Abstract

Przedmiotem zgłoszenia jest sposób wytwarzania modyfikowanej bionanocelulozy (BNC), według wynalazku, polegający na przygotowaniu inokulum poprzez zaszczepienie na podłożu płynnym Hestrin-Schramm zawierającym glukozę, ekstrakt drożdżowy, pepton bakteryjny, kwas cytrynowy, Na<sub>2</sub>HPO<sub>4</sub>, MgSO<sub>4</sub> • H<sub>2</sub>O, w płynnej wysterylizowanej pożywce hodowlanej bakterii fermentacji octowej z rodzaju Komagataeibacter, korzystnie szczepem bakterii Komagataeibacter xylinus. Następnie wymieszaniu przez 15 minut i inkubowaniu przez 7 dni w temperaturze 25 - 30°C, ponownym wymieszaniu przez 5 minut, i, przeniesieniu tak otrzymanego inokulum w ilości 5 – 20% objętościowych do podłoża produkcyjnego i prowadzeniu hodowli stacjonarnej przez 3 - 20 dni w temperaturze 25 - 30°C. Następnym oczyszczaniu za pomocą 0,1 M roztworu NaOH w 80°C przez 30 min i przepłukiwaniu wodą destylowaną do momentu ustabilizowania pH na poziomie 6,5 - 7,5 i wysuszeniu. Istota wynalazku polega na tym, że materiał BNC poddaje się modyfikacji za pomocą plazmy niskociśnieniowej, czyli zjonizowanych cząsteczek gazu przez czas 1 - 30 minut. Proces prowadzi się w generatorze plazmy przy przepływie gazu w zakresie 5 - 50 sccm.The subject of the application is a method of producing modified bionanocellulose (BNC), according to the invention, consisting in the preparation of an inoculum by inoculation on a Hestrin-Schramm liquid medium containing glucose, yeast extract, bacterial peptone, citric acid, Na<sub>2</sub>HPO<sub >4</sub>, MgSO<sub>4</sub> • H<sub>2</sub>O, in a liquid sterilized culture medium of acetic fermentation bacteria of the Komagataeibacter genus, preferably a Komagataeibacter xylinus bacterial strain. Then, mixing for 15 minutes and incubation for 7 days at 25 - 30°C, mixing again for 5 minutes, and transferring the inoculum thus obtained in the amount of 5 - 20% by volume to the production medium and carrying out stationary culture for 3 - 20 days in at 25 - 30°C. Subsequent purification with 0.1 M NaOH solution at 80°C for 30 min and rinsing with distilled water until the pH stabilized at 6.5 - 7.5 and drying. The essence of the invention is that the BNC material is modified using low-pressure plasma, i.e. ionized gas molecules, for 1 - 30 minutes. The process is carried out in a plasma generator with a gas flow in the range of 5 - 50 sccm.
