Lーリボースの製造方法 技術分野 本発明は、 安価な糖を出発原料とした多段階の生物反応による L—リボースの 製造方法に関する。 従来の技術 近年、 非天然型の糖類は医薬及び農薬の中間原料として注目されている。 リボ —スに関しては天然型の D体は D N Aの構成糖としては勿論、 種々のビタミンゃ 補酵素の構成糖として全生物界に豊富に分布しているにもかかわらず、 非天然型 の L体はほとんど供給の目処が立っていないのが現状である。 現在知られている Lーリボースの主な生産方法としては、 L—ァラビノースを原料としたコバルト 触媒による合成法が挙げられる。 一方、 微生物を用いて L—リボースを生産する 方法としてはァシネト/ クタ一 ·力ノレコアセティカス (Acinetobacter calcoace ticus ) D L— 2 8株の産出する酵素が L—リブロースを異性化し、 L—リボー スを生産することが唯一報告されている (Journal of Fermentation and Bioeng ineering Vol. 81, No. 6, 493-497. 1996) 。
上記の酵素反応の原料である L—リブロースの製造方法としては、 Lーァラビ ノースイソメラーゼによる Lーァラビノースの異性化による方法 (Methods Enzy m. Anal. (3rd Ed. ) (1984) vol. 6 442-449) と、 精製されたリビトールを原料とし た微生物反応により製造する方法 (Biochem. J. vol. 64 : 385, 1956) が知られて いるが、 両方法とも原料である L—ァラビノースとリ ビトールは安価とは言い難 く、 工業的規模で安定的に Lーリブロースを供給するプロセスまでは報告されて いない。
上記 2通りの Lーリブロース生産方法の一方の原料であるリ ビトールの微生物 による製造方法として D—リボースを原料として酵素反応により製造する方法
(特公昭 4 5 - 2 0 7 1号公報、 特公昭 4 9 - 1 2 7 1 8号公報) 、 グルコース などの発酵により製造する方法 (特公平 6 - 3 0 5 9 1号公報、 特公平 6 - 3 0 5 9 2号公報、 特公平 6— 3 0 5 9 3号公報、 特公平 6— 3 0 5 9 4号公報) 、 及ぴ藻類の炭酸固定による方法 (特公昭 4 4一 1 6 3 5 0号公報) が報告されて いる。
しかしながら、 上記のいずれの技術を組み合わせても、 工業的規模で L—リボ —スを生産するに至ってはおらず、 従って、 工業的規模で L—リボースを製造し、 かつ生産された Lーリボース含有液から Lーリボースを高純度、 高収率で単離精 製する技術は全く知られていなかった。
実際に従来の方法を用いて工業的規模で Lーリボースを生物学的に生産しょう とした場合、 種々の問題点が生じる。 つまり第 1段階である、 リ ビトールを製造 するに当たって、 従来法の中では安価な原料であるグルコースなどの発酵による 方法が望ましいが、 この従来法でもエリスリ トールなどの他の糖アルコールは効 率的に製造できても、 リビトールに関しては必ずしも満足のいく生産性を得られ てはいない。
第 2段階である、 リビトールを L—リブロースに変換する技術に関して、 従来 法では原料のリビトールは精製されたものでなければならなかった。 結晶リビト —ルは原料としては極めて高価である。 また上述の発酵リビトールを利用する場 合には、 エリスリ トールゃグリセロールといったポリアルコールが多く含まれ、 この中から収率よく リ ビトールのみを分離精製することは極めて困難であった。 そこで、 工業的に安価に L一リブロースを生産するために、 リ ビトールを含有す る発酵液または粗精製リビトールからリビトールを精製することなく直接 L—リ ブロースに変換させる方法が望まれていた。
また、 リビトールの代わりに L—ァラビノースを原料とする場合であっても、 Lーァラビノースは安価とはいえず、 Lーリブロースの収率も低い。
第 3段階である、 L—リプロ一スを L—リボースに変換する反応においても、 従来の技術では原料の L—リブロースは精製されたものである必要があった。 精 製 L—リブロースは極めて高価な原料であり、 上述の Lーリブロース含有液から 単離精製することは極めて困難であった。 そこで目的物質である Lーリボースを
工業的規模で安価に生産するために、 リビトールを含有する発酵液または粗精製 リ ビトールからリ ビトールを精製することなく直接 L一リブロースに変換させ、 さらにその L—リブロース含有反応液から Lーリブロースを精製することなく直 接 Lーリボースに変換する方法が望まれていた。
一方、 多段階生物反応による夾雑物の極めて多い L—リボース含有液からの L 一リボースの単離精製法は全く知られていないため、 有効な方法を開発する必要 がめつた。 発明の開示 本発明者らは、 上述の問題を解決すべく鋭意検討した結果、 安価な原料である グルコース等を出発物質として用い、 数段階の微生物反応と精製を組み合わせる ことにより、 重要な医農薬中間体である非天然型の L _リボースを安価に効率よ く製造できる方法を見出し、 本発明を完成するに至った。
即ち本発明の要旨は、 グルコース等の糖質原料を出発原料とし、 数段階の生物 学的手法を用いて; L—リボースを生産することを特徴とする L—リボースの製造 方法に存する。 具体的には、 本発明は、 グルコース等の糖質原料からリビトール を生産する能力を有する微生物を用いてグルコースからリビトールを生成するェ 程 (以下、 「第 1工程」 と称することもある) と、 リビトールから L—リブ口一 スを生産する能力を有する微生物を用いてリビトールから L—リブロースを生成 する工程 (以下、 「第 2工程」 と称することもある) と、 L—リブロースから L 一リボースを生産する能力を有する微生物を用いて L—リブロースから L—リボ —スを生成する工程 (以下、 「第 3工程」 と称することもある) を含む、 L—リ ボースの製造方法である。
さらに、 本発明の好ましい態様として、 上記の生物学的手法を用いて生産した L—リボース含有液をゲル型濾過材に接触させること、 及び、 該 L—リボース含 有液に有機溶媒を添加し、 反応液中に残存する未反応の Lーリブロースを析出さ せずに L—リボースを単独で析出させることを含む方法が挙げられる。
以下、 本発明を詳細に説明する。
本発明は、 グルコース等の糖質原料を出発原料とし、 数段階の生物学的手法を 用いて L—リボースを生産することを特徴とするが、 上記で延べた第 1工程、 第 2工程及び第 3工程を経る方法がその一例として挙げられる。 以下はその方法を 例として説明するが、 本発明はこれに限定されるものではない。
本発明の製造方法における第 1工程は、 グルコース等の糖質原料からリ ビトー ルを製造する工程である。 第 1工程で使用する微生物としては、 グルコース等の 糖質原料からリビトールを生産する能力を有する微生物であれば、 特に限定され ないが、 好ましくはトリコスポロノイデス(Trichosporonoides) 属に属する微生 物が挙げられ、 さらに好ましくは、 トリコスポロノイデス .マディダ (Trichosp oronoides. madida) 、 卜ジ コスポロノィデス · ニグレスセンス (Trichosporonoi des. nigrescens) 、 卜リ コスポロノイデス - エドセファリス (Trichosporonoide s. oedocephalis) 、 卜リ コスポロノイデス , メガチリェンシス (Trichosporonoi des. megachillensis) 、 卜 リコスポロノイデス · ス / チュラータ (Trichosporon oides. spathulata) の種に属する微生物が挙げられ、 さらに好ましくは、 トリコ スポロノイデス ' エドセファリスと ト リ コスポロノイデス . メガチリェンシスの 種に属する微生物が挙げられる。 例えばトリコスポロノイデス ·マディダに属す る微生物として C B S 240. 79カ 、 トリ コスポロノイデス .ニグレスセンス に属する微生物として CB S 26 8. 81、 CB S 269. 8 1が、 トリコスポ ロノイデス ' エドセフアリスに属する微生物として C B S 568. 85、 CB S 64 9. 6 6が、 トリコスポロノイデス · メガチリェンシスに属する微生物とし て CB S 56 7. 85が、 トリコスポロノイデス ' スパチュラ一タに属する微生 物として CB S 24 1. 79、 C B S 24 2. 79A、 CB S 242. 79 Bが 挙げられる。 これらの菌抹は、 Cenatraalbureau voor Schimmelcultures力 ら入 手することができる。 なお、 上記微生物は、 野生株の他、 UV照射、 N—メチル — N' —ニ トロソグァ二ジン (NTG) 処理、 ェチルメタンスルホネート (EM S) 処理、 亜硝酸処理、 ァクリジン処理等による変異株、 あるいは細胞融合もし くは遺伝子組換え法などの遺伝学的手法により誘導される組換え株などのいずれ の株であってもよい。 変異株の具体例としてはトリコスポロノイデス · メガチリ

ェンシス M C I 3 4 4 2株が挙げられる。 同株は、 平成 9年 6月 2日に、 通産省 工業技術院生命工学工業技術研究所 (郵便番号 305-8566 日本国茨城県つくば市 東 1-1-3) に F E R M P - 1 6 2 5 4の受託番号で寄託され、 平成 9年 1 1月 1 9日にブダペスト条約にもとづく国際寄託に移管され、 F E R M B P— 6 1 7 6の受託番号が付与されている。
第 1工程においては、 上記微生物は 1種あるいは 2種以上が用いられる。
次に、 本発明の第 1工程について具体的に説明する。
第 1工程において、 微生物は糖質原料を炭素源として定法通り培養される。 つ まり、 微生物は 6 0 % (W/V) 以下、 好ましくは 2 0〜 4 5 %の糖質原料を含む 培地に植菌される。 糖質原料としては、 グルコース、 フルク ト一ス、 シュ一クロ ース、 マルトース、 グリセロール等、 及びそれらの混合物が挙げられる。 これら の中ではグルコースが好ましい。 用いる糖質原料の形態は特に制限されず、 粉体、 糖蜜、 廃糖蜜等を用いることができる。 また、 炭素源としては微生物が資化しう るフルク ト一スなどの炭水化物、 グリセロールなどのアルコール類、 有機酸など を適宜加えることができる。 さらにこの培地には微生物が資化しうる窒素源が含 まれる。 窒素源としては、 酵母エキス、 コーンスティ一プリカ一、 N Zァミン、 トリプト一ス、 ペプトン、 ポリペプトン、 魚肉エキス、 肉エキスその他の有機窒 素源、 あるいは硝酸ナトリウム、 その他の無機窒素源、 好ましくは、 酵母エキス やコーンスティ一プリカ一、 が適宜使用される。 培地中に含まれる窒素源の初発 濃度は好ましくは、 酵母エキスの場合 0 . 5〜 4 . 0 %、 コーンスティープリカ 一の場合、 1 . 0〜 8 . 0 %である。 好ましい窒素源の濃度はグルコース等の糖 質原料の濃度に影響される。 上述の炭素源、 窒素源のほかに、 無機イオンゃビタ ミン類を必要に応じ添加することは有効である。 無機イオンとしては、 リン酸ィ オン、 マグネシウムイオン、 鉄イオン、 マンガンイオン、 モリブデンイオンその 他が用いられる。 ビタミン類としては、 チアミン、 イノシトール、 パントテン酸、 ニコチン酸アミ ドなどが挙げられる。 また、 上述のグルコースその他の炭素源、 窒素源、 無機イオン、 ビタミン類は、 必要に応じて培養中の適当な時点で追補添 カロしてもよレ、。
培養は好気的条件下で行う。 本発明において、 微生物はリ ビトール以外にエリ
スリ トールとグリセロールを副生する場合があるが、 リ ビトールの生成比率を効 率よくするためには十分な通気を行うことが重要である。
培養温度は通常 2 0〜 3 7 °C、 好ましくは 2 7〜 3 2 °Cで、 通常 2 4時間から 2週間行う。 本発明の製造方法における第 2工程は、 リビトールから L—リブロースを製造 する工程である。 第 2工程で使用する微生物としては好気的反応条件下、 リ ビト ールを L _リブロースに変換する能力を有する微生物であれば特に限定はされな いが、 リビトールから L—リブロースを製造し、 かつリビトール以外の副生産さ れたポリアルコールを資化する能力を有する微生物が好ましく、 より好ましくは、 ァセ トパクター(Acetobacter) 属、 アルカリゲネス (Alcaligenes ) 属、 ァシネ トバクタ一(Acinetobacter )属、 ァグロパクテリゥム(Agrobacterium) 属、 アル ス口 / クタ一(Arthrobacter)属、 ァェロモナス(Aeromonas) 属、 才ーレ才ノくクテ リウム(Aureobacterium)属、 ノくチノレス(Bacillus)属、 ブレビノ クテリゥム (Brev ibacterium) 属、 クレブシエラ(Klebsiella)属、 オタロバク 卜ラム(Ochrobactru m)属、 リゾビゥム(Rhizobium) 属、 セラチア(Serratia)属、 ロ ドバクタ一(Rhodo bacter) 属、 コリ /くクテリウム(Corynebacterium) 凰、 ェンテロ/ベクター(Ent erobacter)属、 グノレコノノ クタ一 (Gluconobacter) 属、 ミクロコッカス (Microco ecus) 属、 ノヽラコッカス (Paracoccus)属、 シユードモナス (Pseudomonas ) 属に 属する微生物である。 特にダルコノバクタ一属に属する微生物が、 L—リブ口一 スの生成効率の点からも、 ポリアルコールの資化性の点からも好ましい。
本発明において使用するダルコノバクタ一 (Gluconobacter) 属に属する微生 物は、 例;^ば Gluconobacter frateurii , Gluconobacter oxydans等力 ^挙げられる。 さらに具体的な代表株として Gluconobacter frateuri i IFO 2508 、 Gluconobact er oxydans IFO 3293 が挙げられる。
本発明において使用するァセトパクター属に属する微生物は、 例えば、 Acetob actor acinus等が挙げられ、 具体的な菌株としては、 Acetobactor acinus FERM P-2507が挙げられる。
本発明において使用するアルカ リゲネス属に属する微生物は、 例えば Alcalige
nes aquamarinus, Alcaligenes epoxylyticus, Alcal igenes margaritae, Alcal igenes paradoxus, Alcal igenes xylosoxidans subsp. xylosoxidansカ挙げられ ligenes epoxylyticus FERM P - 2511, Alcaligenes margaritae FERM P-2512, Al caligenes xylosoxidans subsp. xylosoxidans I AM 12684, Alcal igenes parado xus Biotype I DSM66, Alcal igenes paradoxus Biotype II DSM30162力 S挙げら れる。
本発明において使用するァシネトパクター属に属する微生物は、 例えば Acinet obacter tartarogenesカ挙げられる。 さらに具体的には、 Acinetobacter tartar ogenesIF013644が挙げられる。
本発明において使用するァルスロバクタ一属に属する微生物は、 例えば Arthro bacter atrocyaneus, Arthrobacter pascens, Arthrobacter ramousus等力挙け られる。 さら iこ具体的 ίこ(ま Arthrobacter pascens IF012139, Arthrobacter atr ocyaneus JCM 1329, Arthrobacter ramousus JCM1334等力 ΐ挙げられる。
本発明において使用するァグロバクテリゥム属に属する微生物は、 例えば Agro bacterium radiobacter, Agrobacterium tumefaciens, Agrobacterium viscosum が挙げられる。 さらに具体的な代表株として Agrobacterium radiobacter NRRL B - 11291, Agrobacterium tumefaciens IAM13129, Agrobacterium viscosum IF013 652 が挙げられる。
本発明において使用するォ一レオパクテリゥム属に属する微生物は、 Aureobac terium testaceum力挙けられる。 さらに具体的な代表株として Aureobacterium testaceum JCM1353 が挙げられる。
本発明において使用するバチルス属に属する微生物は、 例えば Baci llus alvei, Bacillus coagulansが挙げられる。 さらに具体的な代表株として Bacil lus alve i IF03343, Bacillus coagulans AHU 1631 が挙げられる。
本発明において使用するブレビバクテリゥム属に属する微生物は、 Brevibacte rium busi llum が挙げられる。 さらに具体的な代表株として Brevibacterium bus ilium IA 1489 が挙げられる。
本発明において使用するコリネバクテリゥム属に属する微生物は、 Corynebac terium f laccumf aciens カ けられる。 さらに具体的な代表株として Corynebac
terium f laccumfaciens ATCC12813 が挙げられる。
本発明において使用するェンテロバクタ一属に属する微生物は、 Enterobacter aerogenes力 S挙デられる。 さらに具体的な代表株として Enterobacter aerogenes IF013534 が挙げられる。
本発明において使用するクレブシエラ属に属する微生物は、 例えば Klebsiella planticola, Klebsiel la oxytoca , Klebsiella terrigena力 ΐ挙けられる。 さら に具体的な代表株として Klebsiella planticola NCIB11885, Klebsiella oxytoc a JCM1665, Klebsiella terrigena JCM1687 が挙げられる。
本発明において使用するミクロコッカス属に属する微生物は、 例えば Micrococ cus roseus力 S挙デられる。 さらに具体的な代表株として Micrococcus roseus IFO 3764 が挙げられる。
本発明において使用するォクロバク トラム属に属する微生物は、 Ochrobactrum s anthropi力 ΐ挙げられる。
さらに具体的な代表株として Ochrobactrums anthropi I AM 13993が挙げられる。 本発明において使用するパラコッカス属に属する微生物は、 例えば Paracoccus denitrificansカ挙 Iテられる。 さらに具体的な代表株として Paracoccus denitri ficans IF013301 が挙げられる。
本発明において使用するシユードモナス属に属する微生物は、 例えば Pseudomo nas f luorecens, Pseudomonas synxantha カ挙けられる。 さらに具体的な代表株 として Pseudomonas f luorecens IFO 3903, Pseudomonas f luorecens ATCC13525, Pseudomonas synxantha IAM13256 挙げられる。
本発明において使用するァエロモナス属に属する微生物は、 例えば Aeromonas panctata、 具体的に ίま、 Aeromonas panctata IF013288 力 s挙げ、られる。
本発明において使用するリゾビゥム属に属する微生物は、 例えば Rhizobium va lidum IF013648が挙げられる。
本発明において使用するセラチア属に属する微生物は、 例えば Serratia marce scens 力挙けられ、 例;^ば Serratia marcescens IF03054 、 Serratia marcescen s ATCC13880 が挙げられる。
本発明において使用する口 ドパクター属に属する微生物は、 例えば Rhodobacte
r sphaeroides 、 例; ίま Rhodobacter sphaeroides IF012003力挙げ'られる。 上記微生物は、 Institute for Fermentation ( I F O ) 、 Institute of Appli ed Microbiology ( I AM) 、 Deutsche Sammlung fon Mlkroorganismen und Zel lkulturen GmbH ( D S M) 、 Japan Collect ion of Microorganisms ( J C M) 、 Laboratory of Culture Collection of Microorganisms ( A H U ) 、 American T ype Culture Collection ( A T C C ) 、 The National Collections of Industri al and Marin Bacteria ( N C I M B ) から入手することができる。
なお、 上記微生物は、 野生株の他、 U V照射、 N—メチル一 N, 一ニトロソグ ァニジン (N T G ) 処理、 ェチルメタンスルホネート (E M S ) 処理、 亜硝酸処 理、 ァクリジン処理などによる変異株、 あるいは細胞融合もしくは遺伝子組み換 え法などの遺伝的手法により誘導される組み換え株などいずれの株であってもよ レ、。 上記微生物はリビトール以外の副生されたポリアルコールを資化し、 リ ビト —ルを効率的に; L—リブロースに変換できるので、 その後の L—リブロースの分 離 ·精製工程における負荷を著しく軽減することが出来る。
本工程においては、 上記微生物は 1種あるいは 2種以上が用いられる。
次に、 本発明の第 2工程について具体的に説明する。
本工程で用いるリビトールは、 精製されたものであってもよく、 リビトール以 外の物質を含有する未精製の発酵液または該発酵液を部分精製したものであって もよい。 具体的には、 リビト一ル含有液は、 菌体を含んでいても良いし、 遠心分 離などにより菌体を除去したものでもよい。 またリ ビトールの発酵に用いた微生 物を蒸気滅菌などにより殺菌したものでも良いし、 殺菌を行っていない発酵液で も良い。 またこの発酵液を簡便な方法を用いて精製させたものでもよい。 例えば 発酵液を除菌後、 濃縮し晶析させたものでも良いし、 また、 カラムクロマトダラ フィ一などで部分精製させたものでも良い。 また発酵液を適当な緩衝液、 例えば、 酢酸バッファ一、 リン酸バッファ一、 トリ スバッファー、 等、 あるいは水を用い て希釈しても良い。 あるいは濃縮させても良い。 これらの発酵液に微生物を好気 的条件で作用させ、 L—リブロースを生成させる。
本発明の第 2工程での微生物培養に用いる培地は微生物が生育でき、 L—リブ ロース生産能を誘導し、 かつ、 好ましくはポリアルコールの資化性を誘導する培
地であれば、 いずれでもよい。 さらに培地は液体状でも良いし固体培地でも良い。 炭素源としては上記微生物が資化しうるグルコースなどの炭水化物、 グリセロー ルなどのアルコール類、 有機酸などが挙げられ、 適宜一種あるいは 2種以上が選 ばれる。 好ましくはポリアルコ一ルの資化性を誘導するグリセロールなどのアル コール類が挙げられる。 窒素源としては本微生物が資化しうる窒素源であればい ずれでも良いが、 好ましくは、 酵母エキス、 コーンスティ一プリカ一、 N Zアミ ン、 トリプト一ス、 ペプトン、 ポリペプトン、 肉エキス、 魚肉エキス、 その他の 有機窒素源、 あるいは硝酸ナトリウム、 その他の無機窒素源が適宜 1種あるいは 2種使用される。 培地中に含まれる炭素源及ぴ窒素源の濃度は 0 . 0 1 %〜2 0 %の間で用いられる。 炭素源、 窒素源のほかに、 無機イオンやビタミン類を必要 に応じ添加することは有効である。 無機イオンとしては、 リン酸イオン、 ナトリ ゥムイオン、 カリ ウムイオン、 マグネシウムイオン、 鉄イオン、 マンガンイオン、 モリブデンイオン、 カルシウムイオン、 コバルトイオン、 その他金属イオン類等 が適宜数種類用いられる。 ビタミン類としては、 チアミン、 イノシトール、 パン トテン酸、 ニコチン酸アミ ドなどが挙げられ、 適宜用いられる。 また、 上述のグ ルコースその他の炭素源、 窒素源、 無機イオン、 ビタミン類は、 必要に応じて培 養中の適当な時点で追補添加してもよい。
培養は好気的条件下で行う。 培養温度は 4 °Cから 5 0 °Cの範囲で行われるが、 好ましくは 2 0から 4 0 °Cの範囲である。 通気量は 0. OOlvvn!〜 2vvm好ましくは 0. 5〜1. 5vvmの範囲で行われる。 11は2〜 1 0、 好ましくは 3〜 9の間である。 培養は、 通常 1 0時間から 2日間行う。
次に、 第 2工程に供する菌体は、 培養液そのままを用いてもよいし、 遠心分離 等により集菌したものであってもよい。 培養して得られた菌体をそのまま、 ある いは培養して得られた菌体を公知の手法で処理したもの、 即ち、 アセ トン処理、 凍結乾燥処理、 界面活性剤処理あるいはトルエン処理など菌体を物理的または酵 素的に破砕したもの等の菌体処理物を用いることができる。 また、 これらの菌体 または菌体処理物から、 リビトールに作用し、 L—リブロースを生産する能力を 有する酵素画分を粗精製物あるいは精製物として取り出して用いることも可能で ある。 さらには、 このようにして得られた菌体、 菌体処理物、 酵素画分等をポリ
アクリルアミ ドゲル、 カラギーナンゲル等の担体に固定化したもの等を用いるこ とも可能である。 そこで本発明において、 「微生物」 の用語は、 上述の菌体、 菌 体処理物、 酵素画分、 およびそれらの固定化物全てを含有する概念として用いら れる。
反応は好気的条件下で行う。 反応温度は 4 °Cから 5 0 °Cの範囲で行われるが好 ましくは 2 0から 4 0 °Cの範囲である。 p Hは 2〜 1 0、 好ましくは 3〜 8の間 である。 撹拌が十分に行われていれば特に通気の必要は無いが、 好ましくは 0 . 0 1 V v m〜 1 V v mの間で通気する。 反応に用いる菌体量は湿潤菌体量換算で リビトール含有発酵液に対し 0 . 0 0 1 %〜 5 0 % (重量 Z体積) の間で用いら れる。 リビトールの濃度は特に限定されないが、 好ましくは 0 . 1から 1 5 %の 間である。 含有されるポリアルコールの濃度は特に限定されない。 反応は、 通常 1時間から 2週間行う。
反応により得られた L—リブロースは、 上記で得られた培養液を精製せずにそ のまま用いても良いし、 培養液から公知の方法により分離精製した後に第 3段階 の反応に供することもできる。 即ち、 培養終了後、 まず微生物菌体を遠心分離等 の既存の方法で除去する。 除菌に先立ち、 培養液に熱を加え、 殺菌を行ってもよ レ、。 除菌された液は通常の分離方法、 即ちクロマトグラフィーなどを用いること で容易に精製できる。 工業的には、 イオン交換樹脂を用いた疑似移動相型クロマ トグラフィ一により分離することも可能である。 分離精製の工程においては、 必 要に応じて脱塩、 脱色など、 通常の糖の精製における操作を加えることもできる。 本発明の製造方法における第 3工程は、 L—リブロースから Lーリボースを製 造する工程である。 第 3段階で使用する微生物としては、 L一リブロースを異性 化して Lーリボースを生産する能力を有する微生物であれば、 特に限定されない 力 好ましくはァシネトパクター属またはセル口モナス属に属する微生物であり、 更に好ましくは、 ァシネトバクタ一 'カルコァセティカス (Acinetobacter calc oaceticus ) もしく セノレ口モナス · ゲリタ、、 (Cel lulomonas gerida ) 、 セノレ口 モナス ビアゾテア (Cellulomonas biazotea ) またはセルロモナス ' フラビゲ ナ (Cel lulomonas flavigena) の種に属する微生物が挙げられる。 ァシネ トバク タ一属に属する微生物としてァシネトバクタ一 .カルコァセティカス (Acinetob
acter calcoaceticus ) D L— 2 8株、 セノレ口モナス · ゲリダ (Cellulomonas g erida ) に属する微生物と して J C M 1 4 9 0が、 セルロモナス ♦ ビアゾテア
(Cellulomonas biazotea ) に属する微生物として A T C C 4 8 6が、 セルロモ ナス フラビゲナ (Cellulomonas flavigena) に属する微生物として A T C C 4 8 2等が挙げられる。 これらの微生物は、 Japan Collect ion of Microorganisms
( J C M) 及び American Type Culture Collection ( A T C C ) カ ら入手するこ とができる。
また上記微生物は、 野生株の他、 紫外線照射や N- メチル- Ν' -ニトロ- Ν - ニトロソグァ二ジン (N T G ) 処理、 ェチルメタンスルホネート (E M S ) 処理、 亜硝酸処理、 ァクリジン処理等通常知られた方法による変異株、 あるいは細胞融 合もしくは遺伝子組換え法などの遺伝学的手法により誘導される組換え株などの いずれの株であってもよい。 また、 上記微生物の代わりに、 欧州公開第 8 0 7 6 8 2号公報に記載のような酵素 (L—リブロース異性化酵素) を用いてもよい。 本工程においては、 上記微生物を 1種あるいは 2種以上が用いられる。 次に、 本発明の第 3工程について具体的に説明する。
本工程で用いる L—リブロースは、 精製されたものであってもよく、 L—リブ ロース以外の物質を含有する未精製の発酵液または該発酵液を部分精製したもの であってもよい。 具体的には、 L—リブロース含有液は、 第 1および第 2工程で 用いた菌体を含んでいても良いし、 遠心分離などにより菌体を除去したものでも よい。 また、 第 1及び第 2工程で用いた微生物を蒸気滅菌などにより殺菌したも のでも良いし、 殺菌を行っていない発酵液でも良い。 またこの発酵液を簡便な方 法を用いて精製させたものでもよい。 例えば L—リブロース発酵液を除菌後、 濃 縮し晶析させたものでも良いし、 また、 カラムクロマトグラフィーなどで部分精 製させたものでも良い。 また発酵液を適当な緩衝液、 例えば、 酢酸バッファー、 リン酸バッファ一、 トリスバッファ一、 等、 あるいは水を用いて希釈しても良い。 あるいは濃縮させても良い。 これらの発酵液に微生物を好気的条件で作用させ、 L—リボースを生成させる。
本発明の第 3工程において微生物は定法通り培養される。 つまり、 炭素源とし
ては上記微生物が資化しうるグルコースなどの炭水化物、 グリセロールなどのァ ルコール類、 有機酸など、 好ましくはグルコース、 グリセロール、 スクロース等 が、 更に好ましくはスクロースが用いられる。 またこれらの炭素源は培養中に必 要に応じて 1種類あるいは複数種類適宜加えることができる。 さらにこの培地に は上記微生物が資化しうる窒素源が含まれる。 窒素源としては、 アミノ酸類、 酵 母エキス、 大豆ペプチド、 大豆粉末、 コーンスティープリカ一、 N Zァミン、 ト リプトース、 ペプトン、 ポリペプトン、 肉エキス、 魚肉エキスその他の有機窒素 源、 あるいは硝酸ナトリウム、 その他の無機窒素源、 好ましくは、 酵母エキスや 大豆ペプチド、 ポリペプトンが適宜使用される。 上述の炭素源、 窒素源のほかに、 無機イオンゃビタミン類を必要に応じ添加することは有効である。 無機イオンと しては、 リ ン酸イオン、 マグネシウムイオン、 鉄イオン、 マンガンイオン、 モリ ブデンイオンその他が用いられる。 ビタミン類としては、 チアミン、 イノシト一 ル、 パントテン酸、 ニコチン酸アミ ドなどが挙げられる。 また、 上述のスクロー スその他の炭素源、 窒素源、 無機イオン、 ビタミン類は、 必要に応じて培養中の 適当な時点で追補添加してもよい。
培養は好気的条件下で行う。 本発明において、 L—リブロースから L—リボー スを生産するイソメラ一ゼを高活性に発現させるために通常 0 . 1〜 2 . 0 V V m、 好ましくは 0 . 5〜 2 . O v v mで通気を行う。
培養温度は、 通常 2 0〜3 7 °C、 好ましくは 2 7〜 3 2 °Cで、 通常 1 2時間か ら 4 8時間行う。
次に、 異性化反応に供する菌体は、 培養液そのままを用いてもよいし、 遠心分 離等により集菌したものであってもよい。 培養して得られた菌体をそのまま、 あ るいは培養して得られた菌体を公知の手法で処理したもの、 即ち、 アセトン処理、 凍結乾燥処理、 界面活性剤処理あるいはトルエン処理など菌体を物理的または酵 素的に破砕したもの等の菌体処理物を用いることができる。 また、 これらの菌体 または菌体処理物から、 L一リブロースに作用し、 L _リボースを生産する能力 を有する酵素画分を粗精製物あるいは精製物として取り出して用いることも可能 である。 さらには、 このようにして得られた菌体、 菌体処理物、 酵素画分等をポ リアクリルァミ ドゲル、 カラギ一ナンゲル等の担体に固定化したもの等を用いる
ことも可能である。 そこで本発明において、 「微生物」 の用語は、 上述の菌体、 菌体処理物、 酵素画分、 およびそれらの固定化物全てを含有する概念として用い られる。 特に好ましい実施形態としては菌体をトルエン処理することが挙げられ る。 Lーリブロース液に菌体と トルエンを加えてもよい。 好ましくは、 培養終了 液から菌体を濃縮分離しトルエン処理を行った後、 異性化反応に供する。
異性化反応は、 p H 4〜9 . 5、 好ましくは P H 8 . 5〜9 . 0に調整した L 一リブロース水溶液に上記菌体あるいは菌体処理物をリブロース 1 g当たり、 2 ュニット以上、 好ましくは 6ュニット以上になるように添加し、 温度 1 0 °C〜 5 0 °C、 好ましくは、 2 0 °C〜4 0 °Cで行う。 反応が進行するにつれ、 反応液の p Hが低下するため、 水酸化ナトリ ウム等のアルカリ溶液にて p Hを 9 . 0付近に 制御することが好ましい。 用いた菌体の量により、 反応は数秒から数日で L—リ ボースと L—リブロースが平衡に達して終了する。 以上 3工程により生成した L—リボースを精製する。 本発明者らは鋭意検討の 結果、 通常の種々の精製技術をある順序、 条件に従って組み合わせることにより、 反応液から L -リボースを分離精製するに至った。 本発明の製造方法において、 Lーリボースはグルコース等の糖質原料を出発原料として、 例えば 3段階の生物 反応により得られるが、 この様に多段階の生物反応を行った場合、 得られた L一 リボース含有液は、 未反応の Lーリブロース以外にも微生物由来のタンパク質等 の高分子物質、 塩類、 副反応で生成したと思われる有機酸類や、 ジヒ ドロキシァ セトン、 その他少量の糖類、 着色成分等の夾雑物を含む。 これらの夾雑物は精製 の早い時期に出来るだけ取り除いておくことが、 その後の精製にとって望ましい。 反応終了後、 まず微生物菌体を遠心分離等の既存の方法で除去する。 除菌に先 立ち、 培養終了液に熱を加え、 酵素失活およぴ澱下げを行ってもよい。 さらに除 菌前または除菌後に炭酸飽充を行うことはその後の精製に大変有効である。 除菌 された液は L—リボース以外に未だ上述の夾雑物が多く含まれる。 これらは通常 の方法、 即ちクロマトグラフィーゃ晶析技術を用いることで除去できる。 夾雑物 が多い場合は特に晶析に先立ってゲル型濾過材に接触させ、 夾雑物の多くを除去 することが好ましい。 本発明で用いるゲル型濾過材としては、 Lーリボースと他
の成分とを効率よく分離することが出来るものであれば特に限定されないが、 好 ましくはカチオン系イオン交換体が挙げられ、 更に好ましくは架橋度 4〜 8 %の アル力リ金属またはアル力リ土類金属型のカチオン系イオン交換体が挙げられる。 かかるイオン交換体の具体例としては、 U B K— 5 3 0、 U B K— 5 3 5、 U B K— 5 5 0、 U B K— 5 5 5 (いずれも三菱化学製) 等が挙げられる。 これらを 用いてクロマト分離を行うことが好ましい。
具体的な精製手段としては、 例えば上述したゲル型濾過材を脱塩水にてスラリ —化し、 十分に気泡を除去した後分離カラムに充填する。 該カラムに、 充填した 濾過材容量の 1 / 2〜 1 1 0 0、 好ましくは 1 / 5〜: 1 Z 2 0量の粗 Lーリボ ース含有液を送入し、 次いで水で溶出させて Lーリボース画分とその他の画分と に分画する。 この L—リボース画分を、 通常知られた方法により脱塩脱色後、 晶 析を行うことにより高純度の Lーリボースを得ることができる。
また工業的には分離効率を上げ、 溶離水の使用量を減らす目的で、 公知の種々 のクロマト分離手法や疑似移動床法を用いることにより、 さらに効果的な分画を 行うこともできる。
分離精製の工程においては、 必要に応じて脱塩、 脱色など、 通常の糖の精製に おける操作を加えることもできる。
最終的に精製された L—リボース液には未反応の L—リブロースが残存するが、 この混合液より L—リボース結晶を得るには、 Lーリボース含有液に有機溶媒を 添加し、 反応液中に残存する未反応の L—リブロースを析出させずに Lーリボー スを単独で析出させる方法が好ましい。 L—リボース含有液は十分に濃縮され、 好ましくはプリ ックス 6 0以上、 さらに好ましくはブリ ックス 8 5以上、 さらに 好ましくはプリックス 9 0以上に濃縮される。 この濃縮シロップを 4 °Cに放置す ると L—リボースのみが析出する。 しかしこのような低温晶析では析出するまで の時間が長く、 また析出率も満足のいくものではない場合が多く、 好ましくは有 機溶媒添加による方法が挙げられる。 つまり、 上述の十分に濃縮された L—リボ ース含有シロップに、 シロップの 1 / 2〜 1 0倍量好ましくは 1〜 3倍量の有機 溶媒を加え、 十分に撹拌後、 微量の Lーリボース結晶を加え、 4でで2 4時間放 置する。 添加する有機溶媒としては、 メタノール、 エタノール等の低級アルコー
ル類、 アセ トン、 へキサン、 ァセ トニ トリル、 トルエンなどが挙げられ、 好まし くはエタノール、 1—プロパノール、 2—プロパノール、 更に好ましくはェタノ ールが挙げられる。 晶析に先立って再度クロマト分離を行い、 Lーリボース含有 液中から L一リブロースを除いておけば、 さらに晶析率は上昇する。 発明を実施するための最良の形熊 以下に実施例を挙げて本発明をさらに具体的に説明するが、 その要旨を越えな い限り本発明の技術分野における通常の変更をすることができる。 実施例 1
1 ) リビト一ル発酵上清の製造
30 L容量のジャーにグルコース 30%、 魚肉エキス 2%からなる培地 1 8 L を入れ、 トリコスポロノイデス メガチリェンシス (MC I 3442) の種培養 1 80mLを接種した。 種培養液は、 予めバッフル付きフラスコを用いてダルコ —ス 30 %、 酵母エキス 1 %からなる培地中で 30°C、 2日間、 1 60 r pmの 振とう培養により得られた。 ジャーは 330 r pm、 l v vm、 30°Cの条件で 制御され、 5日間培養を行った。 得られた培養液から遠心分離により清澄な培養 上清 1 2 Lを得た。 この発酵を 5回行い、 リ ビトール発酵上清 60 Lを得た。 各 成分の濃度はリ ビトール 80 g/L, エリスリ トール 4 g/L、 グリセロール 4 0 g ZLであった。
2) 粗 L—リブロース液の製造
グルコノバクタ一 . フラテゥリ (Gluconobacter frateurii) (IF02508) を 1 % グリセロール、 0. 5%酵母エキス、 0. 5 %ポリペプトンよりなる培地 1 00 m 1 を入れた 500m lバッフル付きフラスコ 2本に接種した。 1 60 r p mで 回転する振とう培養機にセッ トし、 30°Cで 1 日間培養を行った。 30 Lジャ一 フアーメンターに 3 %グリセロール、 0. 5%酵母エキス、 0. 5%ポリぺプト ンよりなる培地を 20 1仕込み、 1 20度 20分殺菌した。 これに先の培養液を 200m l植菌し、 3 0度で、 p Hを 5以下にならないように 5 N水酸化ナトリ
ゥムでコントロールしながら 1 日培養した。 培養終了後、 培養液を遠心分離して、 培養上清除き、 ダルコノバクタ一 ' フラテゥリの菌体を得た。
上記 1) で製造したリビトール発酵上清液を 1 5 L仕込んだ 30 Lジャーファ —メンターに入れ、 得られたダルコノバクタ一の菌体を入れ、 30°C、 通気 l v vm、 撹拌 330 r pmの条件で反応した。 反応 1 日後、 反応液 1 m 1を遠心分 離にかけて菌体を除き、 反応上清を下記表 1に示す条件で高速液体クロマトダラ フィー分析した。 リビトールは完全に変換され、 エリスリ トール及ぴグリセ口一 ルは検出されなかった。 生成した L—リブロースは 78. 2 g/Lであった。 グ リセロールはジヒ ドロキシアセトンに変換されており、 生成量は 23. 3 g/L であった。 エリ スリ トールはエリ スルロースに変換されていたが、 生成量の測定 はスタンダードの試薬が、 純度不明瞭のため、 正確な測定ができなかった。

上記 2) で得られた L—リブロース含有発酵液に、 異性化反応に供する菌体の トルエン処理物が加えられた。 即ち、 2) で得られた L—リブロース液は、 除菌 その他の処理を施すことなく、 同一ジャー内で連続して異性化反応が行われた。 具体的には 2) の反応終了時点で Lーリブロース含有発酵液を水酸化ナトリウム 水溶液により p H9. 0に調整し、 続いて下記 4) で得られたトルエン処理菌体 1 Lを加えて、 異性化反応を開始した。 ジャーは反応温度 28 °C、 1 1 2 r p m に制御され、 p Hは 1 ON水酸化ナトリ ウム水溶液にて 8〜8. 5の間で緩やか に制御された。 24時間後 Lーリボースと L一リブロースの混合比が約 7 : 3に
達し、 反応は終了した。
遠心分離により反応終了液から菌体を除き清澄な粗 Lーリボース液 15 L を得た。 含有する L—リボースは 39. 1 g/L, L—リブロースは 16 0 g ZL、 ジヒ ドロキシアセトンは 7 g ZLであった。
4) 異性化反応用トルエン処理菌体の製造
ァシネ トノ クタ一♦ 力ノレコアセティカス (Acinetobacter calcoaceticus ) D L— 28株の種培養液をスクロース 2. O gZL、 酵母エキス 5. 0 g/L, 大豆ペプチド 5. 0 g/L, N a C 1 5 g/L、 K2H P O4 3 g/L, K H2P θ 4 l gZL、 リ ジン一塩酸塩 5 g L ( p H 7. 0) よりなる培地 20 Lを入れた 30 Lジャーに 1 %接種した。 種培養は予め同一培地を用いてバッフ ル付きフラスコで 160 r pm、 30。Cで 12時間振とう培養して得られた。 ジ ヤーは 300 r pm、 l v vm、 30 °Cの条件で制御され、 1 6時間培養を行つ た。 得られた培養終了液を遠心分離して上清を除き、 1 0倍濃縮菌体液を 1. 7 Lを得た。 この濃縮菌体 1. 7 Lにトルエンを 27 mL加え、 15分間激しく混 合した。
ァシネトバクタ一 'カルコァセティカス DL— 28株に代えて、 セルロモナス . ゲリダ J CM1490、 セルロモナス . ビアゾテア ATCC486、 セルロモ ナス フラビゲナ ATCC 482等も同様に用いることができる。
5) 粗 Lーリボース液の精製
3) で得られた粗リボース液をエバポレーターにてプリックス 40まで濃縮し た後、 カラムクロマトグラフィーの原料に供した。
特開昭 63 - 158105号公報に記載された疑似移動床型のクロマト分離系 (充填された充填床の前端と後端とが流体通路で連結され、 流体の循環を可能に したもの) において、 充填材として N a型の強力チオン交換樹脂 (ダイヤイオン UBK— 530, 三菱化学社製) を使用し、 脱着剤として水を使用した。 内径 2 8 mm, 充填層高 500 mmの直列に連結した 4本のカラムに前記充填材を合計 で 1 24 OmL充填した充填床を 65°Cに保ち、 床內を容積流速 60 OmL/h rで通液し、 供給はすべて第 1カラム、 抜き出しはすべて第 4カラムとし、 下記
表 2に示すタイムスケジュールで定常となるまで分離操作を行った。 定常状態に 達した後の各画分の組成を、 下記表 3に示す。 L—リボースの回収率は 8 1 . 8 %であった。 また、 原料に含まれていた着色成分も多くは L—リボース以外のそ の他の画分に分離されており、 L—リボース画分の着色はクロマト分離前に比べ て大幅に軽減されていた。 表 2 供給 ·抜出工程 1 0 8分

得られた L—リボース含有画分を分子量分画 1 0 0 0 0の中空糸型限外濾過膜 にて濾過した後、 カチオン交換樹脂を充填したカラムに通液し、 続いてァニオン 交換樹脂を充填したカラムに通液した。 以上の脱塩脱色工程を経た L—リボース 含有液は無色透明であるが、 場合により若干の着色成分の残ることがあった。 そ の場合は少量の活性炭を加え 6 0 °Cで 1時間撹拌後、 ケィソゥ土濾過により活性 炭を除くことで残る着色成分を除いた。 このようにして得られた無色透明の L一 リボース含有液をエバポレーターにてブリ ックス 8 7まで濃縮後、 シロップの 2
倍容量のエタノールを添加し、 微量の L—リボース結晶を種晶として加え、 4°C で 24時間静置したところ、 L—リボース結晶が析出した。 この結晶を冷ェタノ —ルで数回洗浄した後乾固し、 純度 99. 9%の結晶を得た。 晶析率は 70%で めった。
尚、 晶析後の母液からエタノールを留去し、 再度濃縮してエタノールおよび種 晶を加え、 低温で放置することで 2番晶、 3番晶を得ることができた。 実施例 2
1 ) リビト一ル発酵上清の製造
実施例 1の 1 ) と同様に培養を行い、 培養上清 1 2 Lを得た。 培養上清中の成 分の濃度はリ ビト一ル 80 gZL、 エリスリ トール 5 g / L、 グリセ口ール 50 gZLであった。
2) 粗 L—リブロース液の製造
ダルコノバクタ一 · フラテゥリ ( I FO 2508) を実施例 1と同様に培養し て、 培養液 1 8 Lを得た。 培養終了後、 培養液を遠心分離して培養上清を除き、 菌体を 10 OmMのリン酸緩衝液バッファ一 (p H 7. 0) 1. 8 Lに懸濁した。
30 Lジャーに、 1) で得られたリビトール発酵上清 1 2 Lを入れ、 ダルコノ パクター懸濁液を 1. 8 Lを加えて、 30°Cにて 300 r p mで撹拌し、 通気 1 ^ 111で1 日間反応を行った。 反応終了液を遠心分離して菌体を除き清澄な粗 L —リブロース液 1 2. 5 Lを得た。 生成した L—リブロース濃度は 75 g/L、 ジヒ ドロキシアセトン 29 g/Lであった。
3) 粗 L—リボース液の製造
セルロモナス ' ゲリダ (J CM1 490) をスクロース 2. 0 g/L, 酵母 エキス 5. 0 g/L, 大豆ペプチド 5. 0 g/L, N a C 1 5 g/L, K 2ΗΡ〇4 3 g/L、 ΚΗ2Ρ 04 1 g/L, L—リボース 1 5 g/L ( p H 7. 0) よりなる培地 1 8 Lを入れた 30 Lジャーに 1 %接種した。 種培養は予め同 一培地を用いてバッフル付きフラスコで 1 60 r pm、 30°Cで 1 8時間振とう
培養して得られた。 ジャーは 300 r pm、 l v vm、 30 °Cの条件で制御され、 1 8時間培養を行った。 得られた培養終了液を遠心分離し、 集菌した。 菌体ペレ ットにグリシン—塩酸緩衝液 (5 OmM、 p H 8. 5) 1. 8 Lを加え均一に懸 濁した後、 トルエンを 60mL加え、 1 5分間激しく混合した。 このように調製 したトルエン処理菌体 1. 2 Lを、 2) で得られた粗 L—リブロース液 1 2. 5 Lに加え、 均一になるよう緩やかに撹拌しつつ 30°Cで 24時間反応を行った。 途中、 I N Na OHによって p H8. 5に制御した。 得られた反応終了液を遠 心分離して菌体を除き、 さらに 0. 22 μ m孔径の精密濾過膜を通して清澄な粗 L—リボース液 1 2 Lを得た。 L—リボース濃度は 30 g "L、 L—リブロース 濃度は 1 3 gZ であった。
4) 粗 Lーリボース液の精製
3) で得られた粗リボース液をエバポレ一ターにて濃縮し、 2. 7 Lの粗リボ —スシロップを得た。 このシロップの濃度はプリックス 63、 L—リボースの対 固形分比率は HP LCのピーク面積換算で約 38%、 L—リブロースの比率は 1 5%であった。 このシロップをカラムクロマトグラフィーの原料に供した。 即ち イオン交換榭脂 UBK— 550 (三菱化学社製) 32 OmLを充填したカラム 4 本を直列に接続し、 カラム温度を 65°Cに保った状態でその一方の端から流速 6 00 mL/h rで粗リボースシロップ 64mLを供給した。 次いで水を流速 60 OmL/h rで送入し、 シロップをカラム内で展開、 分離させながらカラム末端 に移動させた。 水を送入して 78分後から目的とする L—リボースが流出し始め た。 さらに 1 2分してから流出液を Lーリボース含有画分として採取を開始した。 水の送入開始から 1 24分後 L—リボースの流出がほぼ終わったので、 サンプル 回収を停止した。 この L—リボース含有画分中の Lーリボースの対固形分比率は HP LCのピーク面積換算で 80%、 L—リブロースの比率は 1 4 %であり、 L —リボースの回収率は 80. 1 %であった。 この L—リボース画分をさらに実施 例 1記載と同様の脱塩脱色処理をした後、 プリックス 90まで濃縮し 4°Cで 1ケ 月静置したところ、 L—リボース結晶が析出した。 この結晶を冷アルコール等で 洗浄し、 乾固したところ純度 99%の L一リボース結晶が得られた。
実施例 3 実施例 1の疑似移動床法にて得られた L—リボース含有画分をさらにカラムク 口マトグラフィ一により精製した。
実施例 1 と同様のクロマト分離系を用い、 充填材として C a型の強力チオン交 換樹脂 (ダイヤイオン UBK— 5 55, 三菱化学社製) を使用した。 下記表 4に 示すタイムスケジュールで定常となるまで分離操作を行った。 定常状態に達した 後の各画分の組成を、 下記表 5に示す。 L—リボースの回収率は 72. 4%であ つた。 表 4 供給 ·抜出工程 9. 6分

得られた L—リボース画分を実施例 1と同様に脱塩脱色処理した後、 ブリック ス 8 7まで濃縮し、 2—プロパノールを 2倍量と微量の Lーリボース結晶を加え、 4°Cで 1 2時間撹拌したところ純度 9 9. 5 %の L—リボース結晶が得られた。
なお、 各実施例において得られた培養上清に含まれるリビトールなどの糖類含 量は高速液体クロマトグラフィ一により各々下記表 6に示す条件で測定した。 各 糖質の保持時間は下記分析条件 Aにおいて、 グルコース 1 0. 57分、 リ ビトー ノレ 1 2. 22分、 エリスリ トール 1 3. 36分、 グリセ口一ル 1 5. 09分であ る。 また、 下記分析条件 Bにおいて、 L—リブロース 22. 8分、 L—リボース 24. 0分である。 表 6 高速液体クロマトグラフィー分析条件 A

Claims
1. 糖質原料からリビトールを生産する能力を有する微生物を用いて糖質原 料からリビトールを生成する工程と、 リビトールから L一リブロースを生産する 能力を有する微生物を用いてリビトールから Lーリブロースを生成する工程と、 L—リブロースから L—リボースを生産する能力を有する微生物を用いて L—リ ブロースから L—リボースを生成する工程を含む Lーリボースの製造方法。
2. 前記糖質原料がグルコースである請求項 1記載の方法。
3. グルコースからリビトールを生産する能力を有する微生物がトリコスポ 口ノイデス属に属することを特徴とする請求項 1記載の方法。
4. 微生物がトリコスポロノイデス 'マディダ、 トリコスポロノイデス ·二 グレスセンス、 トリコスポロノイデス 'エドセフアリス、 トリ コスポロノイデス • メガチリェンシスおよびトリコスポロノイデス .スパチュラータから選ばれる とを特徴とする請求項 3記載の方法。
5. 微生物がトリコスポロノイデス ' エドセファリスおよびトリコスポロノ イデス ·メガチリェンシスから選ばれることを特徴とする請求項 3に記載の方法。
6. トリコスポロノイデス属に属する微生物が、 トリコスポロノイデス 'マ デイダ CB S 240. 79、 トリ コスポロノイデス ' ニグレスセンス CB S 2 68. 8 1及び〇83269. 8 1、 トリコスポロノイデス 'エドセフアリス CB S 568. 85および CB S 649. 66、 トリコスポロノイデス ' メガチ リエンシス CB S 567. 85、 トリ コスポロノイデス ' スパチュラータ CB S 241. 79、 CB S 242. 79 A及び CB S 242. 79 Bから選ばれる ことを特徴とする請求項 4に記載の方法。
7. トリコスポロノイデス属に属する微生物がトリコスポロノイデス ·エド セファリス CB S 568. 85およびトリコスポロノイデス · メガチリエンシ ス CB S 567. 85から選ばれることを特微とする請求項 5に記載の方法。
8. リビトールから L—リブロースを生産する能力を有する微生物が、 リビ トール以外の副生産されたポリアルコールを資化する能力を有する微生物である ことを特徴とする請求項 1又は 3記載の方法。
9. リビトールから L一リブロースを生産する能力を有する微生物が、 ァセ トバクタ一属、 アルカリゲネス属、 ァシネトパクター属、 ァグロバクテリウム属、 ァルスロバクター属、 ァエロモナス属、 オーレォバクテリウム属、 バチルス属、 ブレビバクテリウム属、 クレブシエラ属、 ォクロバク トラム属、 リゾピウム属、 セラチア属、 ロドパクター属、 コリネバクテリウム属、 ェンテロバクター属、 グ ルコノバクター属、 ミクロコッカス属、 パラコッカス属およびシユードモナス属 に属する微生物から選ばれることを特徴とする請求項 1又は 3記載の方法。

1 3. リビトールが、 リビトール以外の物質を含有する未精製の発酵液また は該発酵液を部分精製したものであることを特徴とする請求項 1又 8記載の方法。
1 4. L—リブロースから L—リボースを生産する能力を有する微生物が、 セル口モナス属およびァシネトパクター属に属する微生物から選ばれることを特 徴とする請求項 1、 3、 8又は 9のいずれか一項に記載の方法。
1 5. 微生物が、 セルロモナス 'ゲリダ、 セルロモナス ' ビアゾテアおよび セルロモナス · フラビゲナから選ばれることを特徴とする請求項 14記載の方法。
1 6. 微生物がセルロモナス ·ゲリダ J CM 1 490、 セルロモナス · ビ ァゾテア ATCC 486およびセルロモナス フラビゲナ ATCC482から 選ばれることを特徴とする請求項 1 5記載の方法。
1 7. L—リブロースが、 L一リブロース以外の物質を含有する未精製の発 酵液または該発酵液を部分精製したものであることを特徴とする請求項 14に記 載の方法。
1 8. 生成した L—リボースを含む反応液をゲル型濾過材に接触させること を特徴とする請求項 1記載の方法。
1 9 . ゲル型濾過材との接触により L—リボースをクロマト分離することを 特徴とする請求項 1 8記載の方法。
2 0 . ゲル型濾過材がカチオン系のイオン交換体であることを特徴とする請 求項 1 9記載の方法。
2 1 . 生成した L—リボースを含む反応液に有機溶媒を添加し、 反応液中に 残存する未反応の L—リブロースを析出させずに L—リボースを単独で析出させ ることを特徴とする請求項 1記載の方法。
2 2 . 有機溶媒が、 低級アルコール類、 アセトン、 へキサン、 ァセトニトリ ルまたはトルエンであることを特徵とする請求項 2 1記載の方法。
