Abstract

Die vorliegende Erfindung betrifft Arzneimittel und Diagnostika sowie diagnostische, therapeutische und vorbeugende Verfahren zur Behandlung von Autoimmun- und Viruskrankheiten des Menschen, die durch pathogene Bakterien hervorgerufen werden. Diese Bakterien sind insbesondere Mikroorganismen, die die Schleimhäute des Menschen kolonisieren und bestimmte Exoproteine sezernieren, die strukturelle Verwandtschaften zu menschlichen Proteinen aufweisen. Besondere Bedeutung kommt dabei den IgA-Protease-ähnlichen Proteinen zu, die besonders gut von Zellen aufgenommen werden und als Antigene auf MHC-Molekülen präsentiert werden. Besonders ausgeprägte Homologie zu menschlichen Proteinen besitzt das Polyprotein der IgA-Protease (IPP) von pathogenen Neisserien. Beispielsweise besitzt ein bestimmtes Peptid des IPP ausgeprägte Homologie zu den Gelenkproteinen Link und Aggrekan. So wird z.B. der Nachweis geführt, daß das IPP ein ätiologisches Agens der rheumatoiden Arthritis (RA) ist. Die Autoimmunreaktion wird ferner durch andere Eigenschaften der IPP-produzierenden Neisserien und der IPPs begünstigt. Eine besonders wichtige Eigenschaft ist die Fähigkeit des IPP, Viren und virale Elemente zu aktivieren. Dies betrifft insbesondere die Aktivierung von proviralen Retroviren und endogenen Retroviren des Menschen. Aufgrund der Aktivierung von Viren und viralen Elementen werden weitere Autoimmunreaktionen des Menschen induziert. Durch die IPP-abhängige Aktivierung des HIV-Provirus wird darüber hinaus auch die Entwicklung des acquired immune deficiency syndrome (AIDS) erklärt. Aus den erfindungsgemäßen Eigenschaften der pathogenen Neisserien und der von ihnen gebildeten IPPs leiten sich zahlreiche neue diagnostische, therapeutische und vorbeugende Verfahren ab.
