技術分野
本発明はビフ ィ ドバク テ リ ゥム菌の増殖を選択的に促 進する増殖促進剤並びにその具体的用途への応用に関す る ものであ る。
背景技術
ビフ ィ ドバク テ リ ゥム属の微生物即ち ビフ ィ ドバクテ リ ウム菌は、 ヒ ト を含む哺乳類の腸管内常在菌であ.り、 それ自体.は病原性がな く 、 しか も乳酸産生や栄養要求性 等の点か ら病原性腸内細菌に拮抗 し、 これらの菌の腸管 内での増殖を妨げる こ とが知 られている。 ま たこ の菌が 腸管内に純培養の よ う に存在する天然栄養児は、 こ の菌 の少ない人工栄養児よ り も腸管内感染におかされに く い こ と が知 られてい る。 こ の よ う な知見を基に近年ビフ ィ ドバク テ リ ゥム菌に関する研究が進み、 こ の菌の免疫賦 活効果、 菌交代症の防止効果、 更には発癌性物質に対す る抑制効果等が明 らかにな り 臨床への応用が試み られて レ、 る。
上述 した よ う な ビフ ィ ドバク テ リ ゥム菌の有する種々 の効果を利用するためには、 こ の菌を腸管内で増殖させ る こ とが必要であ るので、 ビフ ィ ドパク テ リ ゥ厶菌の増 殖を促進する物質 (以下ビフ ィ ズス フ ァ ク タ ー と いう ) に関する研究が盛んに行なわれてい る。 ビフ ィ ズス フ ァ ク タ 一 と して望ま しい条件は、 上部消 化管において消化吸収されずに回腸や大腸へ到達する こ と、 ま た ビフ ィ ドバク テ リ ゥム菌の利用性が高 く 、 他の 菌に利用 さ れに く い こ と と されて い る。 こ の よ う な ビ フ ィ ズス フ ア ク タ ー と して既にい く つか提案され、 中で も ォ リ ゴ糖類を種々の食品へ添加する こ とが広 く 利用さ れて い る。
しか しながら上記条件の観点か ら見た場合、 従来報告 されたォ リ ゴ糖類は、 ビ フ ィ ズス フ ァ ク タ ー と しては不 充分な ものであ る。 例えばフ ラ ク ト オ リ ゴ糖は、 胃酸に よ り 分解されて上部消化管で一部吸収されて し ま う と い う 問題があ る。 またイ ソマル ト オ リ ゴ糖は小腸粘膜酵素 に よ り 加水分解を受けて吸収されるので、 大量の摂取が 必要 と な る。 ガラ ク ト オ リ ゴ糖の場合は酸や熱に対して 比較的安定で上部消化管での消化吸収を受けに く いが、 製法上収率が低 く 製造コ ス ト が高い と い う 欠点を有して い る。 ま たこれ らはいずれも糖類であ るので、 食品に添 加する場合甘味料と して働き、 食味のう ち甘味ばか り が 強調されて し ま う と い う 問題 も あ る。 更にはこれら糖類 の利用能の選択性が低 く 、 ビフ ィ ドバク テ リ ゥム菌以外 の腸内細菌に も利用されるので、 十分な効果を あげるた めには大量の摂取が必要になる と い う 欠点があ る。
本発明は上記の よ う な従来技術の問題点に着目 してな された も のであっ て、 その 目的は、 上部消化管において 消化吸収されず、 ビフ ィ ドバク テ リ ゥム菌の増殖促進効 果に優れ、 しか も選択性の高い ビフ ィ ズス フ ァ ク タ ーを 提供する こ と にあ る。
発明の開示
上記課題を解決する こ と のでき た本発明は、 ダルコ ン 酸、 その無毒性塩お よび Zま たはダルコ ノ デルタ ラク ト ンを有効成分と して含有する こ と に要旨を有する。
本発明者 ら はビフ ィ ズス フ ァ ク タ ーに関 して種々検討 した結果、 ダルコ ン酸、 その無毒性塩お よび/またはグ ルコ ノ デルタ ラ ク ト ンがビフ ィ ドバク テ リ ゥ厶菌の選択 的増殖促進作用を有する と い う 新 しい知見を得、 こ の新 しい知見に基づき、 さ らに研究を続けた結果、 本発明の 完成に至 っ た も のであ る 。 即ち、 本発明者 らがダルコ ン酸の利用能につ い て検討 した結果、 ビフ ィ ドバク テ リ ウム菌の う ち、 特に成人の腸内に優势と される ビフ ィ ド ノヽ*ク テ リ ウム * ァ ド レ セ ンテ ィ ス ( Bifidobacterium adolescentis) に効率よ く 利用され、 一方腸管内に常在 して産褥熱、 虫垂炎、 腸炎、 食中毒の原因菌 と なる ゥェ ルシ ュ菌 ( Clostridium perf ringens) には禾 ij用 されず 逆にその発育を抑制する こ と を見出 した。 ま たダルコ ン 酸 (塩) は腸内優勢菌であ るパク テロ イ デス属の菌には 利用 されないこ と も解明 した。
ダルコ ン酸 (塩) がイ ン ビ ト ロでビフ ィ ドパク テ リ ゥ ム · ァ ド レセ ンテ イ ス に利用され得る こ と は 「パージ一 ズ ' マニュアル ' ォブ ' システマテ ィ ッ ク ' パク テ リ オ ロ ジ一, 第 8 版, ザ · ウ イ リ ア ムズ ' ア ン ド . ウ ィ ルキ ン ス社」 等に記載 さ れて い る、 一方、 ビ フ ィ ズス フ ァ ク タ 一 と しての重要な条件と して、 既述の如 く 上部消化 管で消化吸収されずに大腸に到達する こ と が挙げられる が、 有機酸は一般に小腸で吸収される こ とが知 られてお り 、 ダルコ ン酸 も小腸で吸収されるだろ う と考え られて き た。 ま た、 ダルコ ン酸はグルコ ースに も化学構造的に 類似す る ので、 グル コ ース と 同様小腸で吸収さ れる と 従来考え られて き た。 しか しながら後記する よ う に腸管 ループ法に よ っ て グルコ ースが 1 00 %吸収される条件下 でダルコ ン酸の小腸における吸収性を試験 した結果、 驚 く べき こ と に大部分が小腸に吸収されず、 大腸に到達す る と い う こ と を見出 したのであ る。 有機酸がビフ ィ ドバ ク テ リ ゥム菌増殖促進作用を有する こ と を見出 したのは 初めてであ る。 さ ら にま た、 本発明者ら はダルコ ン酸が いわゆる 「悪玉菌」 であ る ウエルシュ菌の発育を抑制す る効果があ る こ と を初めて見出 した。 この よ う に、 グル コ ン酸は ビフ ィ ドパク テ リ ゥム菌を選択的に増殖促進さ せる と い う 作用を有する。
ビフ ィ ドバク テ リ ゥム菌については、 前記ビフ ィ ドバ ク テ リ ウム · ァ ド レセ ンテ イ スのほか、 ビフ ィ ド ノ クテ リ ウム · シユー ドカテニユ レ一タ ム、 ビフ ィ ド ノ クテ リ ゥム ♦ カテニユ レ一タ ム等の菌がダルコ ン酸を利用 し得 る と レ、 う こ と が知 られているので、 ダルコ ン酸、 その無 毒性塩お よびダルコ ノ デルタ ラ ク ト ンがこれらの菌の増 殖促進作用を有 し得る。 ダルコ ン酸は温和な酸味を有するので食品の酸味料と して用い られ、 ま たそのカルシウム塩は水に よ く 溶ける のでカルシ ウム剤と して食品に添加 した り 医薬品 と して 用い られてい る。 ま た、 ダルコ ノ デルタ ラ ク ト ンは水に 溶解す る と 徐々 に ダルコ ン酸塩に変化 し、 豆腐の凝固 剤、 パ ン等の膨張剤と して用い られて い る。
本発明で用い る ダルコ ン酸の無毒性塩と しては、 人体 に無害であ り 、 且つ ビフ ィ ドバク テ リ ゥム菌の利用性が 良好な も のであれば特に限定 さ れないが、 ナ ト リ ウム 塩、 カ リ ウム塩等のアルカ リ 金属塩のほか、 カルシウム 塩、 マグネ シウム塩、 亜鉛塩、 銅塩等が好ま しい もの と して例示される。
ビフ ィ ドパク テ リ ゥム菌の増殖促進作用を有するダル コ ン酸、 その塩お よび Zま たはダルコ ノ デルタ ラ ク ト ン は、 ヒ ト ま たは動物のビフ イ ドパク テ リ ゥム菌増殖促進 剤、 ク ロ ス ト リ ジゥム菌の増殖抑制剤、 整腸剤、 腸内腐 敗発酵抑制剤、 下痢予防治療剤、 糞便の臭いの軽減剤、 便性状改善剤、 便秘改善剤、 或は更に動物肥育促進剤等 と して用い る こ と もでき る。
本発明の ビフ ィ ドバク テ リ ゥム菌増殖促進剤をは じめ とする各種用途の剤は、 ダルコ ン酸、 その無毒性塩お よ び Zま たはダルコ ノ デルタ ラ ク ト ン を単独で、 或いは他 の ビ フ ィ ドバク テ リ ゥ ム菌増殖促進剤を二種以上混合 して粉剤、 顆粒剤、 錠剤等の所望の形状に加工すればよ い。 ま た用途や適用対象に応 じて液剤 と して提供する こ と も でき る。
更に本発明の ビフ ィ ドバク テ リ ゥム菌増殖促進剤をは じめ とする各種用途の剤には常用される各種添加剤を加 え る こ と も有効であ る。 添加剤と しては、 ア ップルフ ァ ィ バー、 コ ー ンフ ァ イ バー、 ァノレギン酸、 キ ヤ ロ ッ ト ノヽ0 ウダ一、 ぺク チ ン、 海藻多糖類、 カルボキ シメ チルセル ロ ー ス等の食物繊維 ; 乳糖、 でんぷん等の賦形剤 ; 庶 糖、 麦芽糖、 果糖、 ソルビ ト ール、 マ ンニ ト ール、 ステ ビォサイ ド、 アスパルテーム等の甘味料 ; ビタ ミ ン、 ミ ネ ラル、 ミ ルクパウダー、 肉エキス等の栄養補給剤 ; 香 料 ; ア ラ ビア ゴム末、 ポ リ ビュルピロ リ ド ン、 ヒ ドロキ シプロ ピルセルロ ース等の結合剤 ; ス テア リ ン酸マグネ シゥム、 ス テア リ ン酸カルシウム、 タルク等の滑沢剤な ど が挙げられ、 これ ら の中か ら適宜選択 して用いればよ い o
本発明の ビフ ィ ドバク テ リ ゥム菌増殖促進剤お よび各 種用途の剤は、 ダルコ ン酸、 その無毒性塩お よび/また はダルコ ノ デルタ ラ ク ト ンをそのま ま、 或は前記製剤に した も のをそのま ま単独でヒ ト ま たは動物に投与 して も 良い し、 ま た各種食品に添加 して ヒ ト に、 飼料に添加 し て動物にそれぞれ投与 して も良い。
具体的に は、 発酵乳、 清涼飲料水、 シ ャ ーベ ッ ト 、 キ ャ ンデ ィ ー、 ゼ リ ー等の菓子、 豆腐、 食肉 · 魚肉練製 品、 そ の他の食品 (例えば、 各種漬物、 ド レ ッ シ ング 等) に添加 して も良い。 ま た、 糞便の臭いの軽減の 目的 で老人食、 病院食に添加 した り 、 ま た粉剤、 顆粒剤は振 り かけ、 スープの素、 味噌汁の素等に添加 して も良 く 、 ゥ シ、 ブタ、 ト リ 等の家畜やィ ヌ 、 ネ コ等のペッ ト の飼 料添加物と して下痢の予防、 治療、 糞便の臭いの軽減の ために、 あ る いは肥育促進剤と して使用する こ とができ る。 ま たは、 ダルコ ン酸、 その無毒性塩お よび Zまたは ダルコ ノ デルタ ラ ク ト ン を水に添加 して ヒ ト ま たは動物 の飲料水と して用い る こ と も でき る。
吏にま た、 ダルコ ン酸、 その無毒性塩お よび Zまたは ダルコ ノ デルタ ラ ク ト ンは、 酸味料、 カルシウム剤 (グ ノレコ ン酸カルシウム) 、 凝固剤あ る いは膨張剤と して従 来か ら 使用 さ れて い る の で、 こ れ ら の用途を兼ねたビ フ ィ ズス フ ァ ク タ ー と して用いる こ とができ る。
食品 ま たは飼料に添加す る 際の含有量と しては 0. 1 ~ 10重量%の範囲が望ま しい。 ま たダルコ ン酸の無害性は 既に広 く 知 られてい る と こ ろ であ るが、 1 日 の総摂取量 は成人で 0. 1 g Z kg、 乳幼児で 0. 05 g Z kg程度を 目安と する。
ま た、 こ の発明の ビフ ィ ドバク テ リ ゥム菌の増殖促進 作用等各種の効果は、 水中で加水分解されてダルコ ン酸 を生成す る 物質、 例えばダルコ ン酸のエス テル (例え ば、 メ チノレエス テノレ、 ェチノレエス テル等の ァ ノレキ クレエス テル等) 、 ダルコ ノ ガンマラ ク ト ン等、 あ る いはまた D 一ガラ ク ト ン酸、 D —ガラ ク ト ー 1 , 4 ー ラ ク ト ン等で も期待でき る。 ビ フ ィ ドパク テ リ ゥム菌を選択的に増殖させる作用を 有する ダルコ ン酸、 その無毒性塩お よび/ま たはダルコ ノ デルタ ラ ク ト ン を添加 した食品お よび食品素材は機能 性食品お よび機能性食品素材と して有用であ る。
食品お よび食品素材にダルコ ン酸、 その無毒性塩また はダルコ ノ デルタ ラ ク ト ンを含有させる方法と しては、 直接添加する方法と、 食品や食品素材に含まれる成分を 化学的あ る いは生化学的な方法でダルコ ン酸等に変化さ せる方法があ る。
前者の方法を用い る場合には、 目的に よ っ て、 ダルコ ン酸、 その無毒性塩ま たはダルコ ノ デルタ ラ ク ト ンから 適当な ものを選択すればよ く 、 例えば酸味を付加 したい 場合には、 ダルコ ン酸ゃダルコ ノ デルタ ラ ク ト ンなど を 添加すればよ く 、 塩味や味の改善を 目的とする場合には ダルコ ン酸ナ ト リ ウムゃダルコ ン酸カ リ ウムを添加する 方法が考え られる。 又、 後者の方法を用い る場合には、 食品や食品素材にグルコ ースが含まれてい る場合にはグ ルコ ースォキ シダーゼな どの酵素あ る いは、 ダルコ ン酸 生産菌を作用 させて、 グルコ ース をダルコ ン酸に変化さ せる方法があ り 、 シュ ク ロースやデンプン類を含む食品 や食品素材の場合は、 例えばィ ンベルタ一ゼゃァ ミ ラー ゼな どの酵素を用いて、 グルコ ース を生成させた後、 同 様にグルコ ース をダルコ ン酸に変化させる方法があ る。
ダルコ ン酸、 その無毒性塩お よび ま たはダルコ ノデ ノレタ ラ ク ト ン を含有させう る食品お よび食品素材と して は、 前記の食品の ほか甘味料、 ハチ ミ ツ、 ロ ーヤルゼ リ ー、 乳製品、 大豆製品、 食塩、 酸味料、 調味料、 デン プ ン 、 デキ ス ト リ ン 、 魚肉 * 畜肉加工製品、 麵類、 飲 料、 パ ン 、 ケ ー キ、 菓子、 漬物等の塩蔵製品、 pH調整 剤、 氷温貯蔵用凝固点降下剤、 水分活性調整剤、 防腐 剤、 賦型剤、 増量剤な ど あ らゆ る食品お よび食品素材が 挙げられる。 これらの中か ら代表的な ものを選び説明す る。
( 1 ) ハチ ミ ツ
ハチ ミ ッにはダルコ ン酸が少量含まれてい るが、 ダルコ ン酸の ビフ ィ ドバク テ リ ゥム菌の増殖促進の機能を発現 する ためには、 さ ら に濃度を高める必要があ る。 ハチ ミ ッの甘味と ダルコ ン酸の酸味がよ く 調和する こ とか ら、 ハチ ミ ッにダルコ ン酸ゃダルコ ノ デルタ ラ ク ト ンなど を 添加するか、 ハチ ミ ッ中のグルコ ース を酵素的にダルコ ン酸に変え る こ と に よ り 酸味を有する新規なハチ ミ ツが 得 られる。
( 2 ) 甘味剤
異性化糖、 水飴、 黒砂糖、 グルコ ースな どの甘味剤は、 飲料、 キ ャ ンデ ィ ー、 菓子類な ど に広 く 用い られている が、 こ れにダルコ ン酸ゃダルコ ノ デルタ ラ ク ト ンを添加 するかあ る いは、 その成分を一部ダルコ ン酸に変化させ る こ と で機能性を有する酸味を呈する新規な甘味剤と し て使用でき る。 ( 3 ) オ リ ゴ糖類
各種の機能を有するオ リ ゴ糖類が開発されてお り 、 これ ら にダルコ ン酸、 その無毒性塩お よび/またはダルコ ノ デルタ ラ タ ト ンを添加 して、 その機能性を増強する こ と に よ り 、 新規な呈味性甘味剤と して使用でき る。
( 4 ) 乳製品
牛乳や脱脂粉乳は各種の食品の原料と して用い られてお り 、 ダルコ ン酸を添加する こ と に よ り 新規な呈味を有す る機能性食品素材 と して使用でき る。
こ の他に も、 あ ら ゆる食品や食品素材に対 してダルコ ン酸、 その無毒性塩お よび/ま たはダルコ ノ デルタ ラク ト ンの含有が考え られ、 酸味、 塩味の付加、 カルシウム やマグネ シウムの強化な ど、 目的に よ り 選択 して使用で き る。
さ ら に ま た、 ビフ ィ ドバク テ リ ゥム菌の増殖促進作用 を有する グルコ ン酸のアル力 リ 金属塩は上記に加えて次 の よ う な効果を有する。
( 1 ) アスパルテーム と併用 した場合、 アスパルテームの 甘味の改善作用を有する。
( 2 ) 豆腐用凝固剤と併用 して、 豆腐を製造 した場合、 豆 腐の風味、 舌ざわ り を損なわずに維持でき る作用を有す る。
( 3 ) 食塩と併用 した場合、 食塩の味の改善、 保存性等を 低下させる こ と な く 維持でき る作用を有する。 ま た、 ダルコ ン酸は従来か ら使用されてい る酸味剤に 添加する こ と に よ り 、 酸味剤の有する味、 風味、 酸味の 強さ等を損う こ と な く 、 機能性を付加でき る と いう 作用 を有する。
以下、 これら について説明する。
ア ス パルテ ー ム と ダル コ ン酸の ア ル力 リ 金属塩を含有 する機能性甘味剤について :
ァ ス ノヽ0ノレテ ー ム一 ( ひ 一 L ー ァ ス ノヽ0ルチノレ一 L一フ エ 二ルァ ラ ニ ン メ チルエス テル、 以下ア ス パルテ ーム と言 う ) は甘味が砂糖の 100 ~ 200倍程度と強いため食品に用 いた場合.、 エネルギーは低 く ダイ エ ツ ト 甘味料と して各 種飲料やケーキな ど に広 く 用い られてい る。 甘味の質は 砂糖に比較的近い も のの、 甘味の発現がやや遅 く 、 舌に 残 り やすい と い う 欠点があ る こ と か らその改善方法が検 討されてい る。 例えば、 ア ス パルテーム とサイ ク ロデキ ス ト リ ン を併用する方法 (特開昭 60— 114166 ) 、 ァスパ ルテーム と水あめを併用する方法 (特開昭 60— 114167 ) な どが知 られてい る。 しか し、 サイ ク ロデキス ト リ ンや 水あめは消化吸収され、 エネルギー源と なる こ とか らァ スパルテームの特徴であ る低エネルギー性の面か らは好 ま しい方法 と はい えない。
一方、 ダルコ ン酸のアルカ リ 金属塩が合成甘味剤であ るア セ ス ルフ ァ ム K の風味を改善する と い う こ とが知ら れて い るが (特開昭 59— 66857 ) 、 該公報にはダルコ ン 酸のア ルカ リ 金属塩が具体的に どのよ う に風味を改善す るかは記載されていない。
アスパルテームの特徴を損なう こ と な く 、 味の質をで き るだけ砂糖に近づける方法を検討 した結果、 ダルコ ン 酸のアルカ リ 金属塩を併用する こ と に よ り 、 上記課題を 解決でき る こ と を見出 した。 すなわち、 ダルコ ン酸のァ ルカ リ 金属塩は少量の添加でアスパルテームの甘味の質 を改善でき る こ と、 このためアスパルテームにダルコ ン 酸のアル力 リ 金属塩を添加 した甘味剤は低エネルギー で、 しか も ビフ ィ ドバク テ リ ゥム菌を選択的に増殖させ る 作用 を有す る ため健康上に もす ぐれた も のであ り 、 従っ て機能性甘味剤と して有用であ る。
こ の発明の機能性甘味剤は、 甘味剤と してあ らゆる食 品に使用する こ とができ る。
ダル コ ン酸のア ルカ リ 金属塩の添加量は、 ァスパル テーム 1 重量部に対 して当量以上で甘味の改善効果が認 め られるが、 ダルコ ン酸のアルカ リ 金属塩の添加量が多 く なる と塩味を感 じる よ う になる こ と か ら、 使用する食 品に よ っ て添加量を調整する必要があ る。 例えば、 清涼 飲料な ど の塩味が問題となる よ う な食品では、 食品中の ダルコ ン酸のアルカ リ 金属塩の量が 1 %以下と なる よ う にする こ と が望ま し く 、 逆に、 漬物の よ う に甘味と塩味 が同時に必要な食品では、 食品中のダルコ ン酸のアル力 リ 金属塩の量が 1 %を越えて も ま った く 問題と はな らな い o こ の発明の甘味剤の使用方法と しては、 ァスパルテー ム と ダルコ ン酸のアル力 リ 金属塩をそれぞれ別々に食品 に添加 して も良い し、 アスパルテーム と ダルコ ン酸のァ ルカ リ 金属塩を使用する食品に応 じてあ らか じめ混合し て使用 して も 良い。 甘味剤の形態 と して は粉末、 顆粒 状、 液状等常用の ものが挙げられる。 又、 ァスパルテー 厶 と グル コ ン酸のアルカ リ 金属塩の他に例えば、 酸味 剤、 他の甘味剤、 調味料、 賦形剤等各種成分を配合して も良い。
こ の発明の機能性甘味剤は、 アスパルテームの味を低 エネルギーのま までき るだけ砂糖に近づけた ものである ため、 低エネルギー甘味剤と して優れてい るばか り でな く 、 該甘味剤を構成するダルコ ン酸のアルカ リ 金属塩の 作用 と して腸内のビフ ィ ドバク テ リ ゥ厶菌を選択的に増 殖させる効果を有するので、 機能性甘味剤と して有用で め る 。
豆腐用凝固剤にダルコ ン酸のアル力 リ 金属塩を含有さ せた機能性豆腐用凝固剤について :
豆腐は大豆か ら製造 した豆乳に凝固剤を添加 して製造 するが、 凝固剤 と して一般に用い られてい る ダルコ ノ デ ルタ ラ タ ト ンは、 水に溶かす と加水分解されてダルコ ン 酸にな るために添加量が多 く な る と豆腐に酸味が付加さ れる と い っ た問題があ り 、 添加量は 0 . 3 %が限度と され てい る。
国民の健康志向の向上と と も に機能性食品の開発がさ かんに行われているが、 伝統食品であ る豆腐に も新たな 機能性を持たせる こ とができ れば、 食卓でも親 しみやす い食品であ る と い う 点か ら も その意義は大き い。
こ の発明者等は、 前記の よ う にダルコ ン酸等がビフ ィ ドバク テ リ ゥム菌の増殖作用を有 してい る と い う 新知見 に基づき 、 上記課題解決のため鋭意研究の結果、 ダルコ ン酸のアル力 リ 金属塩を豆腐の製造時に添加する こ と に よ っ て風味や舌ざわ り を損な う こ と な く ダルコ ン酸等を 高濃度で含有 し、 機能性を有する豆腐が製造でき る こ と を 見 出 し た。 豆腐凝固剤 と しては、 従来の豆腐用凝固 剤、 例えば、 ダルコ ノ デルタ ラ ク ト ン、 ダルコ ン酸カル シゥム、 硫酸カルシウム、 塩化マグネ シウム等やそれら の合剤が挙げられる。
豆腐用凝固剤にダルコ ン酸のアル力 リ 金属塩を含有さ せた機能性豆腐用凝固剤中の凝固剤 と ダルコ ン酸のアル 力 リ 金属塩 と の配合割合は一般的には凝固剤 1 に対 して 0 . 3 ~ 30程度の範囲で配合すればよ い。
こ のダルコ ン酸のアル力 リ 金属塩を含有させた機能性 豆腐用凝固剤を使っ て豆腐を製造する場合、 該凝固剤の 添加量は豆腐が凝固する量でよ いが、 一般的には豆腐中 のダルコ ン酸量が 0 . 5 % (重量%、 以下同 じ) 以上が好 ま しい。
さ ら に ま た、 豆腐を製造する場合には、 上記凝固剤の ほか、 リ ン ゴ酸ナ ト リ ウム、 クェン酸ナ ト リ ウム等の有 機酸の塩等が併用 される こ とがあ る。 グルコ ン酸のアル力 リ 金厲塩の添加量は特に制限はな いが、 機能性を考慮 した場合、 豆腐中の含量がダルコ ン 酸に換算 して 0 . 5 %以上 と な る よ う に添加する こ とが好 ま しい。 この場合、 凝固剤と して、 グルコ ノ デルタ ラ タ ト ンやダルコ ン酸カルシウムあ るいは、 これ ら を配合 し た も のを用い る場合には、 合計量がダルコ ン酸に換算し て 0 . 5 %以上 と な る よ う にダルコ ン酸のア ル力 リ 金属塩 の添加量を調整すれば良い。
ダル コ ン酸のア ル力 リ 金属塩を添加す る に当たっ て は、 豆腐の製造工程中の任意の時期に添加すれば良 く 、 例えば、 大豆を水と と も に磨砕 した " ゴ" あ る いはゴを 加熱 した後におか ら を除去 して得た豆乳に添加する方法 な どが考え られる。 又、 添加の方法と しては、 単独で添 加 して も良い し、 消泡剤や凝固剤な どの豆腐の製造に用 い る添加物や食品素材と あ らか じめ混合 して添加 して も 良い。
又、 ダルコ ン酸のアルカ リ 金属塩は、 あ ら ゆる豆腐に 使用が可能であ り 、 例えば、 絹ご し豆腐、 充填包装豆 腐、 木綿豆腐、 ソ フ ト 豆腐な どが挙げられ、 この場合に はこれ らの豆腐を製造する 目的で使用されてい る凝固剤 (ダルコ ノ デルタ ラ タ ト ン、 硫酸カルシウム、 塩化マグ ネ シゥム等) な どの添加物はそのま ま使用でき る。
こ の発明の機能性豆腐は、 従来の豆腐にダルコ ン酸の アル力 リ 金属塩等のダルコ ン酸類を添加する こ と によ り 豆腐の風味、 舌ざわ り な ど を損なう こ とな く 、 ビフ イ ド バク テ リ ゥム菌の増殖促進作用 と い う 機能性を付加 した 点で有用であ る。
食塩にダルコ ン酸のアル力 リ 金属塩を添加 した機能性 塩味剤について :
近年の成人病の増加は食生活と深 く 関 り があ る こ とが 指摘されてお り 、 その一つ と して、 高血圧と食塩の摂取 量の因果関係が明 らかにされている。 その対策と して、 食塩の摂取量を少 く するために各種の減塩食品が販売さ れる よ う になっ て き たが、 食塩は食品に塩味を付加す.る 目的だけで使用 されてい る と は限らず、 食品の加工や保 存の面で も大き な役割 り を果た してい る場合が多い。 例 えば、 漬物では減塩に よ り 、 浸透圧の低下や水分活性の 向上を伴い、 加工特性や保存性に悪影響を及ぼすこ とが 問題と なっ てい る。
こ の問題を解決する方法と して、 食塩の不足分を塩化 カ リ ウムで補う 方法や、 食醉ゃ有機酸な ど の酸味剤を併 用する方法な どがと られてい るが、 いずれの場合に も、 食品の味への影響が大き く 、 有効な方法と はいえない。 この よ う な状況か ら、 食塩を使用 した食品の味、 保存性 な ど を変化させる こ と な く 、 食塩の使用量を低減でき る 添加剤の開発が強 く 望まれてい る。
上記の課題の解決のため鋭意研究の結果、 ダルコ ン酸 のアルカ リ 金属塩は塩味は食塩よ り 弱いが、 その質は食 塩に近 く 、 食塩の代替塩と して有効であ る こ と を見出 し た。 又、 前記の よ う にダルコ ン酸等は、 小腸ではほとん ど吸収されずに大腸に到達 して ビフ ィ ドバク テ リ ウ厶菌 を選択的に増殖させる作用を有 してい る こ とが新たに判 明 し、 食塩の代替塩と して有効であ るばか り でな く 、 整 腸作用を併せ もつ点でも有益であ る。 こ の発明は食塩を 使用する食品の塩味の質、 保存性な ど を低下させる こ と な く 、 ビフ ィ ドバク テ リ ゥム菌の増殖作用 と い う 機能性 を有する新 しい塩味剤を提供する ものであ る。
グルコ ン酸のアル力 リ 金属塩の使用量は特に制限はな く 、 食塩の半分以上を代替 して も食品の味に悪影響は及 ぼさないばか り でな く 、 代替率 40 %以上では食塩の味を マイ ノレ ドにする効果も あ り この点でも有効であ る。
使用方法と しては、 食塩と別々 に添加 して使用 して も よ い し、 あ ら か じ め混合 して使用 して も 良い。 又こ の 他、 酸味剤、 調味料、 甘味料、 賦形剤等各種成分を配合 して も良い。
次にダルコ ン酸ナ ト リ ウ厶 と ダルコ ン酸カ リ ウムの塩 味の強さ を示 した。
食品の塩味の強さ はこの値を参考に して添加量を決定 すればよ い。

こ の発明の機能性塩味剤は、 食塩にダルコ ン酸のアル カ リ 金属塩を添加使用する こ と に よ り 、 従来か ら使用さ れて い る食塩の用途にそのま ま使用 して も食品の塩味の 質、 保存性な ど を低下させる こ と な く 、 しか も ビフ イ ド バク テ リ ゥム菌の増殖促進作用 と い う 機能性を付加 した も のであ り 、 機能性塩味剤と して有用であ る。
酸味剤にダルコ ン酸を添加 した機能性酸味剤について : 食品に酸味を付加する物質、 いわゆる酸味剤と して各 種の有機酸が使われてい るが、 それぞれに味、 酸味の強 さ な どの特徴が有 り 、 食品に よ り 使い分け られている。 例えば、 ジュース類にはク ェ ン酸、 キ ャ ンデ ィ ーには酒 石酸、 漬物には醉酸がよ く 使われるな ど それぞれの酸味 剤の特徴を生か した使われ方がされてい る。 又、 食品に よ っ てはい く つかの酸味剤を組み合せて使用する場合も あ る。
最近は、 機能性食品の開発がさ かんに行われて い る が、 従来か ら使用されてい る酸味剤について も これに機 能性を も たせる こ とができ れば酸味剤と してす ぐれた も のにな る。
こ の発明者等は上記課題解決のため鋭意研究の結果、 ダルコ ン酸がビフ ィ ドバク テ リ ゥム菌の増殖促進作用が あ る と い う 新知見に基づき 、 ダルコ ン酸を従来か ら使用 されてい る酸味剤に添加使用する こ と に よ り 、 酸味剤の 有する味、 風味、 酸味の強さ等の特徴を損う こ とな く 、 機能性を付加でき る こ と を見出 し、 さ ら に研究を続けた 結果、 こ の発明を完成 した。
こ の発明は、 従来か ら使用されている酸味剤の一部を ダルコ ン酸に置き かえ る こ と に よ り 、 該酸味剤の味、 風 味、 酸味の強さ等を損う こ と な く 、 ビフ ィ ドバク テ リ ウ ム菌の増殖作用 と い う 機能性を有する新 しい酸味剤およ び該酸味剤を添加 した食品を提供する も のであ る。
こ の発明で使用する ダルコ ン酸と しては、 ダルコ ン酸 自体のほか、 水溶液中でダルコ ン酸にな る物質であれば 使用でき 、 その よ う な例と して、 例えば、 ダルコ ノ デル タ ラ ク ト ン、 グルコ ノ ガンマラ ク ト ン、 ダルコ ン酸ェチ ルエス テル等のダルコ ン酸のアルキルエス テル等が挙げ られるが、 一般的には食品に使用が認め られて い るグル コ ン酸、 ダルコ ノ デルタ ラ ク ト ンが好ま しい。
ま た、 酸味剤 と しては、 従来か ら食品の酸味剤と して 使用 されてい るすべての酸味剤に適用でき 、 好ま しい例 と しては例えば、 ク ェ ン酸、 乳酸、 酒石酸、 リ ンゴ酸、 酢酸、 コハク酸等が挙げられる。
酸味剤の一部を ダルコ ン酸に置き換え る場合、 その割 合は酸味剤の種類、 酸味の強さ (酸味度) に よ り 異な る。 各有機酸について、 ダルコ ン酸に置き換え る こ とが 可能な割合 (すなわち、 従来の酸味剤の味、 風味、 .酸味 の強さ等を損う こ と な く 、 ダルコ ン酸を添加でき る量) の上限値を酸味度で示 した (表 2 ) 。 なお、 こ こ に示し た値は、 ダルコ ン酸を用いた場合を示 してお り 、 他の前 記ダルコ ン酸にな る物質を用い る場合は、 水溶液中での ダルコ ン酸の生成量を基に換算すれば良い。 なお、 参考 までにダルコ ン酸の酸味度を 1 と した時の各種酸味剤の 酸味度を示 した (表 3 ) 。

コ ノヽク酸 3 . 6 次に上記表の見方を ク ェ ン酸を例に説明する。 ク ェ ン酸の酸味度の 40 %をダルコ ン酸に置き換える場 合、 ク ェン酸を 目的量の 60 %添加 し、 目的量の 40 %のク ェン酸をその酸味度に相当する量のダルコ ン酸に置換 し て も 、 ク ェ ン酸 を 単独で使用 した場合に比べ、 味、 風 味、 酸味の強さ など に差が認め られないこ と を示 してい る。 例えば、 0· 5%のク ェ ン酸溶液の酸味度の 40%をグ ル コ ン酸に置 き 換え る場合、 ク ェ ン酸が 0.3%、 ダル コ ン酸が 0.6 % と なる よ う に配合すればよ い。
ダルコ ン酸の使用方法と しては、 各種の酸味剤と別々 に添加 して使用 して も良い し、 あ らか じめ混合 して使用 して も良い。 この場合のダルコ ン酸類の使用割合は酸味 度で上記の表の値以下 と な る よ う にす る こ と が好ま し く 、 この値よ り 多 く 使用 した場合には、 酸味剤の味、 風 味が変化 し、 本来の特徴が生かせな く な る可能性があ る。 又、 い く つ かの酸味剤を組み合せて使う 場合は、 各 々 の酸剤の酸味度に対するダルコ ン酸への置換可能な上 限値を参考に配合する こ とができ る。
次に従来の酸味剤にダルコ ン酸を配合 した機能性酸味 剤の う ち、 最 も好ま しい酸味剤の例を示と、 クェン酸と グルコ ン酸を 10 : 3 ~ 10: 20の割合で含有する酸味剤、 酒石酸と ダルコ ン酸を 10 : 5 ~ 10: 30の割合で含有する 酸味剤お よび乳酸と ダルコ ン酸を 10: 3 〜 10: 15の割合 で含有す る 酸味剤等が挙げ られる。 そ して、 これ らの 酸味剤を添加する食品に応 じて単独あ る いは 2 種以上混 合 し て使用すれば よ い。 例えば、 ジュ ース類、 キ ャ ン ディ ー等にこれ ら酸味剤の 1 種ま たは 2 種以上を混合し た も の を添加す る場合には、 0.5重量%以上を添加すれ ばよ い。 こ の発明の機能性酸味剤は、 従来の酸味剤にダルコ ン 酸を添加使用する こ と に よ り 、 酸味剤の味、 風味、 酸味 の強さ等の酸味剤と しての従来の特徴を損う こ とな く 、 ビフ ィ ドバク テ リ ゥム菌の増殖促進作用 と い う 機能性を 付加 した機能性酸味剤と して有用であ る。
次にこ の発明の試験例お よび実施例を示す。
試験例 1 ぐ腸管ループに よ る ダルコ ン酸の吸収試験〉 本発明例と してダルコ ン酸ナ ト リ ウムを、 比較剤と し てグル コ ー ス を用いて試験を行った。 被験動物はラ ッ ト ( ウ ィ ス タ ー系、 雄 7 週齢、 体重約 280 g ) を 1 群 3 匹 と し、 計 12匹を用いた。
被験ラ ッ ト を 24時間絶食させ、 エー テルで麻酔後開腹 し、 小腸上部 (空腸の ト ラ イ ツ靭帯下部) または小腸下 部 ( 回腸) で約 10cmの ループを形成 した。 ループ部に lOOmMのダルコ ン酸ナ ト リ ウ ム ま たはグルコ ース を溶解 した生理食塩水 0.5mlを注入 し、 閉腹 して 30分間吸収さ せた後ループを摘出 し、 内容液を生理食塩水 20mlで洗い 出 した。
内容液中に残存 してい る ダル コ ン酸量を 「 F—キ ッ ト D—グノレ コ ン酸」 (ベー リ ンガ一 ' マ ンハイ ム ' 山之 内製) を用いて測定 し、 ま たグルコ ー ス量を 「ダルコ 一 ス CIIテ ス ト ヮ コ 一」 (和光純薬製) を用いて測定し、 こ の値を投与時回収量と した。
尚上記ループ摘出時に小腸の隣接部分を余分に切 り取 り 、 添加回収実験用ループ (約 10cm) を作製 した。 そ こ 93/01473

へ供試剤 0.5mlを注入 し、 上記方法 と 同様に回収 し、 グ ルコ ン酸量ま たはグルコ ース量を測定 して これを添加時 回収量 と し、 下式に基づいて各試料の残存率を算出 し た。

表 4 に小腸上部ま たは下部における残存率を示 した。 表 4 か ら明 らかな よ う に、 ダルコ ン酸は小腸で約 80〜 90 %残存 してお り 、 こ のこ と か ら小腸では一部 しか消化吸 収されないこ とが判る。 これに対 してグルコ ースは小腸 上部で完全に吸収されて し ま う ので大腸にはほと んど到 達でき ず、 ビフ ィ ドバク テ リ ゥム菌を増殖させる効果は 期待でき ない。 試験例 2 < ダルコ ン酸塩の選択的利用性 >
表 5 に示す様に、 ビフ ィ ドバク テ リ ゥム菌の代表と し て ビフ ィ ドバク テ リ ゥ ム · ァ ド レセ ンテ ィ ス ( Bif idoba cterium adolescentis ATCし 15703、 Biiidobacterium adolescentis ATCC 15705) を、 ま たレ、わゆる悪玉菌の 代表 と し て ウ エノレ シ ュ菌 ( Clostridium perf ringens GKK 16、 Clostridium perf ringens CWiu) 及び腸内優勢 菌 と してノヽ♦ク テ ロ イ デス属の菌 ( Bacteroides iragilis W-7) を用いた。 供試菌株を G A Mブイ ヨ ン培地 (ニッ ス ィ 製) に接種 し、 37'C、 20時間嫌気培養 して前培養菌 液と した。
供試剤と してはダルコ ン酸ナ ト リ ウム、 フ ラ ク ト オ リ ゴ糖、 グルコ ース を用い (表 5 参照) 、 糖分解試験用培 地は、 GAM糖分解用半流動培地 (二 ッ ス ィ 製) の 1 / 2 処方か ら 寒天 を 除去 した培地を基礎培地 と し、 上記供 試剤を 0.5WZ V %添加 し、 ρΗ6· 9に調整 した も のを用い 該試験用培地に前培養菌液を 0.01V/V%接種 し、 37 で 、 20時間嫌気培養 して濁度 ( 660nm) 及び pHを測定し た。 培地は直径 18mmの試験管に 5 mlずつ分注 した。 嫌気 培養はァネ ロパッ ク (三菱ガス化学製) を使用 した。 濁 度の測定は島津 ミ ル ト ン · ロ イ 分光光度計" スぺク ト ロ ニ ッ ク 20 A " を使用 し、 試験管のま ま測定 した。 PHの測 定は pHメ ー タ ー HM— 30S (東亜電波工業製) のガラス電 極で直接測定 した。 基礎培地を用いたコ ン ト ロ ールと比 較 して濁度を増加させるか或いは PHを低下させた場合、 そ の供試剤が利用 さ れる と 判定 した。 結果を表 5 に示 す。
ま た、 代表的な ¾株と して Bi f i dobac te ri um adolesce ntis ATCC 15703を選んで、 グノレ コ ン酸カノレシゥ ムに関 して試験 した結果を表 6 に示す。

表 5 及び表 6 か ら 明 ら かな よ う に、 本発明に係る ダル コ ン 酸ナ ト リ ゥ ム 及びダル コ ン酸カ ルシ ウ ム は いずれ も、 ビフ イ ドバク テ リ ゥム菌に よ く 資化され、 ま たグル コ ン酸ナ ト リ ウムは病原菌 と もなる ウエルシュ菌には資 化されず、 む しろ増殖を抑制する こ とが分かる。 さ らに また、 ダルコ ン酸ナ ト リ ウムは腸内優势菌であ るパク テ ロ イ デス属の菌には利用されず、 本発明の増殖促進剤が 選択的な増殖促進剤と して優れてい る こ とが判かる。 一 方、 グルコ ース はビフ ィ ドバク テ リ ゥムの資化性は強い ものの本発明例のよ う な選択性は認め られない。 また試 験例 1 か ら明 らかな よ う にグルコ ースは小腸上部でほぼ 完全に吸収されて し ま う ので、 大腸に到達 し得ず、 ビフ ィ ズス フ ァ ク タ ー と しては不適当であ る こ と が分かる。 試験例 3 ぐ ヒ ト の糞便菌叢に及ぼすダルコ ノ デルタ ラク ト ン投与の影響 >
本発明例と してダルコ ノ デルタ ラ ク ト ン (ダルコ ン酸 のラ ク ト ン) を、 ヒ ト に投与 して、 腸内菌叢と便性に及 ぼす影響について試験を行っ た。
健康成人男性 ( 50歳) 1 名に、 1 日 当 り 9 g のダルコ ノ デルタ ラ ク ト ン粉末を 1 日 3 回に分けて摂取させ、 摂 取 0 日、 摂取 10日、 摂取 30日 お よび摂取終了後 20日休止 した時点での糞便中の腸内菌叢を、 光岡の方法 (光岡知 足 : 「腸内菌の世界」 , 叢文社, 束京, P 53 ( 1984 ) ) に準 じて測定 した。 その結果を表 7 に示す。 ま た糞便の 性状 と して硬さ, 色, 臭いを被験者自身が毎日官能で観 察 した。 糞便 1 g 当た り の菌数の対数値 ( )内は全体に対する比率(% ) 属 名

約 10か月齢の ビーグル犬雄 8 頭 (体重 10〜 13kg) を 1 群 4 頭の 2 群に分け、 ダルコ ノ デルタ ラ ク ト ン投与区 と 、 ダル コ ノ デルタ ラ ク ト ン を投与 しない対照区 と し ダルコ ノ デルタ ラ ク ト ンはゼラチ ンカプセルに入れ、 SOnigZ kg体重 Z回で 10時、 13時、 16時の 1 日 3 回'、 2 週 間投与 した。 投与前 と 2 週間投与後に、 投与区と対照区 の新鲜糞便を採取 し、 パネ ラーに よ る糞便臭気の比較を 行っ た。
その結果、 ダルコ ノ デルタ ラ ク ト ン投与区で瀵便臭気 の低減が観察された。
試験例 5 ぐ甘味剤官能検査 >
ア ス パルテ ー ム にダルコ ン酸ナ ト リ ゥムを添加 した時 の甘味の改善度を官能検査に よ り 比較 した。
官能検査の方法

実施例 6 (オ リ ゴ糖)
イ ソマル ト オ リ ゴ糖 (昭和産業、 イ ソマル ト 500 ) 1 50 g に水 3 50 g を加え、 これに、 実施例 2 で用いた酵素剤 0 . 5 g を添加 し、 23で、 通気条件下で 90分間反応させ、 ダルコ ン酸を含有するオ リ ゴ糖液を得た。 こ のオ リ ゴ糖 液中のダルコ ン酸を高速液体ク ロマ ト ダラ フ ィ 一によ り 定量 し、 表 14の結果が得 られた。 この場合も実施例 3 と 同様ォ リ ゴ糖の製造工程で酵素に よ り ィ ソマル ト オ リ ゴ 糖を生成させる反応を行っ た後上記の反応を行い濃縮す る方法 も考え られる。 又、 この他、 フ ラ ク ト オ リ ゴ糖、 ガラ ク ト オ リ ゴ糖、 乳果オ リ ゴ糖類な ど も 同様の方法で ダルコ ン酸を含有するオ リ ゴ糖とする こ とができ る。

脱脂粉乳 20 g に水 180 g を加え、 これにダルコ ン酸カ ルシ ゥム 2 g と ダルコ ン酸ナ ト リ ウム 3 g を添加 した。 こ の方法で、 加熱 して も沈澱物の生成 しない安定した力 ルシゥ ム強化乳飲料が製造でき た。
実施例 9 (牛乳凝固物)
牛乳 100 g に砂糖 10 g と グルコ ノ デルタ ラ ク ト ン 3 g を 加 え、 80 C で 30分間加熱 して、 牛乳を凝固 させ、 デ ザ一 ト 用牛乳凝固物を得た。
実施例 10 ( リ ン ゴ果汁飲料)
市販の 100% リ ン ゴ果汁に水、 ク ェ ン酸、 グラニュー 糖、 ダルコ ン酸ナ ト リ ウムを加えて リ ンゴ果汁飲料を調 製 した (組成は下記の通 り ) 。
本品は ダル コ ン酸ナ ト リ ゥ 厶 を加 えない も の に比べ て味は濃厚であ り 、 100% リ ン ゴ果汁に近い品質となつ 100% リ ン ゴ果汁 20g グラニュ ー糖 8 g クェン酸 0.32 g ダルコ ン酸ナ ト リ ウム 1.08g 水 80 g 実施例 11 (豆腐)
ー晚水に浸漬 し、 充分吸水させた大豆 200 g に水 350ml を加え、 ミ キサーで磨砕し、 ゴを調製する。 こ のゴに蒸 気を吹 き 込みなが ら加熱 し、 100°C に達 した後、 さ らに 3 分間加熱 し、 直ち に、 こ し布でおか ら を分離 し、 豆 乳を得る。 こ の豆乳にダルコ ン酸ナ ト リ ウムを 0.6%、 1.0% と な る よ う に添加 した後、 ダルコ ノ デルタ ラ ク ト ン を 0.3% と な る よ う に添加 し、 80 °C の湯浴で 30分間加 熱 し、 豆腐を製造 した。
結果は表 16に示す通 り であ る。
ダルコ ン酸ナ ト リ ウムを添加 した豆腐は無添加の豆腐 に比べて " コ ク " があ り 、 味は良好であっ た。 なお、 豆 腐のかた さ、 舌ざわ り な ど に、 ほ と ん ど差は認め られな かっ た。 表 16

実施例 13 (豆腐)
実施例 11と 同様の方法で調製 した豆乳にダルコ ン酸ナ ト リ ウム を 1.0% と な る よ う に添加 した後、 ダルコ ノデ ルタ ラ ク ト ン 67%、 硫酸カ ル シ ウ ム 33%か らな る凝固剤 0.30お よ び 0.34% を添加 し 80°C の湯浴で 30分間加熱 し て、 豆腐を製造 した。 対照にダルコ ン酸ナ ト リ ウムを添 加 しない豆乳に同 じ組成の凝固剤を 0.30%加え同様に豆 腐を製造 した。
結果は表 18に示す通 り であ る。
ダルコ ン酸ナ ト リ ウムを添加 した豆腐は、 やや軟 く な るが、 凝固剤を 1 割程度増すこ とで、 かたさ は対照と同 程度 とする こ と ができ た。

実施例 14 (凝固剤)
ダルコ ン酸ナ ト リ ゥ厶 3 kgにダルコ ノ デルタ ラク ト ン 1 kgを混合 し機能性豆腐用凝固剤 4 kgを製造 した。
実施例 15 (凝固剤)
ダル コ ン 酸 カ リ ゥ 厶 1 kgにダル コ ノ デノレタ ラ ク ト ン 300 g を混合 し、 機能性豆腐用凝固剤 1.3kgを製造 した。 実施例 16 (凝固剤)
ダルコ ン酸ナ ト リ ウ 厶 3 kgにダルコ ノ デルタ ラ ク ト ン 600 g 、 硫酸カルシウ ム 400 g を混合 し機能性豆腐用凝固 剤 4 kgを製造 した。
実施例 17 (豆腐)
実施例 11と 同様の方法で調製 した豆乳に実施例 15で製 造 し た機能性豆腐用凝固剤を 1. 3% と な る よ う に添加 し、 80°C の湯浴で 30分間加熱 し、 豆腐を製造 した。 製造 した豆腐は味が良好で、 かたさ、 舌ざわ り も ま った く 問 題はなかっ た。
試験例 6 (塩味剤官能検査)
食塩単独 と食塩の一部をダルコ ン酸ナ ト リ ゥムで置き 換えた塩味剤を調製 し、 両者の塩味の強さ を同一に した 水溶液の味を官能検査で評価 した。
(官能検査の方法)
食塩の一部を ダルコ ン酸ナ ト リ ウ ム ( GNA) で置き換 えた塩味剤を 5 %濃度にな る よ う に調製 した水溶液と、 これと 同 じ塩味度の食塩溶液を調製 し両者の味の違いを 三点比較法に よ り 評価 した。
結果を次表に示す。

試験例 7 (酸味剤)
酸味剤と して ク ェ ン酸、 乳酸、 酒石酸、 リ ンゴ酸、 酢 酸、 コ ハク酸を選び、 それぞれ単独と酸味剤の一部又は 全部をダルコ ン酸に置き換えた酸味剤の酸味度を同一に し、 両者に官能的に差を認めないダルコ ン酸の最大配合 割合を酸味度を基準に して求めた。
官能検査の方法 :
酸味剤単独 ( A ) と酸味剤の一部又は全部をダルコ ン 酸で置き換えた酸味剤 ( B ) の酸味度を同一に し、 酸味 剤 と酸味剤の一部又は全部を ダル コ ン酸に置き換えた も の の間に官能的に差が認め られるかど う かを三点比較法 で評価 した。 パネル数 1 0 名 結果 : 結果を次表に示す 表 21

実施例 19 (酸味剤) 市販の 1 00 % オ レ ン ジ果汁に水、 グラニュー糖、 酸味 剤 と して ク ェ ン酸と ダルコ ン酸を加え、 オ レ ン ジジユ ー ス果汁飲料を調製 した。 比較例と して、 酸味剤と して実 施例 と 同 じ酸味度 と な る よ う にクェン酸を用いて同様に オ レ ン ジ果汁飲料を調製 し、 両者の味を官能検査 (三点 比較法) で評価 した。 両者の飲料の味に有意な差は認め られなかっ た。 実施例の組成

厚手の鍋にグラニュー糖、 水飴、 水を入れ、 弱火で加 熱す る 。 沸 と う し て き た ら、 フ タ を して、 さ ら に加熱 し、 液温が 1 50 °C になっ た ら水に溶か した酒石酸 と グル コ ン酸か らな る酸味剤を加え火を止める。 これをサラ ダ 油を塗っ たバッ ト に移 し、 ヘラ を使っ て飴を押さ えなが ら均一に冷却する。 飴が冷えて き た ら適当な大き さ に切 断 し、 丸 く 成形 し、 飴を製造 した。 比較例 と して、 酸味 剤 と して実施例 と 同 じ酸味度とな る よ う に酒石酸を用い て同様に飴を製造 した。 両者の飴には味、 舌ざわ り と も に差は認め られなかっ た。

通常の方法で飲飯 した米飯に食酢、 ダルコ ン酸、 食 塩、 グ ラ ニ ュ ー糖 を あ ら か じめ混合 した合わせ酢を加 え、 均一に混合 し、 冷却 してち ら し寿司用の寿司飯を調 製 した。 比較例 と して、 酸味剤と して実施例と 同 じ酸味 度 と な る よ う に食酢を用い同様に寿司飯を調製 した。 両 者の寿司飯には、 味、 風味、 舌ざわ り 等、 官能的に有意 な差は認め られなかっ た。 実施例の仕込量 親 3 0 0 g

1 . 6 g 試験例 8 ぐ ヒ ト の糞便菌叢に及ぼすダルコ ン酸カルシゥ ム投与の影響 >
本発明例と してダル コ ン酸カ ル シ ウ ムを、 ヒ ト に投与 して、 腸内菌叢に及ぼす影響について試験を行っ た。 健康成人男性 ( 28才 ~ 56才) 6 名 に、 1 日 当 り 1 . 7 g のダル コ ン酸カ ル シ ウ ム粉末を 1 日 1 回 (昼食後) 摂取 させ、 摂取開始 1 週間前、 摂取開始日、 摂取 1 週間目お よび 2 週間 目、 摂取終了後 1 週間お よび 2 週間休止 した 時点での糞便中の腸内菌叢を、 試験例 3 と 同様の方法で 測定 した。 その結果を表 22に示す。 表 22 (その 1 )

産業上の利用可能性
本発明の ビフ ィ ドバク テ リ ゥム菌増殖促進剤は選択的 な ビフ ィ ドバク テ リ ゥム菌増殖活性を示 し、 ビフ ィ ズス フ ァ ク タ 一 と して優れた特性を有 してい る。 従って本発 明の ビフ ィ ドバク テ リ ゥム菌増殖促進剤は単独であるい は各種食品や飲料に添加 して各機能性食品や飲料 と し て用 い る こ と がで き 、 健康増進の点か ら極めて有益であ る。

請 求 の 範 囲
1 ダル コ ン酸、 そ の無毒性塩お よびノま たはダルコ ノ デルタ ラ ク ト ン を含有す る こ と を特徴 と す る ビフ ィ ドバ ク テ リ ウ ム菌の増殖促進剤。
2 ダルコ ン酸、 その無毒性塩お よ び ま たはダルコ ノ デルタ ラ ク ト ン を含有す る整腸剤。
3 ダルコ ン酸、 そ の無毒性塩お よ び/ま たはダルコ ノ デルタ ラ ク ト ン を含有す る糞便の臭いの軽減剤。
4 ダルコ ン酸、 そ の無毒性塩お よ び/ま たはダルコ ノ デルタ ラ ク ト ン を含有す る下痢予防治療剤。
5 ダル コ ン酸、 そ の無毒性塩お よ び ま たはダルコ ノ デルタ ラ タ ト ン を含有す る腸内腐敗発酵抑制剤。
6 ダルコ ン酸、 その無毒性塩お よ び ま たはダルコ ノ デルタ ラ ク ト ン を含有す る便性状改善剤。
7 ダルコ ン酸、 その無毒性塩お よ び/ "ま たはダルコ ノ デル タ ラ ク ト ン を含有す る便秘改善剤。
8 ダルコ ン酸、 その無毒性塩お よ び ま たはダルコ ノ デルタ ラ ク ト ン を含有す る ク ロ ス ト リ ジゥ ム菌の増殖抑 制剤。
9 豆腐用凝固剤にダルコ ン酸のアルカ リ 金属塩を含有 させた機能性豆腐用凝固剤。
1 0 ダル コ ン酸、 そ の無毒性塩お よ び Zま たはダルコ ノ デル タ ラ ク ト ン を グル コ ン酸に換算 して 0 . 5 %以上含有 す る機能性豆腐。 11 食塩 と ダルコ ン酸のアルカ リ 金属塩 と を含有する機 能性塩味剤。
12 アスパルテーム と ダルコ ン酸のアルカ リ 金属塩 と を 含有す る機能性甘味剤。
13 ク ェ ン酸 と ダルコ ン酸を 10 : 3 〜: 10 : 20の割合で含 有す る酸味剤、 酒石酸 と ダルコ ン酸を 10 : 5 ~ 10 : 30の 割合で含有す る酸味剤お よ び乳酸 と ダルコ ン酸を 10 : 3 〜 10 : 15の割合で含有す る酸味剤か ら選ばれた酸味剤。 14 ク ェ ン酸 と ダルコ ン酸を 10 : 3 〜: 10~ 20の割合で含 有す る酸味剤、 酒石酸 と ダルコ ン酸を 10 : 5 〜 10 : 30の 割合で含有す る酸味剤お よ び乳酸 と ダルコ ン酸を 10 : 3 〜 10 : 15の割合で含有す る酸味剤か ら選ば^た 1 種ま た は 2 種以上の酸味剤 0.5重量%以上を 含有す る ジュース 類ま たはキ ャ ンデ ィ ー。
15 ダルコ ン酸、 その無毒性塩お よ び/ま たはダルコ ノ デルタ ラ ク ト ン を人に投与す る こ と を特徴 と す る ビフ ィ ドバク テ リ ゥ ム菌の増殖促進法。
16 ダルコ ン酸、 その無毒性塩お よ び/ま たはダルコ ノ デル タ ラ ク ト ン を 人 に投与す る こ と を特徴 と す る 整腸 法。
