trametes versicolor on the growth of Staphylococcus and Streptococcus bacteria was studied. Before the assays were performed, the bacteria were cultured in vitro under aerobic conditions on solid Mueller-Hinton medium (MHA, Biomaxima, Poland) at 35 ± 2°C for 18 ± 2 hours. Clinical isolates and reference strains from the American Type Culture Collection (ATCC) were used in the study: S. aureus ATCC 29213, S. aureus ATCC 25923, S. aureus ATCC 6538, S. aureus ATCC 43300 methicillin-resistant Staphylococcus aureus (MRSA), S. aureus ATCC 1707 MRSA, Streptococcus sobrinus ATCC 33478, Streptococcus mutans ATCC 25175, Streptococcus pyogenes ATCC 19615, from the museum collection of the Department of Pharmaceutical Microbiology of the Medical University of Lublin. The activity of the tested extracts and hydrolysates obtained by enzymatic hydrolysis was assessed by serial microdilution in a liquid medium, in accordance with the 2023 recommendations of the European Committee on Antimicrobial Susceptibility Testing (EUCAST). 96-well titration plates were used for this purpose. A fresh bacterial culture was supplemented with the tested extract of Trametes versicolor as a homogeneous suspension and a commercially available drug, chloramphenicol (positive control).
Podczas eksperymentu sporządzano roztwór podstawowy 50 mg badanego ekstraktu z wrośniaka różnobarwnego (Trametes versicolor) poprzez jego rozpuszczenie w 1 mL dimetylosulfotlenku (DMSO). Następnie, z roztworu podstawowego przygotowywano wyjściowy roztwór o stężeniu 32 mg/mL w jałowym płynnym podłożu bulionowym Mueller-Hinton (MHB, Biomaxima, Polska) dla gronkowców lub MHB wzbogaconym 5% odwłóknioną krwią końską (Biomaxima, Polska) i 20 mg/L β-NAD (MHF) dla paciorkowców18. Do pierwszych dołków płytki wielodołkowej dodawano pipetą automatyczną po 200 μL ekstraktu w stężeniu 32 mg/mL, a do pozostałych dołków po 100 μL jałowego płynnego podłoża, zachowując przy tym warunki aseptyczne. Zawartość dołków dokładnie mieszano i wykonywano seryjne rozcieńczenia za pomocą wielokanałowej pipety automatycznej. Dzięki temu uzyskano podwójnie malejące stężenia ekstraktu z wrośniaka różnobarwnego w zakresie 0,0156-32 mg/mL. Następnie, przygotowywano zawiesiny badanych bakterii. W tym celu, za pomocą zwilżonej jałowej wymazówki pobierano 1-2 kolonie do jałowej probówki zawierającej 2 mL jałowego roztworu 0,85% NaCl (Biomerieux, Francja). Sporządzoną zawiesinę doprowadzano do gęstości 0,5 w skali McFarlanda (5 χ 105 CFU (Colony Forming Units)/mL; CFU - jednostki tworzące kolonie). Tak uzyskane zawiesiny badanych drobnoustrojów rozcieńczano w jałowym podłożu MHB lub MHF do uzyskania gęstości końcowej odpowiadającej 5 χ 106 CFU/mL i wprowadzano do wszystkich dołków płytki w objętości 2 μL za pomocą pipety automatycznej. Równocześnie z oznaczeniami aktywności ekstraktu w stosunku do badanych szczepów bakterii z rodzaju Staphylococcus i Streptococcus, przeprowadzano: (a) kontrolę żywotności badanych szczepów bakterii poprzez dodanie 2 μL rozcieńczonej zawiesiny do trzech dołków zawierających czyste podłoże dla każdego szczepu, (b) kontrolę czystego podłoża poprzez umieszczenie w dołkach płytki titracyjnej 100 μL jałowego podłoża bez dodatku badanego związku oraz (c) kontrolę badanego ekstraktu przy tej samej serii rozcieńczeń bez dodatku zawiesiny mikroorganizmów.During the experiment, a stock solution of 50 mg of the tested extract from Trametes versicolor was prepared by dissolving it in 1 mL of dimethyl sulfoxide (DMSO). Subsequently, a stock solution with a concentration of 32 mg/mL was prepared from the stock solution in sterile liquid Mueller-Hinton broth (MHB, Biomaxima, Poland) for staphylococci or MHB supplemented with 5% defibrinated horse blood (Biomaxima, Poland) and 20 mg/L β-NAD (MHF) for streptococci 18 . Using an automatic pipette, 200 μL of the extract at a concentration of 32 mg/mL was added to the first wells of the multiwell plate, and 100 μL of sterile liquid medium was added to the remaining wells, maintaining aseptic conditions. The well contents were thoroughly mixed and serial dilutions were performed using a multichannel automatic pipette. This resulted in doubly decreasing concentrations of the extract from variegated tarsiers ranging from 0.0156 to 32 mg/mL. Suspensions of the tested bacteria were then prepared. To do this, using a moistened sterile swab, 1-2 colonies were collected into a sterile tube containing 2 mL of sterile 0.85% NaCl solution (Biomerieux, France). The prepared suspension was adjusted to a density of 0.5 on the McFarland scale (5 χ 105 CFU (Colony Forming Units)/mL). The suspensions of tested microorganisms were diluted in sterile MHB or MHF medium to obtain a final density of 5 x 106 CFU/mL and added to all wells of the plate in a volume of 2 μL using an automatic pipette. Simultaneously with the determination of the extract activity against the tested bacterial strains of the Staphylococcus and Streptococcus genera, the following were performed: (a) a check of the viability of the tested bacterial strains by adding 2 μL of the diluted suspension to three wells containing clean medium for each strain; (b) a check of the clean medium by placing 100 μL of sterile medium without the test compound in the wells of the titration plate; and (c) a check of the tested extract using the same dilution series without the addition of the microorganism suspension.
Przygotowane w ten sposób mikropłytki inkubowano w temperaturze 35 ± 1°C przez 18 ± 2 godziny w warunkach tlenowych (gronkowce) lub w atmosferze wzbogaconej w 5% CO2 (paciorkowce). Po upływie tego czasu dokonywano odczytu wzrostu drobnoustrojów w poszczególnych dołkach płytki przy użyciu czytnika spektrofotometrycznego Bio-Tek Elx800 (Biokom, Polska) przy długości fali 570 nm oraz obserwacji makroskopowej widocznego zmętnienia. Następnie, pomiary przetwarzano z udziałem programu komputerowego Gen5 ver. 3.03.14 (Biokom, Polska). O wzroście bakterii świadczyło widoczne wzrokowo zmętnienie w studzienkach mikropłytki. Działanie przeciwbakteryjne badanego ekstraktu w stosunku do szczepów gronkowców i paciorkowców oceniano na podstawie wartości minimalnego stężenia hamującego wzrost bakterii (MlC - minimal inhibitory concentration) oraz minimalnego stężenia bakteriobójczego (MBC - minimal bactericidal concentration). Za wartość MIC uznawano dane uzyskiwane w dołkach, w których zaobserwowano prawie całkowite (>90%) zahamowanie namnażania bakterii. Najmniejsze stężenie ekstraktu z wrośniaka różnobarwnego, przy którym stwierdzano brak wzrostu bakterii (99,9%) uznawano za wartość minimalnego stężenia bakteriobójczego (MBC). Na podstawie otrzymanych wyników i w oparciu o współczynnik MBC/MIC, oznaczano aktywność bójczą (MBC/MIC < 4) lub statyczną (MBC/MIC > 4) testowanych ekstraktów z wrośniaka różnobarwnego wobec badanych bakterii. Uzyskane dane przedstawiono w Tabeli 2.The prepared microplates were incubated at 35 ± 1°C for 18 ± 2 hours under aerobic conditions (staphylococci) or in an atmosphere enriched with 5% CO2 (streptococci). After this time, microbial growth in individual plate wells was read using a Bio-Tek Elx800 spectrophotometric reader (Biokom, Poland) at a wavelength of 570 nm, and the visible turbidity was observed macroscopically. The measurements were then processed using the Gen5 computer program version 3.03.14 (Biokom, Poland). Bacterial growth was indicated by visually visible turbidity in the microplate wells. The antibacterial activity of the tested extract against staphylococci and streptococci strains was assessed based on the minimal inhibitory concentration (MlC) and minimal bactericidal concentration (MBC). The MIC value was defined as data obtained in wells where almost complete (>90%) inhibition of bacterial growth was observed. The lowest concentration of the variegated taraxacum extract at which no bacterial growth was observed (99.9%) was considered the minimum bactericidal concentration (MBC). Based on the obtained results and the MBC/MIC ratio, the bactericidal (MBC/MIC < 4) or static (MBC/MIC > 4) activity of the tested variegated taraxacum extracts against the tested bacteria was determined. Zaletą badanego wodnego ekstraktu z wrośniaka różnobarwnego (Trametes versicolor), otrzymanego zmodyfikowaną metodą będącą przedmiotem wynalazku, jest stwierdzone działanie przeciwbakteryjne zależne od gatunku: obejmujące wpływ bójczy na gronkowce (Staphylococcus spp., w tym na szczep wzorcowy metycylino-oporny - MRSA) i hamujący na namnażanie paciorkowców (Streptococcus spp.) w przypadku ekstraktów uzyskanych z liofilizatów wrośniaka różnobarwnego, a także zależne od szczepu: obejmujące wpływ bójczy na paciorkowce i statyczny na gronkowce poza pewnymi wyjątkami w przypadku ekstraktów uzyskiwanych z pominięciem etapu liofilizacji; a także właściwości antyoksydacyjne bez potrzeby frakcjonowania czy separacji produktu końcowego ekstrakcji na pojedyncze składowe lub ich grupy. Poprzez zastosowanie wodyjako rozpuszczalnika ekstrahującego, możliwe jest łatwe i nietoksyczne wytworzenie ekstraktu oraz hydrolizatu. Produkt końcowy ekstrakcji może stanowić alternatywę wobec tradycyjnych substancji przeciwdrobnoustrojowych, wykorzystywanych w celach wspomagania terapii oraz odkażania, zarówno dezynfekcji jak i antyseptyki, a także konserwujących.The advantage of the tested aqueous extract of Trametes versicolor, obtained by the modified method of the invention, is its observed species-dependent antibacterial activity: including a staphylococcal (Staphylococcus spp.) killifish (including the methicillin-resistant reference strain - MRSA) and an inhibitory effect on the growth of streptococci (Streptococcus spp.) in the case of extracts obtained from lyophilized Trametes versicolor, as well as a strain-dependent activity: including a streptococcal killifish and a staphylococcal killifish, with some exceptions in extracts obtained without the lyophilization step; and its antioxidant properties without the need for fractionation or separation of the final extraction product into individual components or groups thereof. By using water as the extraction solvent, easy and non-toxic production of the extract and hydrolysate is possible. The final extraction product may be an alternative to traditional antimicrobial substances used to support therapy and disinfection, both for disinfection and antisepsis, as well as for preservatives.
Warunkiem przeprowadzenia efektywnego procesu pozyskania produktu o unikalnych właściwościach jest odpowiednie rozdrobnienie substratu grzybiczego gwarantujące przede wszystkim rozbicie ścian komórkowych grzyba. Kolejność etapów procesu musi zostać zachowana, a więc: zamrożenie owocnika, rozdrobnienie z udziałem ciekłego azotu lub alternatywnie liofilizacji w niskiej temperaturze, ekstrakcja białek, hydroliza enzymatyczna oraz dezaktywacja enzymu. Zastosowanie innych niż wymienione właściwości skutkuje otrzymaniem mieszanki o słabszych właściwościach przeciwbakteryjnych. Ważna jest także dezaktywacja temperaturowa enzymu, ponieważ brak tego etapu wpływa na uzyskanie nieaktywnego produktu hydrolizy.A prerequisite for effectively obtaining a product with unique properties is the appropriate fragmentation of the fungal substrate, which primarily guarantees the disruption of the fungal cell walls. The sequence of process steps must be maintained: freezing the fruiting body, fragmentation using liquid nitrogen or, alternatively, low-temperature freeze-drying, protein extraction, enzymatic hydrolysis, and enzyme deactivation. Using other properties than those listed results in a mixture with weaker antibacterial properties. Thermal deactivation of the enzyme is also important, as the omission of this step results in an inactive hydrolysis product.
