Abstract
Istotą zgłoszenia jest sposób indukowania depigmentacji larw ryb, zwłaszcza danio pręgowanego polegający na przygotowaniu medium odżywczego dla ryb zawierającego mieszaninę soli w wodzie demineralizowanej z dodatkiem substancji aktywnej, a następnie umieszczeniu w nim ryby danio pręgowanego i utrzymywaniu ich w medium odżywczym do pożądanego stopnia odbarwienia, korzystnie całkowitego odbarwienia, gdzie jako substancję aktywną stosuje się cefazolinę w stężeniu od 0,15 mM do 1,4 mM, w ilości od 0,0682 g do 0,6363 g na 1 L wodnego roztworu medium odżywczego. Sposób depigmentacji w alternatywnym rozwiązaniu polega na tym, że jako substancję aktywną stosuje się kompleks cefazoliny z cyklodekstryną o stosunku molowym na etapie formulacji, korzystnie 1:2, odpowiadający stężeniu cefazoliny w finalnym medium w zakresie od 0,11 mM do 0,25 mM, co odpowiada ilości od 0,3888 g do 0,884 g kompleksu 1:2 na 1 L wodnego roztworu medium odżywczego, przy czym stosuje się warunki jak opisane w pierwszym rozwiązaniu. Zgłoszenie pozwala uzyskiwać każdorazowo do celów badawczych transparentną rybę zwłaszcza w badaniach in vivo prowadzonych nad rozwojem narządów i funkcjonowaniem organizmu, patogenezą chorób, testowaniem leków mających na celu opracowanie nowych skutecznych terapii.The essence of the application is a method of inducing depigmentation of fish larvae, especially zebrafish, consisting in preparing a nutrient medium for fish containing a mixture of salts in demineralized water with the addition of an active substance, and then placing zebrafish therein and keeping them in the nutrient medium until the desired degree of discoloration, preferably complete discoloration, wherein cefazolin is used as the active substance at a concentration of 0.15 mM to 1.4 mM, in an amount of 0.0682 g to 0.6363 g per 1 L of an aqueous solution of the nutrient medium. The alternative depigmentation method involves using a cefazolin-cyclodextrin complex as the active ingredient, preferably in a 1:2 molar ratio at the formulation stage, corresponding to a cefazolin concentration in the final medium ranging from 0.11 mM to 0.25 mM, which corresponds to an amount of 0.3888 g to 0.884 g of the 1:2 complex per 1 L of aqueous nutrient solution, using the same conditions as described in the first embodiment. The application allows for obtaining transparent fish for research purposes, particularly in in vivo studies on organ development and body function, disease pathogenesis, and drug testing aimed at developing new effective therapies.
