Abstract

Przedmiotem zgłoszenia jest układ reakcyjno-detekcyjny z wymiennym kartridżem do urządzenia do zautomatyzowanego oznaczania analitu w fazie ciekłej na drodze prowadzenia specyficznej reakcji chemicznej oraz następczego optycznego pomiaru stężenia jej produktów, zwłaszcza do monitorowania postępu procesu dializy, opisanego w zgłoszeniu wynalazku nr P.441721, który to układ charakteryzuje się tym, że cylinder (1), z dwoma tłokami (2) poruszanymi za pomocą silników krokowych (3) połączonych z nimi za pośrednictwem złączy (4), jest ściśle osadzony we wnętrzu bloku obudowy (5) i szczelnie z nim połączony swoją powierzchnią zewnętrzną, który to cylinder (1) ma otwory (10, 12, 14, 16) przechodzące odpowiednio w kanały (11, 13, 15, 17) w bloku obudowy (5), przy czym te otwory i kanały mają parami (10-11, 12-13, 14-15, 16-17) jednakową średnicę, a każdy z co najmniej czterech otworów (10) i odpowiednich kanałów (11), jest połączony rozłącznie z jednym zbiornikiem (20) w kartridżu (30) za pośrednictwem portów (28) z podebraniem schodkowym (29), wyposażonych w uszczelki bocznie uszczelniające (27) oraz dekiel dociskający (26), przyjmujących rozłącznie trzpienie przelotowe (25) kartridża (30), połączone za pomocą kanałów (24) z gniazdami (23) przyjmującymi rozłącznie końcówki dozujące (22) zbiorników (20), zaś otwór (12) i kanał (13) wyposażony w szybkozłącze (40) jest połączony rozłącznie za pomocą węża (41) ze źródłem próbki, tj. z automatycznym układem próbkowania, rurą ze strumieniem próbki lub śluzą powietrzną (50) na rurze, zaś otwory (14, 16) i kanały (15, 17) wyposażone w szybkozłącza (45, 43) są połączone rozłącznie za pomocą węży (46, 44) z kanałem odpadowym (61), przy czym wąż (46) może się korzystnie łączyć wcześniej z wężem (44), a ponadto, blok obudowy (5) ma co najmniej jedno poprzeczne przelotowe wybranie o okrągłym przekroju poprzecznym, stanowiące komorę detekcji (6), odsłaniające transparentne ściany cylindra (1), pozwalające na montaż po obu jego stronach elementów optycznego układu detekcji, przy czym blok detekcji (6) i cylinder (1) są do siebie prostopadłe, a ich osie się przecinają, korzystnie bezpośrednio nad wylotem otworu (14) i kanału (15). Zgłoszenie obejmuje także sposób zautomatyzowanego oznaczania analitu w fazie ciekłej na drodze prowadzenia specyficznej reakcji chemicznej oraz następczego optycznego pomiaru stężenia jej produktów, wykorzystujące urządzenie do zautomatyzowanego oznaczania analitu w fazie ciekłej na drodze prowadzenia specyficznej reakcji chemicznej oraz późniejszego optycznego pomiaru stężenia jej produktów, zwłaszcza do monitorowania postępu procesu dializy opisane w zgłoszeniu wynalazku nr P.441721, który charakteryzuje się tym, że urządzenie zawiera układ reakcyjno-detekcyjny z wymiennym kartridżem opisany powyżej, przy czym czwarty zbiornik (20D) w kartridżu (30) służy jako mieszalnik, a wszystkie roztwory zaciągane sekwencyjnie do cylindra (1) podczas oznaczania badanej próbki, tj. próbka ze źródła próbki (50) lub roztwór wzorcowy ze zbiornika (20A) oraz odczynniki ze zbiorników (20B, 20C) natychmiast po ich zaciągnięciu do cylindra (1) przetłacza się do mieszalnika (20D), a po zaciągnięciu i przepompowaniu wszystkich roztworów do czwartego zbiornika (20D), uzyskany roztwór reakcyjny miesza się poprzez jego przetłaczanie pomiędzy cylindrem (1) a zbiornikiem (20D), przy czym objętość badanej próbki wynosi 30 — 90 µl, objętość stosowanych reagentów wynosi 50 — 250 µl, uzyskując mieszaninę reakcyjną o objętości 240 — 320 µl, zaś po zakończeniu mieszania roztworu reakcyjnego przetłacza się jego porcję, korzystnie 240 µl, z mieszalnika (20D) do cylindra (1), a następnie przesuwa się ją pomiędzy tłokami (2) do obszaru komory detekcji (6), gdzie prowadzi się oznaczenie analitu, a po oznaczeniu wypompowuje się roztwór reakcyjny z cylindra w obszarze komory detekcji (6) i zbiornika (20D) odpowiednio poprzez kanał (15) i kanał (17) do kanału odpadowego (61).The subject of the application is a reaction-detection system with a replaceable cartridge for a device for automated determination of an analyte in the liquid phase by conducting a specific chemical reaction and subsequent optical measurement of the concentration of its products, especially for monitoring the progress of the dialysis process, described in the patent application no. P.441721, which system is characterized in that a cylinder (1), with two pistons (2) moved by means of stepper motors (3) connected to them by means of connectors (4), is closely mounted inside the housing block (5) and tightly connected to it by its external surface, which cylinder (1) has openings (10, 12, 14, 16) passing into channels (11, 13, 15, 17) in the housing block (5), respectively, wherein these openings and channels have the same diameter in pairs (10-11, 12-13, 14-15, 16-17), and each of at least four the openings (10) and corresponding channels (11) are releasably connected to one tank (20) in the cartridge (30) by means of ports (28) with a stepped undercut (29), provided with side seals (27) and a pressure cap (26) releasably receiving the through-hole pins (25) of the cartridge (30), connected by means of channels (24) to sockets (23) releasably receiving the dispensing tips (22) of the tanks (20), and the opening (12) and channel (13) provided with a quick connector (40) are releasably connected by means of a hose (41) to a sample source, i.e. to an automatic sampling system, a sample stream pipe or an airlock (50) on the pipe, and the openings (14, 16) and channels (15, 17) provided with quick connectors (45, 43) are releasably connected by means of hoses (46, 44) with a waste channel (61), wherein the hose (46) can advantageously be connected in advance to the hose (44), and furthermore, the housing block (5) has at least one transverse through-hole of circular cross-section, constituting a detection chamber (6), exposing transparent walls of the cylinder (1), allowing for the mounting of elements of an optical detection system on both sides thereof, wherein the detection block (6) and the cylinder (1) are perpendicular to each other and their axes intersect, advantageously directly above the outlet of the opening (14) and the channel (15). The application also covers a method of automated determination of an analyte in the liquid phase by means of conducting a specific chemical reaction and subsequent optical measurement of the concentration of its products, using a device for automated determination of an analyte in the liquid phase by means of conducting a specific chemical reaction and subsequent optical measurement of the concentration of its products, especially for monitoring the progress of the dialysis process described in the application for invention No. P.441721, which is characterized in that the device comprises a reaction-detection system with a replaceable cartridge described above, wherein the fourth tank (20D) in the cartridge (30) serves as a mixer, and all solutions drawn sequentially into the cylinder (1) during the determination of the tested sample, i.e. the sample from the sample source (50) or the standard solution from the tank (20A) and the reagents from the tanks (20B, 20C) are immediately pumped into the mixer (20D) after being drawn into the cylinder (1), and after being drawn and pumping all the solutions to the fourth tank (20D), the obtained reaction solution is mixed by pumping it between the cylinder (1) and the tank (20D), wherein the volume of the tested sample is 30 - 90 µl, the volume of the used reagents is 50 - 250 µl, obtaining a reaction mixture with a volume of 240 - 320 µl, and after the mixing of the reaction solution is finished, a portion thereof, preferably 240 µl, is pumped from the mixer (20D) to the cylinder (1), and then it is moved between the pistons (2) to the area of the detection chamber (6), where the analyte is determined, and after the determination, the reaction solution is pumped out of the cylinder in the area of the detection chamber (6) and the tank (20D) through the channel (15) and the channel (17), respectively, to the waste channel (61).
