Abstract

Wynalazek dotyczy sposobu otrzymywania nierozpuszczalnego preparatu lipazy Mucor circinelloides w drodze hodowli selektanta pleśni Mucor circinelloides TB80300A, którego zarodniki hoduje się na podłożu stałym zawierającym: agar, namok kukurydziany, zemulgowaną frakcję technicznych kwasów tłuszczowych stanowiącą odpad w procesie rafinacji oleju rzepakowego i wodę wodociągową. Hodowlę inokulum selektanta prowadzi się w podłożu ciekłym zawierającym: namok kukurydziany, wodę wodociągową, a nadto frakcję technicznych kwasów tłuszczowych stanowiącą odpad przy rafinacji oleju rzepakowego, zaś hodowlę produkcyjną selektanta prowadzi się na podłożu ciekłym o składzie jak podłoże inokularne. Selektant pleśni Mucor circinelloides TB80300A otrzymuje się ze szczepu pleśni Mucor circinelloides TB wyizolowanego z pancerzy krewetek, który poddaje się aktywacji na podłożu stałym o składzie: zemulgowaną frakcja technicznych kwasów tłuszczowych stanowiąca odpad przy rafinacji oleju rzepakowego, namok kukurydziany, agar, woda destylowana, po czym wyrosłe kolonie poddaje się selekcji w drodze hodowli wstrząsanej na podłożu ciekłym o składzie: frakcja technicznych kwasów tłuszczowych stanowiąca odpad przy rafinacji oleju rzepakowego, namok kukurydziany, woda wodociągowa, przy czym procedurę aktywowania i selekcji pleśni Mucor circinelloides powtarza się kilkakrotnie, a otrzymany selektant przechowuje się na podłożu stałym o składzie: frakcja technicznych kwasów tłuszczowych stanowiąca odpad przy rafinacji oleju rzepakowego, agar, brzeczka piwowarska, w temperaturze 4°C. Wynalazek dotyczy także wytwarzania estrów kwasów tłuszczowych i alkoholi w obecności nierozpuszczalnego preparatu lipazy Mucor circinelloides, otrzymanego opisanym wyżej sposobem, jako katalizatora enzymatycznego.The invention relates to a method for obtaining an insoluble Mucor circinelloides lipase preparation by culturing the Mucor circinelloides TB80300A mold selector, the spores of which are grown on a solid medium containing: agar, corn soak, an emulsified fraction of technical fatty acids constituting waste in the rapeseed oil refining process and tap water. Cultivation of the selector inoculum is carried out in a liquid medium containing: corn soak, tap water, and also a fraction of technical fatty acids constituting waste during rapeseed oil refining, while the cultivation of the selectorant is carried out on a liquid medium with the composition as an inocular medium. The Mucor circinelloides TB80300A mold selector is obtained from the Mucor circinelloides TB mold strain isolated from shrimp armor, which is activated on a solid substrate composed of: the emulsified technical fatty acid fraction that is waste when refining rapeseed oil, corn soak, agar, and distilled water, the grown colonies are selected by shaking culture on a liquid medium composed of: technical fatty acid fraction being the waste when refining rapeseed oil, corn soak, tap water, the procedure of activating and selecting the Mucor circinelloides mold is repeated several times, and the obtained selector is stored on a solid medium composed of: technical fatty acid fraction being waste from rapeseed oil refining, agar, brewing wort, at 4 ° C. The invention also relates to the production of fatty acid esters in the presence of an insoluble Mucor circinelloides lipase preparation obtained by the method described above as an enzyme catalyst.
