Abstract
Przedmiotem zgłoszenia jest bioinokulat zawierający szczep bakterii endofitycznej oraz biowęgiel, mający zastosowanie jako biopreparat o przeznaczeniu rolniczym. Bioinokulat zawierający szczep bakterii endofitycznej oraz biowęgiel charakteryzuje się tym, że szczep bakterii należy do rodzaju Labedella. Przedmiotem zgłoszenia jest także sposób otrzymywania bioinokulatu zawierającego szczep bakterii endofitycznej oraz biowęgiel, mający zastosowanie jako biopreparat o przeznaczeniu rolniczym. Sposób otrzymywania bioinokulatu według wynalazku powyżej charakteryzuje się tym, że wyjściową płynną hodowlę bakterii rodzaju Labedella przenosi się do sterylnej płynnej pożywki w proporcji 1 ml hodowli bakteryjnej do 100 ml pożywki i inkubuje na wytrząsarce przy 120 - 145 rpm w temperaturze w zakresie 24°C - 26°C do osiągnięcia przez bakterie logarytmicznej fazy wzrostu, następnie biowęgiel przesiewa się przez sito o oczkach co najwyżej 0,75 mm, po czym biowęgiel miesza się z zawiesiną bakteryjną w stosunku 5:100 w/v w warunkach sterylnych i wytrząsa przez 2h na wytrząsarce przy 120 - 145 rpm w temperaturze w zakresie 24°C - 26°C, następnie dodaje się 2% w/v alginianu sodu w warunkach sterylnych i kontynuuje wytrząsanie przez 1h do uzyskania jednorodnej mieszaniny, otrzymaną zawiesinę wkrapla się do wymieszanego i wysterylizowanego 2% w/v roztworu chlorku wapnia na mieszadle magnetycznym z wysokości od 15 cm do 20 cm, następnie otrzymane granulki pozostawia się w roztworze chlorku wapnia na 24h w 4°C do stwardnienia, a następnie zebrane granulki przepłukuje się sterylną wodą i poddaje liofilizacji przez 12 – 20 h.
