抗 H I V剤
技術分野
本発明は後天性免疫不全症候群 (AIDS : Ac qu i r ed Immune De f i c i en cy Synd r ome) の予防及び治療に有用な抗 H I V (H um a n I mmu nod e f i c i ency V i rus) 剤に する。 背景技術
A I DSは H I V感染を原因とする疾患であり、 その患者数は 1983年に米 国で発見されて以来、 急速に増加している。 かかる A I DSの治療には抗 H I V 剤であるアジドチミジン (AZT) 、 ジダノシン (DDI) などが知られている c しかしながら、 AZTは著明な延命効果が認められてはいるものの、 副作用と して頭痛や胃腸障害、 骨髄抑制作用等が発生するという問題がある。 また、 八∑丁ゃ00 Iはいずれも H I V初感染細胞に対しては著明な増殖抑制効果を示 すが、 既に H I Vが感染した細胞に対しては効果を発揮しないため、 A I DS関 連症候群の発症予防作用や患者の延命効果を有するが、 H I V感染後の細胞を抑 制する効果、 すなわち A I DSの根本治療には役立たないとされている。
従って、 本発明の目的は H I V初感染細胞はもとより H I V持続感染細胞に対 して強い増殖抑制作用を示し、 感染後の A I DS治療に有用な抗 H I V剤を提供 ίる と る。
発明の開示
本発明は次の一般式 (1) :

(式中、 R1 は炭素数 2〜 20の了ルケ二ル基を示し、 R2 は炭素数 2〜20の 了ルケニル基又は炭素数 1〜 20のアルキル基を示す)
で表わされるジスルフィ ド、 又は
一般式 (2) :
R!-D-R3 ( 2 )
(式中、 R1 は炭素数 2〜 20の了ルケ二ル基を示し、 R3 は水素原子、 炭素数 2〜 20のァシル基、 リン酸基又はりン脂質基を示す) で表わされる不飽和アルコール又はそのエステルを有効成分とする抗 H I V剤に 係るものである。
また、 本発明は上記一般式 (1) 又は (2) で表わされる化合物を有効成分と する A I DSの予防及び治療剤に係るものである。
また、 本発明は上記一般式 (1) 又は (2) で表わされる化合物の抗 H I V剤 又は A I DSの予防及び治療剤としての使用に係るものである。
さらに、 本発明は H I V持続感染細胞に対して上記一般式 (1) 又は (2) で 表わされる化合物を投与することを特徴とする H I V持続感染細胞の増殖抑制方 法に係るものである。
上記一般式 (1) 又は (2) で表わされる化合物は、 H IV初感染細胞だけで なく H I V持続感染細胞に対しても優れた増殖抑制作用を有し、 かつ安全性が髙 いので A I DSの予防及び治療のために有用である。

発明を実施するための最良の形態
上記一般式 (1) 又は (2) 中、 R1 及び R2 で示されるアルケニル基として は、 炭素数 2〜 20の直鎖又は分岐鎖の了ルケニル基、 例えばビニル基、 ァリル 基、 1一プロぺニル基、 3—ブテニル基等が挙げられる。 このうち、 炭素数 2〜 6の了ルケニル基が好ましく、 特に了リル基が好ましい。 また、 R2 で示される アルキル基としては、 炭素数 1〜2 0の直鎖又は分岐鎖のアルキル基、 例えばメ チル基、 ェチル基、 n—プロピル基、 Ti—ブチル基、 n—ペンチル基、 n—^ *キ シル基等が挙げられる。 このうち、 炭素数 1〜6のアルキル基が好ましい。 また、 R3 で示される炭素数 2〜2 0のァシル基としては、 例えばァセチル基、 プロピ ォニル基、 プチリル基、 ノヽ'レリル基、 カブ ϋィル基、 力プリル基、 カプリノィル 基、 ラウロイル基、 ミ リストィル基、 パルミ トイル基、 ステアロイル基等が挙げ られる。 また R3 がリン酸基である一般式 (2) の化合物としては、 モノー、 ジ 一又はト リ一了ルケ二ルリ ン酸が挙げられ、 R3 がリン脂質基である一般式 (2) の化合物としては、 グリセ口リン脂質のリン酸残基にアルケニル基が結合 した化合物が挙げられる。
—般式 (1) のジスルフィ ドとしては、 例えばジァリルジスルフィ ド、 ァリル メチルジスルフィ ド、 了リル II一プロピルジスルフィ ド等が挙げられるが、 就中、 特にジァリルジスルフィ ドが好ましい。 一方、 一般式 (2) の化合物の中では、 脂肪酸ァリル、 特に飽和脂肪酸了リルが好ましい。
上記ジスルフィ ド ( 1) のうち、 ジァ リルジスルフィ ドは、 例えばニンニク (A l l i um s a t i v urn) の一成分であり、 本化合物については、 げっ 歯類における実験癌に対する抑制効果が報告 (L e e WW e t a 1 : Can c e r Re s e a r c h 49, 2689 - 2692, 1989 ) され ているが、 抗 H I V作用に関する報告例はない。 また、 このジァリルジスルフィ ドは文献 (J o hn FB e t a 1 : C a n c e r R e s e a r c h 48, 5937 - 5940, 1988 ) によればラッ トに 200 mgZkg経口投与 しても死亡せず、 またニンニクの一成分であることから安全性の髙いものである c 上記不飽和アルコール又はそのエステル (2) (以下、 不飽和化合物 (2) と いう) は、 既知の化合物であり、 常法に従って製造することができ、 安全性につ いても良く知られている。
かかるジスルフィ ド (1) 及び不飽和化合物 (2) は、 後記実施例に示すよう に H I V持続感染株として知られている CEMZLAV細胞に対し顕著な抑制効 果を示す。 その抗 H I V作用機構は明らかではないが、 H I V遺伝子の転写、 複 写経路の促進あるいは抑制、 即ち Ta t関連の遺伝子情報の流れに闋与して H I Vの増殖を抑制しているものと考えられる。 更に本発明者は、 不飽和化合物 (2) において R3 のァシル基の炭素数が増加した場合、 H I Vに対し直接的な 殺生効果が発揮されることを実験的事実として観察している。 また、 その抑制効 果発現に際し、 実施例で効果を確認した化合物には構造式上の部分的類似性 - CH=CHCH20-、 -CH=CHCH2S-又は CH2=CH-0-、 CH2=CH- S-が見られる。 従って、 薬物 が生体に適用された後、 代謝変換されてこの部分的類似構造を呈する化合物は H I V抑制効果を発現する可能性がある。
ジスルフイ ド (1) 又は不飽和化合物 (2) を抗 H I V剤又は A I DSの予防 及び治療剤としてヒトに投与する場合は、 必要に応じて賦形剤、 結合剤、 滑沢剤、 崩壊剤、 被覆剤、 乳化剤、 懸濁剤、 溶剤、 安定化剤、 吸収助剤、 軟罾基剤等を適 宜添加するか、 あるいはリボソーム化等を行い常法によって経口投与用、 注射投 与用、 直腸内投与用等の医薬組成物とすることができる。
経口投与用の製剤としては、 顆粒、 錠剤、 糖衣錠、 カプセル剤、 ソフトカプセ ル剤、 丸剤、 液剤、 乳剤、 懸濁剤等が;注射投与用の製剤としては、 静脈内注射、 筋肉内注射、 皮下注射、 点滴注射用等が;直腸内投与用の製剤としては、 坐薬力 プセル等が好ましい。
投与量は、 投与経路、 患者の年齢、 症状等によって異なるが、 通常成人 1日当 たりジスルフィ ド ( 1 ) 又は不飽和化合物 (2) として l mg~ l 0 gが好ましく、 これを 1回あるいは数回に分けて投与する。
実施例
次に実施例を挙げて本発明を詳細に説明するが、 本発明はこれらに限定される ものではない。
実施例 1
( 1 ) ジァリルジスルフィ ドの連続投与による非感染性 CEM細胞に対する作用 培養液 RPM I — 1 6 4 0 ( 1 0 %F C S含有) 中に、 CEM細胞を浮遊させ、 細胞数 2. 0 X 1 05 個 Zm£濃度とした。 この CEM細胞浮遊液 9. 9 を培養 瓶中にとり、 被検薬ジァリルジスルフイ ド溶液 (0. 0 1 %, ポリオキシェチレ ン (6 0 ) 硬化ヒマシ油中に懸濁) 0. 1 mlを加え、 3 7 、 5 %C02 環境下に 培養した。 2 4時間毎にトリパンブルー色素排除試験により生細胞数を測定する と同時に、 1 2 0 O rpm で遠心分雜して細胞を回収し、 新鮮な培養液と被検薬溶 液を加えて培養を続けた。 その結果を図 1に示す。
(2) ジァリルジスルフィ ドの連続投与による H I V持続産生性 CEMZL AV 細胞に対する作用
上記 (1 ) と同様に CEMZLAV細胞浮遊液を作製し、 実験を行った。 その 結果を図 2に示す。
図 1及び図 2から明らかなように、 ジ了リルジスルフィ ドは 4 0 MU投与で非 感染性 CEM細胞に対する増殖抑制作用は弱いが、 H I V持続産生性 CEMZ LAV細胞の増殖を完全に抑制した。 この結果、 ジスルフィ ド (1 ) は H I V感 染後の細胞の増殖を抑制し、 A I DS治療に有用であることがわかる。

培養液 RPMI - 1 640 (1 0 %FCS含有) 中に C EM細胞を浮遊させ、 細胞数 2. 0 X 1 05 個 Zm£濃度とした。 この CEM細胞浮遊液 9. 9? ^を培養 瓶中にとり、 被検薬として酢酸ァリル溶液 (0. 01%ΡΟΕ 60硬化ヒマシ油 中に懸濁) 0. 1? ^を加え、 37 :、 5%C02 環境下で培養した。 24時間毎に トリパンブルー色素排除試験により生細胞数を測定し、 その結果を図 3に示した。
(2) 単回投与による H I V持続産生性 CEMZLAV細胞に対する作用 上記 (1) と同様に CEMZLAV細胞浮遊液を作製し、 実験を行った。 その 結果を図 4に示す。
図 3及び図 4から明らかなように被検液、 酢酸了リルは 40; uM£l上の濃度で H I V持続産生性 CEM/LAV細胞に対して明らかな細胞増殖の抑制、 即ち生 細胞の減少による死細胞の増加を示した。 一方、 非感染性の CEM細胞に対して は用いた量の範囲内で細胞増殖の抑制を示さなかつた。
実施例 3 ァリルアルコールの効果:
被検薬をァリルアルコールとして実施例 1と同様の実験を行った。 その結果を 図 5、 図 6に示す。
図 5、 図 6から明らかなように、 ァリルアルコール 20 uM以上の濃度では H I V持続産生細胞に対して特異的な増殖抑制の効果を示した。
実施例 4 n一酩酸了リルの効果:
被検薬を n—酪酸ァリルとして実施例 1と同様の実験を行った。 その結果を図 7、 図 8に示す。
図 7、 図 8から明らかなように、 n—酪酸ァリル 40 juM以上の濃度では H I V持続産生細胞に対して特異的な増殖抑制の効果を示した。
実施例 5 n—力プロン酸了リルの効果:
被検薬を n—力プロン酸ァリルとして実施例 1と同様の実験を行った。 その結 果を図 9、 図 1 0に示す。
図 9、 図 1 0から明らかなように、 ΙΊ—力プロン酸了リル 80 uM以上の濃度 では H I V持続産生細胞に対して特異的な増殖抑制の効果を示した。
実施例 6 n—力プリル酸ァリルの効果: 被検薬を]!一力プリル酸ァリルとして実施例 1と同様の実験を行った。 その結 果を図 1 1、 図 1 2に示す。
図 1 1、 図 1 2から明らかなように、 T1一力プリル酸了リル 4 0 Μ以上の濃 度では H I V持続産生細胞に対して特異的な増殖抑制の効果を示した。
実施例 7 η—カブリン酸ァリルの効果:
被検薬を η—力プリン酸ァリルとして実施例 1と同様の実験を行った。 その結 果を図 1 3、 図 1 4に示す。
図 1 3、 図 1 4から明らかなように、 η—力プリン酸ァリル 4 0 ί Μ£1上の濃 度では H I V持続産生細胞に対して特異的な増殖抑制の効果を示した。
実施例 8
下記成分を常法により顆粒化した後、 打錠し、 次いでフィルムコートしてジァ リルジスルフィ ド 2 O mg含有フィルムコート錠を得た。

下記成分を常法により混合し、 硬カプセルに充塡してジァリルジスルフィ ド 4 O mg含有カプセルを得た。

産業上の利用可能性
ジスルフィ ド (1 ) 及び不飽和化合物 (2 ) は、 H I V初感染細胞だけでなく、
H I V持続感染細胞に対して強い増殖抑制作用を有し、 A I D Sの治療に有用で ある。

Claims

請 求 の 範 囲
1. 一般式 ( 1 ) :
R1- S - S - R2 ( 1 )
(式中、 R1 は炭素数 2〜20のアルケニル基を示し、 R2 は炭素数 2~20 のアルケニル基又は炭素数 1〜20のアルキル基を示す)
で表わされるジスルフィ ド、 又は
—般式 (2) :
R^O-R3 (2)
(式中、 R1 は炭素数 2〜20の了ルケ二ル基を示し、 R3 は水素原子、 炭素 数 2 ~ 20の了シル基、 リン酸基又はリン脂質基を示す)
で表わされる不飽和アルコール又はそのエステルを有効成分とする抗 H I V剤。
2. 有効成分が、 ジ了リルジスルフィ ド又は脂肪酸了リルである請求項 1記載の 抗 H I V剤。
3. 一般式 ( 1 ) :

(式中、 R1 は炭素数 2〜20のアルケニル基を示し、 R2 は炭素数 2〜20 のアルケニル基又は炭素数 1〜20の了ルキル基を示す)
で表わされるジスルフィ ド、 又は
一般式 (2) :
R'-O-R3 (2)
(式中、 R1 は炭素数 2〜20のアルケニル基を示し、 R3 は水素原子、 炭素 数 2 ~ 20の了シル基、 リン酸基又はリン脂質基を示す)
で表わされる不飽和アルコール又はそのエステルを有効成分とする A I DSの 予防及び治療剤。
4. 有効成分が、 ジァリルジスルフィ ド又は脂肪酸ァリルである請求項 3記載の A I DSの予防及び治療剤。

(式中、 R1 は炭素数 2〜20のアルケニル基を示し、 R2 は炭素数 2~20 のアルケニル基又は炭素数 1 ~ 20の了ルキル基を示す)

(式中、 R1 は炭素数 2〜20のアルケニル基を示し、 R3 は水素原子、 炭素 数 2〜20の了シル基、 リン酸基又はリン脂質基を示す)
で表わされる不飽和アルコール又はそのエステルの抗 H I V剤としての使用。
6. 有効成分が、 ジ了リルジスルフイ ド又は脂肪酸ァリルである請求項 5記載の 抗 H I V剤としての使用。
7. —般式 ( 1 ) :

(式中、 R1 は炭素数 2~20の了ルケ二ル基を示し、 R2 は炭素数 2〜20 の了ルケニル基又は炭素数 1〜20の了ルキル基を示す)
で表わされるジスルフィ ド、 又は

(式中、 R1 は炭素数 2〜20のアルケニル基を示し、 R3 は水素原子、 炭素 数 2 ~ 20のァシル基、 リン酸基又はリン脂質基を示す)
で表わされる不飽和アルコール又はそのエステルの A I DSの予防又は治療剤 としての使用。
8. 有効成分が、 ジ了リルジスルフイ ド又は脂肪酸了リルである請求項 7記載の A I DSの予防又は治療剤としての使用。
9. H I V持続感染細胞に対して一般式 ( 1 ) :
R1 - S-S- R2 (1)
(式中、 R1 は炭素数 2〜20の了ルケ二ル基を示し、 R2 は炭素数 2〜20 のアルケニル基又は炭素数 1~20の了ルキル基を示す)
で表わされるジスルフィ ド、 又は
一般式 (2) :
R'-O-R3 (2) (式中、 R1 は炭素数 2~2 0のアルケニル基を示し、 R3 は水素原子、 炭素 数 2 ~ 2 0のァシル基、 リ ン酸基又はリ ン脂質基を示す)
で表わされる不飽和アルコール又はそのエステルを投与することを特徴とする H I V持続感染細胞の増殖抑制方法。
10. 投与する化合物がジァリルジスルフィ ド又は脂肪酸ァリルである請求項 9記 載の増殖抑制方法。
