Abstract

Przedmiotem wynalazku jest sposób otrzymywania warstwy polimeru, rozpoznającego aminokwasy wybrane spośród aminokwasów występujących w białku wiążącym TATA, przyłączone przez kasetę TATA, metodą molekularnego wdrukowania prowadzącego do wytworzenia polimeru wdrukowanego molekularnie (MIP), za pomocą polimeryzacji elektrochemicznej w warunkach potencjodynamicznych. We wdrukowaniu tym jako monomery funkcyjne do elektropolimeryzacji stosuje się 4-[2-(6-amino-9H-puryn-9-ylo)etoksy]fenylo-4-[bis(2,2'-bitien-5-ylo)metan] i 1-tyminooctan 4-(di-2,2'-bitiofen-5-ylometylo)fenylu, a jako monomer sieciujący stosuje się (2,4,5,2',4',5'-heksa(tiofen-2-ylo)-3,3'-bitiofen. Wdrukowanie to charakteryzuje się tym, że jako szablon i analit stosuje się sześcionukleotydową sekwencję jednoniciowego DNA lub peptydowy analog jednoniciowego DNA, tj. PNA. Ponadto wynalazek obejmuje zastosowanie warstwy rozpoznającego MIPu, otrzymanej sposobem według wynalazku, jako elementu czujnika chemicznego do wykrywania i/lub selektywnego oznaczania sześcionukleotydowej sekwencji DNA, w płynach ustrojowych pacjenta, z wykorzystaniem impedimetrycznych pomiarów pojemnościowych (CI) w warunkach analizy przepływowo - wstrzykowej (FIA). Ponadto wynalazek obejmuje zastosowanie warstwy rozpoznającej MIPu do badania efektywności wdrukowania a następnie ekstrakcji szablonu TATAAA oraz przyłączania analitu TATAAA przez warstwę MIPu za pomocą elektrochemicznej spektroskopii impedancyjnej (EIS), odbiciowo - absorpcyjnej spektroskopii w podczerwieni (IRRAS) i różnicowej woltamperometrii pulsowej (DPV) oraz zastosowanie warstwy rozpoznającej MIPu do badania oddziaływań z aminokwasami oddziaływującymi z kasetą TATA za pomocą mikrograwimetrii piezoelektrycznej (PM) w warunkach analizy strzykowo - przepływowej (FIA). W szczególności wynalazek ma zastosowanie w inżynierii genetycznej, zwłaszcza do testów genetycznych przy wczesnym wykrywaniu dziedzicznych chorób genetycznych oraz do badań podstawowych modelu transkrypcji DNA.The subject of the invention is a method of obtaining a polymer layer that recognizes amino acids selected from among those found in the TATA binding protein, attached via a TATA cassette, by molecular imprinting to produce a molecularly imprinted polymer (MIP) by electrochemical polymerization under potentiodynamic conditions. In addition, the invention includes the use of a MIPu recognition layer for testing the printing efficiency then template extraction TATAAA and the attachment of the TATAAA analyte through the MIP layer by means of electrochemical impedance spectroscopy (EIS), reflection and absorption infrared spectroscopy (IRRAS) and differential pulse voltammetry (DPV) and the use of the MIPu recognition layer to study interactions with amino acids interacting with the Timetra cassette using micrometry piezoelectric (PM) under conditions of teat-flow analysis (FIA). In particular, the invention is applicable in genetic engineering, in particular for genetic testing for early detection of hereditary genetic diseases and for basic research of the DNA transcription model.
