Abstract
Przedmiotem wynalazku jest sposób amplifikacji komplementarnego DNA w reakcji łańcuchowej polimerazy w czasie rzeczywistym z odwrotną transkrypcją (RT-qPCR) za pomocą starterów genowo i regiono-specyficznych dla prekursora miRNA-944, który charakteryzuje się tym, że stosuje się startery posiadające następujące sekwencje nukleotydowe: a) startery genowo-specyficzne (służące przepisaniu sekwencji RNA pri-miRNA-944 oraz GAPDH na komplementarne dla nich DNA - cDNA) wykorzystywane w pierwszym etapie RT-qPCR: 1. starter genowo-specyficzny (G1) dla RNA pri-miRNA-944: 5'- ATGGGAGACACAGC - 3', 2. starter genowo-specyficzny (G2) dla RNA GAPDH: 5' - GGACTGAGATTGGC - 3' b) startery regiono-specyficzne (służące amplifikacji cDNA pri-miRNA-944 oraz GAPDH) wykorzystywane w drugim etapie RT-qPCR: 1. starter sensowny (F1) dla cDNA pri-miRNA-944: 5'- GTTCCAGACACATCTCATCTGATA - 3', 2. starter antysensowny (R1) dla cDNA pri-miRNA-944: 5'- TCCCAGACACAGCTCATCCG - 3', 3. starter sensowny (F2) dla cDNA GAPDH: 5,- CCATCTCAGTCGTTCCCAAAGT - 3', 4. starter antysensowny (R2) dla cDNA GAPDH: 5'- AGGTGATCGGTGCTGGTTCC - 3'.
