本發明的組成物包含鋅離子來源。鋅離子來源可提供Zn2+離子至口腔,包括傳遞到口腔表面。在一實施例中,鋅離子來源可傳遞Zn2+離子至包括牙齦上皮細胞的口腔黏膜。在一實施例中,鋅離子來源包含一或多種鋅鹽。在一實施例中,鋅離子來源包含選自氯化鋅、醋酸鋅、葡萄糖鋅、硫酸鋅、氟化鋅、檸檬酸鋅、乳酸鋅、氧化鋅、單甘油酸鋅、酒石酸鋅、焦磷酸鋅、磷酸鋅、馬來酸鋅、蘋果酸鋅、碳酸鋅、抗壞血酸鋅、氫氯離胺酸鋅和單水氫

在一實施例中,組成物包含介於1和20,000ppm的鋅。在一實施例中,組成物包含介於1和10,000ppm、介於1和5,000ppm的鋅、介於1和2,000ppm的鋅、介於1和1,000ppm的鋅、介於1和500ppm的鋅、介於1和200ppm的鋅、介於1和100ppm的鋅、介於1和50ppm的鋅、介於1和25ppm的鋅、介於1和10ppm或介於3至9ppm的鋅。在一實施例中,組成物包含介於4至8ppm的鋅。在一實施例中,組成物包含從0.0001至2.0重量%的鋅。在一實施例中,組成物包含從0.0001至1.0重量%的鋅、從0.0001至0.5重量%的鋅、從0.0001至0.2重量%的鋅、從0.0001至0.1重量%的鋅、從0.0001至0.05重量%的鋅、從0.0001至0.02重量%的鋅、從0.0001至0.01重量%的鋅、從0.0001至0.005重量%的鋅、從0.0001至0.0025重量%的鋅、從0.0001至0.001重量%的鋅或介於0.0003至0.0009重量%的鋅。例如,組成物可包含介於0.0004至0.0008重量%的鋅。

在一實施例中,組成物為口腔護理組成物。在一實施例中,口腔護理組成物選自潔牙劑、牙膏、牙粉、口腔清洗劑、錠劑、口香糖、凝膠、塗料和薄膜。

(sarcosine stannous fluoride)、甘胺酸氟化鉀(glycine potassium fluoride)、甘胺酸氫氟酸鹽(glycine hydrofluoride)、氟化胺或其混合物;及包括鹼金屬單氟磷酸鹽等離子單氟磷酸,例如氟化鉀、氟化鈉和氟化銨、單氟磷酸鉀、單氟磷酸鈉和單氟磷酸銨及其混合物。在部分實施例中,本發明的口腔護理組成物亦可包含量足以提供約50至約5000ppm氟離子、例如從約100至約1000、從約200至約500、或約250ppm的氟離子之氟離子來源或氟提供物質。可將約0.001重量%至約10重量%、例如從約0.003重量%至約5重量%、0.01重量%至約1重量%或約0.05重量%的氟離子來源加入本發明的組成物。然而,可理解用於提供適當氟離子量的氟鹽重量將隨鹽類中的逆離子重量而有明顯差異,且本領域之習知技藝者可快速決定此含量。較佳的氟鹽可為氟化鈉。
組成物可包含至少一種香料,其可用於例如增強組成物的味道。可使用任何口用天然或合成香料,包括但不限於香草精、鼠尾草、馬鬱蘭、洋芹油、綠薄荷油、肉桂油、冬青油(水楊酸甲酯)、薄荷油、丁香油、月桂油、茴香油、桉樹油、柑橘油、水果油和精油(包括來自檸檬、柳橙、萊姆、葡萄柚、杏、香蕉、葡萄、蘋果、草莓、櫻桃、鳳梨等)、來自豆類和堅果的香味(例如咖啡、可可、可樂果、花生和杏仁等)、吸附或包囊化的香料等。在此香料中亦包含在口中提供香味及/或其他感官效果(包括涼爽或溫暖效果)的成分。此種成分說明性地包括薄荷醇、乙酸薄荷酯、乳酸薄荷酯、樟腦、桉樹油、桉樹腦、茴香腦、丁香酚、肉桂(cassia)、口惡酮(oxanone)、α-紫羅蘭酮、丙烯基乙基愈創木酚(propenyl guaiethol)、百里酚、沈香醇、苯甲醛、肉桂醛、N-乙基-對-薄荷燒-3-羧胺、N,2,3-三甲基-2-異丙基丁醯胺、3-(1-薄荷氧基)-丙烷-1,2-二醇、肉桂醛甘油縮醛(CGA)、薄荷酮甘油縮醛(MGA)等。選擇性地,一種或多種香料的總含量為從約0.01重量%至約5重量%,例如從約0.1重量%至約2.5重量%,以組成物的總重量為基準。
選擇性地,本發明的組成物整合一種或多種抗敏感劑,例如硝酸鉀、碳酸氫鉀、氯化鉀、檸檬酸鉀和草酸鉀等鉀鹽、
辣椒素、丁香酚、鍶鹽和氯鹽及其組合。可加入有效量的此種藥劑,例如從約1重量%至約20重量%(以組成物的總重量為基準),隨所選藥劑而有所不同。
組成物可進一步包含抗氧化劑。可使用任何口用抗氧化劑,包括丁基羥基甲氧苯(BHA)、二丁基羥基甲苯(BHT)、維生素A、類胡蘿蔔素、維生素E、類黃酮、多酚、抗壞血酸、藥草抗氧化劑、葉綠素、褪黑激素及其混合物。
另外選擇性地,本發明的組成物可另外包含以下提供的牙垢控制(抗牙垢)劑。在此有用的牙垢控制劑包括上述指定藥劑的鹽類,包括鹼金屬和銨鹽。藥劑包括:磷酸鹽和多磷酸鹽(例如焦磷酸鹽)、聚胺基丙磺酸(AMPS)、聚烯烴磺酸鹽、聚烯烴磷酸鹽、二膦酸鹽例如氮雜環烷-2,2-二膦酸鹽(例如氮雜環庚烷-2,2-二膦酸)、N-甲基氮雜環戊烷-2,3-二膦酸、乙烷-1-羥基-1,1-二膦酸(EHDP)和乙烷-1-胺基-1,1-二膦酸鹽、膦醯基鏈烷羧酸及其混合物。有用的無機磷酸鹽和多磷酸鹽包括磷酸單鈉、磷酸二鈉和磷酸三鈉、三聚磷酸鈉、四聚磷酸鈉、焦磷酸單鈉、焦磷酸二鈉、焦磷酸三鈉、焦磷酸四鈉、三偏磷酸鈉、六偏磷酸鈉及其混合物。其他有用的防牙垢劑包括聚羧酸鹽聚合物、聚乙烯甲基醚/馬來酸酐(PVME/MA)共聚物,例如GANTREZ®。可加入無效量的此種藥劑,例如從1至20重量%。
在部分實施例中,組成物可包含一種或多種胺基酸。可整合至本發明的組成物之胺基酸包括精胺酸、組胺酸、賴胺酸、天冬胺酸、麩胺酸、絲胺酸、蘇胺酸、天冬醯胺酸、麩胺醯胺、半胱胺酸、甘胺酸、脯胺酸、丙胺酸、纈胺酸、異白胺酸、白胺酸、甲硫胺酸、苯丙胺酸、酪胺酸和色胺酸。在部分實施例中,組成物包含一種或多種胺基酸,選自左式精胺酸、半胱胺酸、異白胺酸、左式賴胺酸、麩胺酸、絲胺酸及其混合物。在部分實施例中,組成物包含左式賴胺酸。
在部分實施例中,組成物可包含多肽。例如,組成物可包括含有25-30個賴胺酸殘餘物的聚賴胺酸。
在部分實施例中,本發明的組成物進一步包含營養成分。適合的營養成分包括維生素、礦物質、胺基酸及其混合物。維生素包含維生素C和D、硫胺素、核黃素、泛酸鈣、菸酸、葉酸、菸鹼醯胺、吡哆醇、氰鈷胺、對胺苯甲酸、生物類黃酮及其混合物。營養補充劑包括胺基酸(例如左式色胺酸、左式賴胺酸、甲硫胺酸、蘇胺酸、左旋肉鹼和左式肉鹼)、親脂物(例如膽鹼、肌醇、甜菜鹼和亞麻油酸)及其混合物。
本發明亦提供一種用於抑制牙周病原菌附著或侵入齒齦上皮細胞的方法,該方法包含施加包含鋅離子來源的組成物至齒齦上皮組織。可使用手或牙刷等工具(例如將牙刷浸入口腔護理組成物再施加至齒齦上皮組織)將組成物施加至組織。或者,能夠在液體口腔清洗劑中施加組成物。在一實施例中,能夠以每日介於1和4次的頻率(亦即在24小時內介於1和4次的頻率)施加組成物至齒齦上皮細胞。在一實施例中,以每日1次的頻率施加組成物至齒齦上皮組織。
實例:
細胞製備:
將人類牙周韌帶成纖維細胞(Human periodontal ligament fibroblasts/HPdLF,Lonza cat#CC-7049)植入平板,並使用包含基質細胞基礎培養基、hFGF-B、胰島素、FBS和GA-1000的SCGM Bullet Kit(Lonza,#CC-3205)在37℃下以5% CO2培養一夜。
細菌製備:
在37℃下將病原細菌(伴於線桿菌(Actinobacillus actinomycetemcomitans)(Aa)ATCC cat#29522或牙齦卟啉單胞菌(Porphyromonas gingivalis)(Pg)ATCC cat#33277)培養一夜(對伴放線桿菌(Actinobacillus actinomycetemcomitans)使用胰蛋白酶,而對牙齦卟啉單胞菌(Porphyromonas gingivalis)使用半強度的腦心浸液培養基(以酵母萃取物、氯化血紅素和vitK1補充)。讀取到的光學密度為610奈米。將細菌以8000轉/分離心10分鐘,使沈澱細胞重新懸浮在細胞培養液中,再加以稀釋至OD610為0.1和0.2。
細胞感染:
將測試樣品(溶於水)加入細胞,在5% CO2培養器內以37℃培養30分鐘,再加入細菌。藉由將重新懸浮的細菌加進細胞並進一步在5% CO2培養器內以37℃培養90至120分鐘,使細菌感染細胞。
細菌侵入之判定:
使用PBS清洗細胞5次以移除非附著細菌。加入含有1x青黴素(100 IU)和鏈黴素(100微克/毫升)新鮮細胞培養基,然後在5% CO2培養器內以37℃培養2個小時以殺死細胞外細菌。然後移除培養基並以PBS清洗細胞3次。在37℃下將溶於PBS的1毫升1%皂素溶液加進受感染的細胞達5分鐘以溶解細胞並釋放內化的細菌。使用100微升樣品,將稀釋物(在PBS中連續稀釋,稀釋因子為10)鋪板至合適的培養基上,並使用消毒過棒狀物塗抹展開,然後在厭氧培養室內以37℃培養2-3天,並計算最終的細菌聚落。
結果如表1和2所示。表1顯示氧化鋅對伴放線桿菌(Actinobacillus actinomycetemcomitans)和牙齦卟啉單胞菌(Porphyromonas gingivalis)的影響。表1顯示使用氧化鋅處理過的細胞具有較少的內化伴放線桿菌(Actinobacillus actinomycetemcomitans)和內化牙齦卟啉單胞菌(Porphyromonas gingivalis)。在本實例中,氧化鋅減少內化伴放線桿菌(Actinobacillus actinomycetemcomitans)約15%和內化牙齦卟啉單胞菌(Porphyromonas gingivalis)約6%。表2顯示氧化鋅和檸檬酸鋅皆減少侵入人類牙周韌帶成纖維細胞的內化伴放線桿菌(Actinobacillus actinomycetemcomitans)。8ppm的檸檬酸鋅減少內化伴放線桿菌(Actinobacillus actinomycetemcomitans)約66%,而4ppm的氧化鋅減少內化伴放線桿菌(Actinobacillus actinomycetemcomitans)約23%。
Claims (15)
- 一種組成物,包含用於抑制牙周病原菌附著或侵入口腔軟組織上皮細胞的鋅離子來源。
- 如申請專利範圍第1項的組成物,其中該口腔軟組織為齒齦上皮細胞。
- 如上述申請專利範圍之任一項的組成物,其中該牙周病原菌引起、傳播或加劇選自牙齦炎和牙周炎的牙周病。
- 如申請專利範圍第1項的組成物,其中該牙周病原菌為轉糖鏈球菌(Streptococcus mutans)、遠緣鏈球菌(Streptococcus sobrinus)、血鏈球菌(Streptococcus sanguis)、口腔鏈球菌(Streptococcus oralis)、牙齦卟啉單胞菌(Porphyromonas gingivalis)、犬齒齦液卟啉單胞菌(Porphyromonas cangingivalis)、伴放線桿菌(Actinobacillus actinomycetemcomitans)、福賽斯坦納菌(Tannerella forsythia)、革蘭氏陰性桿狀菌(Fusobacterium nucleatum)和牙垢密螺旋體(Treponema denticola)之其中一種或多種。
